    • Even though ammonia is considered to underlie nervous system symptoms of dysfunction during hyperammonemia, lactate, which increases as a metabolic consequence of high ammonia levels, might also be a contributing factor. The data presented here show that NH4Cl (5 mM) mediates astroglial cell swelling, and that treatment with NH4Cl or lactate (25 mM) causes rearrangements of actin filaments and reduces astroglial glutamate uptake capacity. Co-application with BaCl2, which blocks astroglial uptake of NH4+, prevents NH4Cl-mediated cell swelling and rearrangement of actin filaments, but does not reduce NH4Cl-induced glutamate uptake capacity inhibition. Neither NH4Cl nor lactate affected glutamate uptake or protein expression in microglial cultures, indicating that astroglial cells are more susceptible to the neurotoxic affects of ammonia. Our results suggest that ammonium underlies brain edema, but that lactate can contribute to some of the cellular dysfunctions associated with elevated cerebral levels of ammonia.

  • Implementation of daily visual management at five small and medium sized enterprises in Produktionslyftet compared to six larger Swedish companies

    • This paper compares the implementation of daily visual management (DVM) in five SMEs (small and medium sized enterprises), with practice in six large Swedish companies, considering the challenge to include sustainability aspects. With method definition from Produktionslyftet (PL), a Swedish development programme, DVM includes daily meetings around key performance indices (KPIs) and visualised information on boards. The SMEs were studied within PL while the large companies were studied in the research project SuReBPMS (Sustainable and resource efficient Business Performance Measurement Systems). The empirical result from SMEs shows that KPIs often cover few areas (most common: delivery, safety and quality) while the larger companies have wider range of areas. In some SMEs the meeting agendas, KPIs and pulse-boards are standardised while in others there are differences between teams or operations. The larger companies were mainly standardised. Both SMEs and larger companies mostly include all employees in DVM, in line with PL-DVM methodology. The general tendency is to focus on short term rather than follow-up of strategic challenges in the DVM. Furthermore, environmental issues, such as climate impact, are rarely found on boards, while often reflected in company policy. However, many KPIs have a strong, indirect, connection to sustainability and environment.

  • Initial DNA Interactions of the Binuclear Threading Intercalator Λ,Λ-[μ-bidppz(bipy)4Ru2]4+: An NMR Study with [d(CGCGAATTCGCG)]2

    • Binuclear polypyridine ruthenium compounds have been shown to slowly intercalate into DNA, following a fast initial binding on the DNA surface. For these compounds, intercalation requires threading of a bulky substituent, containing one RuII, through the DNA base-pair stack, and the accompanying DNA duplex distortions are much more severe than with intercalation of mononuclear compounds. Structural understanding of the process of intercalation may greatly gain from a characterisation of the initial interactions between binuclear RuII compounds and DNA. We report a structural NMR study on the binuclear RuII intercalator -B (-[-bidppz-(bipy)4Ru2]4+; bidppz=11,11’-bis(di-pyrido[3,2-:2’,3’-c]phenazinyl, bipy = 2,2’-bipyridine) mixed with the palindromic DNA [d(CGCGAATTCGCG)]2. Threading of -B depends on the presence and length of AT stretches in the DNA. Therefore, the latter was selected to promote initial binding, but due to the short stretch of AT base pairs, final intercalation is prevented. Structural calculations provide a model for the interaction: -B is trapped in a well-defined surface-bound state consisting of an eccentric minor-groove binding. Most of the interaction enthalpy originates from electrostatic and van der Waals contacts, whereas intermolecular hydrogen bonds may help to define a unique position of -B. Molecular dynamics simulations show that this minor-groove binding mode is stable on a nanosecond scale. To the best of our knowledge, this is the first structural study by NMR spectroscopy on a binuclear Ru compound bound to DNA. In the calculated structure, one of the positively charged Ru2+ moieties is near the central AATT region; this is favourable in view of potential intercalation as observed by optical methods for DNA with longer AT stretches. Circular dichroism (CD) spectroscopy suggests that a similar binding geometry is formed in mixtures of -B with natural calf thymus DNA. The present minor-groove binding mode is proposed to represent the initial surface interactions of binuclear RuII compounds prior to intercalation into AT-rich DNA.

  • The influence of the molecular weight of the water-soluble polymer on phase-separated films for controlled release

    • Hydroxypropyl cellulose (HPC) and ethyl cellulose (EC) can be used for extended release coatings, where the water-soluble HPC may act as a pore former. The aim was to investigate the effect of the molecular weight of HPC on the microstructure and mass transport in phase-separated freestanding EC/HPC films with 30% w/w HPC. Four different HPC grades were used, with weight averaged molecular weights (Mw) of 30.0 (SSL), 55.0 (SL), 83.5 (L) and 365 (M) kg/mol. Results showed that the phase-separated structure changed from HPC-discontinuous to bicontinuous with increasing Mw of HPC. The film with the lowest Mw HPC (SSL) had unconnected oval-shaped HPC-rich domains, leaked almost no HPC and had the lowest water permeability. The remaining higher Mw films had connected complex-shaped pores, which resulted in higher permeabilities. The highest Mw film (M) had the smallest pores and very slow HPC leakage, which led to a slow increase in permeability. Films with grade L and SL released most of their HPC, yet the permeability of the L film was three times higher due to greater pore connectivity. It was concluded that the phase-separated microstructure, the level of pore percolation and the leakage rate of HPC will be affected by the choice of HPC Mw grade used in the film and this will in turn have strong impact on the film permeability.

  • Why do people accept or reject climate policies targeting food consumption? Unpacking justifications in the public debate in online social forums

    • A shift in dietary habits will be required to meet global climate targets. However, from a social dilemma perspective, major voluntary shifts in diet patterns are unlikely. Hence, government interventions are called for. This may be a perilous political endeavor, since food habits and choices are assumed to be personal and contentious matters and any food regulation policy risks stepping over the line for what people accept, risking policy legitimacy. In order to construct feasible policy measures, it is therefore important to gain knowledge of the prerequisites for support of climate food regulations and to understand why people accept or oppose regulations. The aim of this paper is to do so by analyzing the public debate concerning meat-free days in school canteens and a tax on meat in two public online social forums in Sweden. We seek to 1) map the arguments supporting (non)acceptability of the two food consumption regulation issues and 2) analyze what policy-specific and factual beliefs are reflected in the arguments and then detangle their meaning and content as revealed in the arguments. We find that policy-specific beliefs around freedom, fairness, and effectiveness are commonly used in support of or objection to these policies, but to different degrees, and often linked to factual beliefs about consequences for health or disadvantaged social groups. We conclude that the general reluctance of policy makers to interfere with what people eat is not necessarily well founded, and that better policy design, framing, and communication have the potential to increase policy support.

  • Touching Loops - Interactive Tactility in Textiles

    • have been designed. The collection is named “Touching Loops” and is the result of an exploration in interactive tactile properties for knitted textiles. As a first experiment, different yarns were tried out for their ability to change structure due to heat in several knitted constructions. The experiment resulted in three different material prototypes with the ability to sense touch and react by shrinking, breaking or hardening. The aim of the project is to explore new possibilities for interactive tactile knitted materials and structures. In the discussion we relate to these textile structures as possible materials in the context of architecture.

    • We report enhanced protonic and ionic dynamics in an imidazole/protic ionic liquid mixture confined within the nanopores of silica particles. The ionic liquid is 1-octylimidazolium bis(trifluoromethanesulfonyl)imide ([HC8Im][TFSI]), while the silica particles are microsized and characterized by internal well connected nanopores. We demonstrate that the addition of imidazole is crucial to promote a proton motion decoupled from molecular diffusion, which occurs due to the establishment of new N-H⋯N hydrogen bonds and fast proton exchange events in the ionic domains, as evidenced by both infrared and1H NMR spectroscopy. An additional reason for the decoupled motion of protons is the nanosegregated structure adopted by the liquid imidazole/[HC8Im][TFSI] mixture, with segregated polar and non-polar nano-domains, as clearly shown by WAXS data. This arrangement, promoted by the length of the octyl group and thus by significant chain-chain interactions, reduces the mobility of molecules (Dmol) more than that of protons (DH), which is manifested by DH/Dmolratios greater than three. Once included into the nanopores of hydrophobic silica microparticles, the nanostructure of the liquid mixture is preserved with slightly larger ionic domains, but effects on the non-polar ones are unclear. This results in a further enhancement of proton motion with localised paths of conduction. These findings demonstrate significant progress in the design of proton conducting materials via tailor-made molecular structures as well as by smart exploitation of confinement effects. Compared to other imidazole-based proton conducting materials that are crystalline up to 90 °C or above, the gel materials that we propose are useful for applications at room temperature, and can thus find applications in e.g. intermediate temperature proton exchange fuel cells.

    • Room temperature ionic liquids confined in a solid material, for example, nano-porous silica, are particularly propitious for energy related applications. The aim of this study is to probe the molecular interactions established between the protic ionic liquid diethylmethylammonium methanesulfonate (DEMA-OMs) and silica, where the latter consists of nano-porous micro-particles with pores in the size range of 10 nm. The changes in the local coordination and transport properties induced by the nano-confinement of the ionic liquid are investigated by a combination of Raman and solid-state NMR spectroscopy. In particular, one-dimensional (1D) H-1 and Si-29 and two-dimensional (2D) Si-29{H-1} HETOCR solid-state NMR are combined to identify the sites of interaction at the silica-ionic liquid interface. Pulsed field gradient (PFG) NMR experiments are performed to estimate the self-diffusion of both bulk and nano-confined DEMA-OMs. Complementary information on the overall coordination and interaction scheme is achieved by Raman spectroscopy. All these advanced experimental techniques are revealed to be crucial to differentiate between ionic liquid molecules residing in the inter-or intra-particle domains.
