  • Matrix stiffness controls lymphatic vessel formation through regulation of a GATA2-dependent transcriptional program

    • Tissue and vessel wall stiffening alters endothelial cell properties and contributes to vascular dysfunction. However, whether extracellular matrix (ECM) stiffness impacts vascular development is not known. Here we show that matrix stiffness controls lymphatic vascular morphogenesis. Atomic force microscopy measurements in mouse embryos reveal that venous lymphatic endothelial cell (LEC) progenitors experience a decrease in substrate stiffness upon migration out of the cardinal vein, which induces a GATA2-dependent transcriptional program required to form the first lymphatic vessels. Transcriptome analysis shows that LECs grown on a soft matrix exhibit increased GATA2 expression and a GATA2-dependent upregulation of genes involved in cell migration and lymphangiogenesis, including VEGFR3. Analyses of mouse models demonstrate a cell-autonomous function of GATA2 in regulating LEC responsiveness to VEGF-C and in controlling LEC migration and sprouting in vivo. Our study thus uncovers a mechanism by which ECM stiffness dictates the migratory behavior of LECs during early lymphatic development.

    • Electric vehicles (EVs) are widely regarded as valuable assets in the smart grid as distributed energy resources in addition to their primary transportation function. However, connecting EVs to the distribution network and recharging the EV batteries without any control may overload the transformers and cables during peak hours when the penetration of EVs is relatively high. In this study, a two level hierarchical control method for integrating EVs into the distribution network is proposed to coordinate the self-interests and operational constraints of two actors, the EV owner and Distribution system operator (DSO), facilitated by the introduction of the fleet operator (FO) and the grid capacity market operator (CMO). Unlike the typical hierarchical control system where the upper level controller commands the low level unit to execute the actions, in this study, market based control are applied both in the upper and low level of the hierarchical system. Specifically, in the upper level of the hierarchy, distribution system operator uses market based control to coordinate the fleet operators power schedule. In the low level of the hierarchy, the fleet operator use market based control to allocate the charging power to the individual EVs, by using market based control, the proposed method considers the flexibility of EVs through the presence of the response-weighting factor to the shadow price sent out by the FO. Furthermore, to fully demonstrate the coordination behavior of the proposed control strategy, we built a multi-agent system (MAS) that is based on the co-simulation environment of JACK, Matlab and Simulink. A use case of the MAS and the results of running the system are presented to intuitively illustrate the effectiveness of the proposed solutions.

    • The ability of laccases from Trametes villosa (TvL), Myceliophthora thermophila (MtL), Trametes hirsuta (ThL) and Bacillus subtilis (BsL) to improve the dispersion properties of calcium lignosulfonates 398 in the presence of HBT as a mediator was investigated. Size exclusion chromatography showed an extensive increase in molecular weight of the samples incubated with TvL and ThL by 107% and 572% from 28400 Da after 17 h of incubation, respectively. Interestingly, FTIR spectroscopy. C-13 NMR and Py-GC/MS analysis of the treated samples suggested no substantial changes in the aromatic signal of the lignosulfonates, a good indication of the ability of TvL/ThL-HBT systems to limit their effect on functional groups without degrading the lignin backbone. Further, the enzymatic treatments led to a general increase in the dispersion properties, indeed a welcome development for its application in polymer blends. (C) 2010 Elsevier Ltd.
