  • Lymphocytes eject interferogenic mitochondrial DNA webs in response to CpG and non-CpG oligodeoxynucleotides of class C

    • Circulating mitochondrial DNA (mtDNA) is receiving increasing attention as a danger-associated molecular pattern in conditions such as autoimmunity, cancer, and trauma. We report here that human lymphocytes [B cells, T cells, natural killer (NK) cells], monocytes, and neutrophils derived from healthy blood donors, as well as B cells from chronic lymphocytic leukemia patients, rapidly eject mtDNA as web filament structures upon recognition of CpG and non-CpG oligodeoxynucleotides of class C. The release was quenched by ZnCl2, independent of cell death (apoptosis, necrosis, necroptosis, autophagy), and continued in the presence of TLR9 signaling inhibitors. B-cell mtDNA webs were distinct from neutrophil extracellular traps concerning structure, reactive oxygen species (ROS) dependence, and were devoid of antibacterial proteins. mtDNA webs acted as rapid (within minutes) messengers, priming antiviral type I IFN production. In summary, our findings point at a previously unrecognized role for lymphocytes in antimicrobial defense, utilizing mtDNA webs as signals in synergy with cytokines and natural antibodies, and cast light on the interplay between mitochondria and the immune system.

    • In an attempt to keep up with increased competition and shortening of product lifecycles, many companies are starting to implement the methods and principles that are offered by Lean product development. This paradigm of product development has mainly been described in management literature, and there are few industrial cases investigated outside Toyota. Existing literature points towards the Visual planning method as a good way of coordinating product development work, and is a method commonly used as a first step toward implementing Lean product development. This paper investigates the benefits and drawbacks of Lean Visual planning methods at three Swedish industrial companies. Benefits identified are efficient communication, shared understanding of the status of the work, ability to prioritize tasks and efficient leveling of workload. Complications associated with the method include difficulty of using it in geographically scattered teams, to store historical information and to see causal links between activities. The conclusion from this paper is that Visual planning constitutes a method that is useful for product development teams that need efficient ways of communicating and coordinating team work.

    • Lean product development is considered by many authors to promote learning and knowledge transfer, but studies that relate lean product development to the understanding drawn from the knowledge management field seem to be lacking. This article establishes a link between the concept of lean product development and the field of knowledge management. The transfer of knowledge within any organisation is recognised as a challenging task and mastering that challenge is increasingly important. Numerous studies show that common knowledge transfer tools such as post-project reviews are problematic, which calls for improved ways of transferring knowledge. By relating Lean principles and methods to the knowledge creation framework developed by Nonaka and Takeuchi (often referred to as the SECI-model), this article suggests that lean product development offers principles and methods that could provide a way of improving knowledge transfer in product development.

  • Lean Visual Planning and Product Development Complexity: Facilitating Information Processing Capability

    • Increasing complexity of products makes product development hard to manage and increases lead-time, which puts great demands on the information processing capability in organizations. This calls for methods that could facilitate this capability in a good way. Lean product development literature points towards the Visual planning method as a good way of managing information. However, studies that relate Visual planning methods to information processing capability seems to be lacking. This case study investigates how Visual planning can help managing complexity through increasing the information processing capability of an product development organization. Results show that Visual planning increases the extent and timeliness of information shared and is also an enabler for fast decision-making in the organization. The conclusion from this paper is that Visual planning constitutes a method that is useful for product development teams handling considerable complexity in coordination of their tasks.

    • This thesis investigates Visual planning, a method used in teams for planning and controlling product development projects. Cross-functional teams have shown to be effective at accelerating product development, but also make it difficult to coordinate the work and puts high demands on communication and knowledge exchange. Literature indicates that Visual planning might be a useful method to facilitate communication and knowledge exchange in product development teams. However, the research is still in its infancy and description of benefits, limitations and a clear definition of the method is lacking. Due to these early stages of research on the method, the purpose of this thesis is to define visual planning in product development. This purpose is adressed through examining current literature on Visual planning and product development through the lens of the resource-based view of the firm. The empirical data is based on a case study approach where data has been collected throughout three different studies. Visual planning consists of two components, where a board visualizing the team’s tasks is combined with short and frequent meetings. At the board, detailed planning and control of product development projects is done through color-coded artifacts such as sticky notes, representing task-related information. The method facilitates knowledge transfer through that it becomes a “forum” for task-related communication. Moreover, Visual planning seems to facilitate frequent, task related cross-functional team communication through these meetings and decentralization of decision-making trough the autonomy of workflow planning. There are also limitations with Visual planning the method related to amount of management complexity, co-location, size of the team using it and the physical limitations of the board itself compared to software tools. The method is defined as “Visual planning is a method used by a team to plan and control the team’s progress through the use of physical representations of tasks in combination with frequent and interactive meetings” Earlier literature on the method lacks consensus, and does not address the limitations of Visual planning. I have attempted to contribute to the emerging literature on Visual planning through both defining the content and limitations of the method itself and finding the benefits facilitated by the method that are important to product development.
