  • Critical Success Factors in Capturing Knowledge for Retention in IT-Supported Repositories

    • In this paper, the authors demonstrate the suitability of IT-supported knowledge repositories for knowledge retention. Successful knowledge retention is dependent on whatis stored in a repository and, hence, possible to share. Accordingly, the ability to capture theright (relevant) knowledge is a key aspect. Therefore, to increase the quality in an IT-supported knowledge repository, the identification activity, which starts the capture process, must besuccessfully performed. While critical success factors (CSFs) for knowledge retention andknowledge management are frequently discussed in the literature, there is a knowledge gapconcerning CSFs for this specific knowledge capture activity. From a knowledge retention perspective, this paper proposes a model that characterizes CSFs for the identification activity and highlights the CSFs’ contribution to knowledge retention.

  • Neisseria meningitidis-Induced Caspase-1 Activation in Human Innate Immune Cells Is LOS-Dependent

    • Meningococcal disease such as sepsis and meningitidis is hallmarked by an excessive inflammatory response. The causative agent, Neisseria meningitidis, expresses the endotoxin lipooligosaccharide (LOS) that is responsible for activation of immune cells and the release of proinflammatory cytokines. One of the most potent proinflammatory cytokines, interleukin-1 (IL-1), is activated following caspase-1 activity in the intracellular multiprotein complex called inflammasome. Inflammasomes are activated by a number of microbial factors as well as danger molecules by a two-step mechanismpriming and licensing of inflammasome activationbut there are no data available regarding a role for inflammasome activation in meningococcal disease. The aim of this study was to investigate if N. meningitidis activates the inflammasome and, if so, the role of bacterial LOS in this activation. Cells were subjected to N. meningitidis, both wild-type (FAM20) and its LOS-deficient mutant (lpxA), and priming as well as licensing of inflammasome activation was investigated. The wild-type LOS-expressing parental FAM20 serogroup C N. meningitidis (FAM20) strain significantly enhanced the caspase-1 activity in human neutrophils and monocytes, whereas lpxA was unable to induce caspase-1 activity as well as to induce IL-1 release. While the lpxA mutant induced a priming response, measured as increased expression of NLRP3 and IL1B, the LOS-expressing FAM20 further increased this priming. We conclude that although non-LOS components of N. meningitidis contribute to the priming of the inflammasome activity, LOS per se is to be considered as the central component of N. meningitidis virulence, responsible for both priming and licensing of inflammasome activation.

  • The impact of nocturnal road traffic noise, bedroom window orientation, and work-related stress on subjective sleep quality: results of a cross-sectional study among working women.

    • To examine the effect of work-related stress and road noise exposure on self-rated sleep and potential additive interaction effects.Sleep and predictor variables were surveyed within two subsamples with 2191 and 1764 working women in a cross-sectional study. Sleep was assessed using a single question on general sleep quality and four questions on specific sleep problems and subsequently dichotomized (poor sleep vs. no poor sleep). Work-related stress was operationalized by job strain and effort-reward imbalance. Nocturnal exposure to road traffic noise was assessed as (a) the orientation of the bedroom window to a quiet façade vs. a low-, medium- or high-trafficked street and (b) energy-equivalent sound pressure levels for night-time modelled at the most exposed façade (Lnight). We distinguished between low (< 45 dB(A)), medium (45-50 dB(A)) and high exposure (> 50 dB(A)).Poor sleep was associated with job strain and effort-reward imbalance. The prevalence of poor sleep did not increase with increasing Lnight, but bedroom window orientation showed a non-significant trend. A quiet façade had a protective effect on sleep in each Lnight category. We found a non-significant trend for an additive interaction between bedroom window orientation and job strain.Noise levels modelled for the most exposed façade likely overestimate the actual exposure and thus may not be a precise predictor of poor sleep. Bedroom window orientation seems more relevant. Potential additive interaction effects between bedroom window orientation and job strain should be considered when interpreting epidemiological study results on noise-induced sleep disturbances.

  • Temporal trends of organochlorines in Northern Europe, 1967–1995. Relation to global fractionation, leakage from sediments and international measures

    • The time trend monitoring of organochlorine pollution was carried out in Sweden since the late 1960s. This report presents data on concentrations of DDT, PCB, HCHs and HCB in biota samples collected and analysed annually. All the matrices and compounds studied show a significant decrease over time. The data cover severely polluted Swedish marine and fresh water in southern Sweden as well as locally unpolluted waters in remote northern Arctic regions of Sweden. A total of 13 time series representing different locations and species are presented for the different pollutants. The period studied covers the time when pollution was serious as well as the time of recovery. All monitoring activities were carried out at the same laboratories over the entire study period, which means that comparability over time is good in the sets of data presented. The various time trends show a convincing agreement with trends and annual change over time, although the concentrations differ between the species and locations investigated, the highest concentrations being in the south. Since the annual changes are normally similar regardless of locations and species, spatial variations in concentrations remain over time, although concentrations are lower today. The onset of changes in concentrations over time can be related to international measures or other circumstances that lowered releases into the environment. Similarities in the annual changes, as well as the time when changes began, are discussed with respect to suggested hypotheses on the fate of the investigated organochlorines. It was not possible to verify that the oxygenation of anoxic sediments mobilised old pollution in Baltic sediments. Neither was it possible to conclude that eutrophication has caused a measurable effect on the rate and timing of the decreases. Finally, long-range transport to Arctic regions seems to be due more to a one step transport than to the ‘Grass-hopper’ effect. The comprehensive database used, clearly shows how important it is to have datasets big enough to describe between-year variation before attempting to evaluate the time trend. In addition, if between-year variation is not known, it is then also difficult to evaluate spatial variation on the basis of single year observations.

  • Beyond the global brain differences : intraindividual variability differences in 1q21.1 distal and 15q11.2 bp1-bp2 deletion carriers

    • Background: Carriers of the 1q21.1 distal and 15q11.2 BP1-BP2 copy number variants exhibit regional and global brain differences compared with noncarriers. However, interpreting regional differences is challenging if a global difference drives the regional brain differences. Intraindividual variability measures can be used to test for regional differences beyond global differences in brain structure.Methods: Magnetic resonance imaging data were used to obtain regional brain values for 1q21.1 distal deletion (n = 30) and duplication (n = 27) and 15q11.2 BP1-BP2 deletion (n = 170) and duplication (n = 243) carriers and matched noncarriers (n = 2350). Regional intra-deviation scores, i.e., the standardized difference between an individual's regional difference and global difference, were used to test for regional differences that diverge from the global difference.Results: For the 1q21.1 distal deletion carriers, cortical surface area for regions in the medial visual cortex, posterior cingulate, and temporal pole differed less and regions in the prefrontal and superior temporal cortex differed more than the global difference in cortical surface area. For the 15q11.2 BP1-BP2 deletion carriers, cortical thickness in regions in the medial visual cortex, auditory cortex, and temporal pole differed less and the prefrontal and somatosensory cortex differed more than the global difference in cortical thickness.Conclusions: We find evidence for regional effects beyond differences in global brain measures in 1q21.1 distal and 15q11.2 BP1-BP2 copy number variants. The results provide new insight into brain profiling of the 1q21.1 distal and 15q11.2 BP1-BP2 copy number variants, with the potential to increase understanding of the mechanisms involved in altered neurodevelopment.

  • Microstructure, grain size distribution and grain shape in WC-Co alloys sintered at different carbon activities

    • The properties of cemented carbides strongly depend on the WC grain size and it is thus crucial to control coarsening of WC during processing. The aim of this work was to study the effect of sintering at different carbon activities on the final microstructure, as well as the coarsening behavior of the WC grains, including the size distribution and the shape of WC grains. These aspects were investigated for five WC-Co alloys sintered at 1410 degrees C for 1 h at different carbon activities in the liquid, in the range from the graphite equilibrium (carbon activity of 1) to the eta (M6C) phase equilibrium (carbon activity of 0.33). The grain size distribution was experimentally evaluated for the different alloys using EBSD (electron backscatter diffraction). In addition, the shape of the WC grains was evaluated for the different alloys. It was found that the average WC grain size increased and the grain size distribution became slightly wider with increasing carbon activity. Comparing the two three-phase (WC-Co-eta and WC-Co-graphite) alloys a shape change of the WC grains was observed with larger grains having more planar surfaces and more triangular shape for the WC-Co-graphite alloy. It was indicated that in alloys with a relatively low volume fraction of the binder phase the WC grain shape is significantly affected by impingements. Moreover, after 1 h of sintering the WC grains are at a non-equilibrium state with regards to grain morphology. (C) 2013 Elsevier Ltd. All rights reserved.
