    • A desalting step using reversed phase chromatography is a common practice prior to mass spectrometry analysis of proteolytic digests in spite of the detrimental exclusion of the hydrophilic peptides. The detection of such peptides is also important for the complete coverage of protein sequences and the analysis of posttranslational modifications as inquired by regulatory agencies for the commercialization of biotechnological products. The procedure described here, named in-solution buffer-free digestion, simplifies the sample processing and circumvents the above-mentioned limitations by allowing the detection of tryptic hydrophilic peptides via direct ESI-MS analysis. Two DNA recombinant proteins such as HBcAg (hepatitis B core antigen) and fusion VEGF (vascular endothelial growth factor) were analyzed with the proposed in-solution buffer-free digestion allowing the detection of extremely hydrophilic di-, tri- and tetra-peptides, C-terminal His-tail peptide, as well as disulfide-containing peptides. All these molecular species are hardly seen in mass spectrometric analysis using a standard digestion that includes a C18-desalting step. The procedure was also successfully tried on hydrophilic tetra- and hexa-peptides of Ribonuclease B carrying an N-glycosylation site occupied with “high-mannose” N-glycan chains. The in-solution buffer-free digestion constitutes a simple and straightforward approach to analyse the hydrophilic proteolytic peptides which are commonly elusive to the detection by conventional mass spectrometric analysis.

  • Effect of Degradation on Wood Hygroscopicity : The Case of a 400-Year-Old Coffin

    • The hygroscopicity and thermodynamic properties of Pinus sylvestris L. wood from a coffin allegedly holding the remains of famous Spanish author Miguel de Cervantes Saavedra (1547–1616) were studied using the 15 °C and 35 °C isotherms fitted to the Guggenheim–Anderson–de Boer model and comparing them with the isotherms of recently felled wood of the same species. In addition, using infrared spectroscopy (FTIR-ATR) and X-ray diffractograms, the functional groups were determined and the crystallinity and organization of the cell wall components were analyzed. The absence of the 1740 cm−1 group indicates hemicellulose degradation in the archaeological wood, and the X-ray diffractograms show a considerable decrease in cellulose crystallinity and disorganization of the cellulose crystallites. The greater availability of active –OH groups means that the archaeological wood isotherms are above the juvenile and mature wood isotherms, and therefore the thermodynamic balance in the sorption of the archaeological wood is greater.

    • We describe here the synthesis and characterization of new sulfonated and carboxylated-containing N-donor ligands [Na(4)(edts)]·4H(2)O (2), [Na(2)(dmeddp)]·2H(2)O (3) and [Na(4)(edtp)]·H(2)O (4) (edts = ethylene-diamine- N,N,N',N'-tetraethylenesulfonate ion; dmeddp = dimethyl-ethylene-diamine-N,N,N',N'-tetra-3-propionate ion; edtp = ethylene-diamine-N,N,N',N'-tetra-3-propionate ion) and their corresponding metal (Ni, Co, Cu and Zn) complexes. Mainly, UV-Vis and a computer aided analysis of the EPR spectra provided information on the geometry and structure of the complexes in solution. Some of the metal complexes inhibit HIV replication when treating both pre- and post-infected PBMC cells, and hustle the inhibitory effect compared to the metal salts alone.

    • This chapter presents the preliminary results related to obtaining Na2CO3 through membrane distillation in a carbon capture and utilization scheme for the circular economy. This process is developed in a circular economy framework in which value can be given to waste streams or brines with high NaCl contents. The process proposes to capture CO2 from streams from multiple industrial processes such as biogas, combustion streams, or typical standard flue gases, based on a membrane process. The permeate and salt fluxes obtained when using membrane distillation to concentrate Na2CO3 have been explored. This was carried out through an experimental design which served to obtain a reliable model to predict permeate flux according to feed rate and feed temperature. The results showed that, for the range tested, the influence of the feed rate is greater than the influence of the temperature.
