  • A metabolite from commensal Candida albicans enhances the bactericidal activity of macrophages and protects against sepsis

    • The gut microbiome is recognized as a key modulator of sepsis development. However, the contribution of the gut mycobiome to sepsis development is still not fully understood. Here, we demonstrated that the level of Candida albicans was markedly decreased in patients with bacterial sepsis, and the supernatant of Candida albicans culture significantly decreased the bacterial load and improved sepsis symptoms in both cecum ligation and puncture (CLP)-challenged mice and Escherichia coli-challenged pigs. Integrative metabolomics and the genetic engineering of fungi revealed that Candida albicans-derived phenylpyruvate (PPA) enhanced the bactericidal activity of macrophages and reduced organ damage during sepsis. Mechanistically, PPA directly binds to sirtuin 2 (SIRT2) and increases reactive oxygen species (ROS) production for eventual bacterial clearance. Importantly, PPA enhanced the bacterial clearance capacity of macrophages in sepsis patients and was inversely correlated with the severity of sepsis in patients. Our findings highlight the crucial contribution of commensal fungi to bacterial disease modulation and expand our understanding of the host-mycobiome interaction during sepsis development. © 2023, The Author(s), under exclusive licence to CSI and USTC.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

    • To improve industrial sustainability performance in manufacturing, energy management and optimi-sation are key levers. This is particularly true for aluminium extrusions manufacturing dan energy -intensive production system with considerable environmental impacts. Many energy management and optimisation approaches have been studied to relieve such negative impact. However, the effectiveness of these approaches is compromised without the support of refined supply-side energy consumption information. Industrial internet of things provides opportunities to acquire refined energy consumption information in its data-rich environment but also poses a range of difficulties in implementation. The existing sensors cannot directly obtain the energy consumption at the granularity of a specific job. To acquire that refined energy consumption information, a supply-side energy modelling method based on existing industrial internet of things devices for energy-intensive production systems is proposed in this paper. First, the job-specified production event concept is proposed, and the layout of the data acqui-sition network is designed to obtain the event elements. Second, the mathematical models are developed to calculate the energy consumption of the production event in three process modes. Third, the energy consumption information of multiple manufacturing element dimensions can be derived from the mathematical models, and therefore, the energy consumption information on multiple dimensions is easily scaled. Finally, a case of refined energy cost accounting is studied to demonstrate the feasibility of the proposed models. ? 2021 The Author(s). Published by Elsevier Ltd.   • Bark Beetle-Associated Blue-Stain Fungi Increase Antioxidant Enzyme Activities and Monoterpene Concentrations in Pinus yunnanensis

    • Yunnan pine is the most important tree species in SW China in both economical and ecological terms, but it is often killed by pine shoot beetles (Tomicus spp.). Tomicus beetles are secondary pests in temperate regions and the aggressiveness of the beetles in SW China is considered to be due to the warm subtropical climates as well as the beetles' virulent fungal associates. Here, we assessed the virulence of three blue-stain fungi (Leptographium wushanense, L. sinense and Ophiostoma canum) associated with pine shoot beetles to Yunnan pine (Pinus yunnanensis) in SW China. Following fungal inoculation, we measured necrotic lesion lengths, antioxidant enzyme activities and monoterpene concentrations in the stem phloem of Yunnan pine. Leptographium wushanense induced twice as long lesions as L. sinense and O. canum, and all three fungi induced significantly longer lesions than sterile agar control inoculations. The activity of three tested antioxidant enzymes (peroxidase, polyphenol oxidase, and superoxide dismutase) increased after both fungal inoculation and control inoculation. However, L. wushanense and L. sinense generally caused a greater increase in enzyme activities than O. canum and the control treatment. Fungal inoculation induced stronger increases in six major monoterpenes than the control treatment, but the difference was significant only for some fungus-monoterpene combinations. Overall, our results show that L. wushanense and L. sinense elicit stronger defense responses and thus are more virulent to Yunnan pine than O. canum. The two Leptographium species may thus contribute to the aggressiveness of their beetle vectors and could damage Yunnan pine across SW China if they spread from the restricted geographical area they have been found in so far.

    • Ophiostomatalean fungi may facilitate bark beetle colonization and reproduction. In the present study of the fungal community associated with bark beetle spe-cies belonging to Tomicus in Yunnan, China, six ophios-tomatalean fungi (Ophiostoma canum, O. ips, O. tingens, Leptographium yunnanense, Leptographium sp. 1 and Lep-tographium sp. 2) were isolated from the beetles or their galleries; O. canum was the most common fungal species. The distribution of O. canum was associated with stands heavily damaged by Tomicus species and a higher percentage of valid galleries of Tomicus yunnanensis and T. minor in Yunnan pine (Pinus yunnanensis). After inoculation of Yun-nan pine with the fungus, a phloem reaction zone formed and monoterpenes accumulated in the phloem. These results suggested that O. canum was pathogenic to Yunnan pine and that the wide distribution of the fungus might be beneficial to reproduction of pine shoot beetles in Yunnan pine. How-ever, because the reaction zone and monoterpene accumu-lation were mild, fungal damage of Yunnan pine might be limited. A more integrated study considering all the fungal species should be done to better understand the interactions among bark beetles, blue-stain fungi, and the tree hosts in the region.

  • FTO genetic variants, dietary intake and body mass index : insights from 177 330 individuals

    • FTO is the strongest known genetic susceptibility locus for obesity. Experimental studies in animals suggest the potential roles of FTO in regulating food intake. The interactive relation among FTO variants, dietary intake and body mass index (BMI) is complex and results from previous often small-scale studies in humans are highly inconsistent. We performed large-scale analyses based on data from 177 330 adults (154 439 Whites, 5776 African Americans and 17 115 Asians) from 40 studies to examine: (i) the association between the FTO-rs9939609 variant (or a proxy single-nucleotide polymorphism) and total energy and macronutrient intake and (ii) the interaction between the FTO variant and dietary intake on BMI. The minor allele (A-allele) of the FTO-rs9939609 variant was associated with higher BMI in Whites (effect per allele = 0.34 [0.31, 0.37] kg/m(2), P = 1.9 × 10(-105)), and all participants (0.30 [0.30, 0.35] kg/m(2), P = 3.6 × 10(-107)). The BMI-increasing allele of the FTO variant showed a significant association with higher dietary protein intake (effect per allele = 0.08 [0.06, 0.10] %, P = 2.4 × 10(-16)), and relative weak associations with lower total energy intake (-6.4 [-10.1, -2.6] kcal/day, P = 0.001) and lower dietary carbohydrate intake (-0.07 [-0.11, -0.02] %, P = 0.004). The associations with protein (P = 7.5 × 10(-9)) and total energy (P = 0.002) were attenuated but remained significant after adjustment for BMI. We did not find significant interactions between the FTO variant and dietary intake of total energy, protein, carbohydrate or fat on BMI. Our findings suggest a positive association between the BMI-increasing allele of FTO variant and higher dietary protein intake and offer insight into potential link between FTO, dietary protein intake and adiposity.

  • Solar, Atmospheric, and Volcanic Impacts on 10Be Depositions in Greenland and Antarctica During the Last 100 Years

    • Cosmogenic radionuclides (e.g., 10Be) from ice cores are a powerful tool for solar reconstructions back in time. However, superimposed on the solar signal, other factors like weather/climate and volcanic influences on 10Be can complicate the interpretation of 10Be data. A comprehensive study of 10Be records over the recent period, when atmospheric 10Be production and meteorological conditions are relatively well-known, can improve our interpretation of 10Be records. Here we conduct a systematic study of the production and climate/volcanic signals in Antarctica and Greenland 10Be records, including a new 10Be record from the East GReenland Ice-core Project site. Greenland and Antarctica records show significant decreasing trends (5%–6.5%/decade) for 1900–1950, which is comparable with the expected production rate inferred from sunspot observations. By comparing 10Be records with reanalysis data and atmospheric circulation patterns, 10Be records from Southern/Southeastern Greenland are significantly correlated with the Scandinavia pattern. Stacking 10Be records from different locations can enhance the production signal. However, this approach is not always straightforward as uncertainties in some records can lead to a weaker solar signal. A strategy can be employed to select records for the bipolar stack by comparing Greenland records with Antarctica records, assuming the shared signal is a production signal. Finally, we observe significant increases (36%–64%) in 10Be depositions in Greenland related to the Agung eruption. This large increase in Greenland 10Be records after the Agung eruption, could be partly explained by the enhanced air mass transport from mid-latitudes coinciding with the decreased precipitation en-route.
