  • Body composition in patients with Alzheimer's disease and healthy controls
  • 1992
  • Ingår i: Journal of Clinical and Experimental Gerontology. - 0192-1193. ; 14:1, s. 17-31
  • Tidskriftsartikel (refereegranskat)abstract
    • In Alzheimer's disease (AD), decreased physical activity and nutritional problems are parts of the natural course with probable implications on body composition. Body fat and lean body mass were measured with a bioelectrical impedance method in 25 women who had AD according to the criteria of NINCDS- ADRDA and in 63 health age-matched controls. The patients with AD had 9.5 kg lower body weight (p<0.01), due to almost 4.0 kg lower body fat and 6.0 kg lower lean body mass (p<0.001). This might imply a higher risk for morbidity and mortality. Body weight and lean body mass decreased with age in AD patients but not in the healthy control women. In the control group, the unmarried women had more than 13 kg lower body weight and 9 kg lower body fat than the mean values of all other marital status groups (p<0.01). The maintenance of lean body mass in the very healthy old indicate the possibilities for physical activity.

  • Identification of a functional glucocorticoid response element in the promoter of the cylcin-dependant kinase inhibitor p57(Kip2)
  • 2003
  • Ingår i: Journal of Molecular Endocrinology. - : Bioscientifica. - 0952-5041 .- 1479-6813. ; 30:3, s. 359-368
  • Tidskriftsartikel (refereegranskat)abstract
    • Glucocorticoids are known regulators of the cell cycle, normally exerting an anti-proliferative effect. We have previously shown that glucocorticoids stimulate expression of p57(Kip2), a member of the Cip/Kip family of cyclin-dependent kinase inhibitors which, in some cell types, may account for the anti-proliferative responses seen after glucocorticoid treatment. The induction of p57(Kip2) involves primary transcriptional effects where no de novo protein synthesis is necessary, suggesting a direct interaction of the glucocorticoid receptor with the p57(Kip2) gene. In this study we have identified a functional glucocorticoid response element (GRE), located 5 kilo bases (kb) upstream of the transcription start site in the human P57(Kip2) promoter. This GRE was functional also when isolated, suggesting a direct transcriptional effect of the glucocorticoid receptor. Furthermore, mutation of this GRE abolished glucocorticoid induction of the reporter gene, whereas mutation of a nearby Sp1 site did not. Using electrophoretic mobility shift assays, we have shown that the -5 kb p57(Kip2) promoter GRE was able to compete with a well-known GRE for glucocorticoid receptor binding. Sequence comparisons with the mouse genome showed that this GRE is highly conserved, further strengthening the biological importance of this site. All these data emphasize the involvement of this GRE in the glucocorticoid-mediated induction of p57(Kip2) expression.

  • A national stroke quality register : 12 years experience from a participating hospital
  • 2007
  • Ingår i: European Journal of Neurology. - : Wiley. - 1351-5101 .- 1468-1331. ; 14:8, s. 890-894
  • Tidskriftsartikel (refereegranskat)abstract
    • Registration of all hospitalized stroke patients is practiced in Sweden in order to assess care quality. Data in this register, Riks-Stroke (RS), may be biased due to incomplete registration. The purpose of this paper was to report changes in stroke outcome in relation to fluctuations in registration. Patients registered in RS at a hospital during the period 1994-2005 were analyzed. Case fatality at 28 days, living conditions, and activities of daily living (ADL) performance at 3 months were correlated to the number of patients registered and follow-up frequency. A total of 4994 stroke cases were registered during the period. A high annual registration rate was significantly correlated to a high case fatality ratio. A low annual follow-up rate was associated with a low proportion of patients living in their own home without any need of help. Quality parameters are sensible for selection bias, which make them difficult to compare over time and between hospitals. We suggest that by weighing outcome data against stroke severity, safer conclusions may be drawn. Additionally, hospitals considering setting up quality registers should make every effort to attain complete case ascertainment at all times, including patients managed outside the hospital, in order to avoid selection bias.

  • The assessment of fitness to drive after a stroke : The Nordic Stroke Driver Screening Assessment
  • 2003
  • Ingår i: Scandinavian Journal of Psychology. - : Wiley. - 0036-5564 .- 1467-9450. ; 44:1, s. 23-30
  • Tidskriftsartikel (refereegranskat)abstract
    • The British Stroke Driver Screening Assessment (SDSA) is a set of four simple cognitive tests to evaluate driving fitness in stroke patients. To evaluate its usefulness in a Scandinavian context, we adapted the tests and assessed a group of 97 stroke patients from Sweden and Norway, using a driving test as the criterion. When results were calculated according to the original method, based on a discriminant function, less than 70% of the participants were correctly classified. To improve the predictive potential, a new discriminant analysis was performed, using the scores of a subsample of 49 patients, and validated on the remaining 48 participants. In total, 78% of the patients were correctly classified, but specificity was superior to sensitivity. We conclude that the Nordic version of the SDSA is a useful instrument, provided that test scores are interpreted in a balanced manner, taking into account the possibility of compensatory traffic behavior.

  • Contamination and correlation with toxicity of sediment samples from the Skiagerrak and Kattegat
  • 1996
  • Ingår i: Journal of Sea Research. - 1385-1101. ; 35:1-3, s. 223-234
  • Tidskriftsartikel (refereegranskat)abstract
    • The pollution state in the Skagerrak and Kattegat was investigated by determination of pollutant concentrations and toxicity of sediment samples from 11 stations in the area. A comparison was made with the sediment from a reference site near the Faroe Islands. Polycyclic aromatic hydrocarbons (PAH) and organochlorines were determined in whole sediment and heavy metals and ammonia were analysed in filtered pore water. Toxicity was bioassayed in whole sediment with Nitocra spinipes and Daphnia magna, in pore water with Mytilus edulis larvae and in solvent extracts from sediment with tests measuring etoxyresorufin-O-deethylase (EROD) activity in Oncorhyncus mykiss and rate of denitrification. Sites close to Goteborg and in an area from the Oslo fjord to the Norwegian Trench were most polluted. Sediment from the Faroe Islands was least polluted and also least toxic. Multivariate statistical analysis indicates that the different tests were sensitive to different kinds of pollutants. Effects on mussel larvae correlated strongest with the occurrence of ammonia, manganese, cadmium and PAHs, Nitocra with alpha-hexachlorocyclohexane (HCH) and p,p'-DDD, Daphnia with arsenic and gamma-HCH, fish EROD activity with benzo[ghi]perylene and unknown compounds associated with organic carbon, and denitrification with chlordanes, dieldrin and a few PAHs. The results indicate that sampling sites close to Goteborg are so polluted that harmful effects on the ecosystem probably occur.
