    • With widespread commercial applications in various domains, sentiment analysis has become a success story for Natural Language Processing (NLP). Still, although sentiment analysis has rapidly progressed during the last years, mainly due to the application of modern AI technologies, many approaches apply knowledge-based strategies, such as lexicon-based, to the task. This is particularly true for analyzing short social media content, e.g., tweets. Moreover, lexicon-based sentiment analysis approaches are usually preferred over learning-based methods when training data is unavailable or insufficient. Therefore, our main goal is to scale-up and apply a lexicon-based approach which can be used as a baseline to Swedish sentiment analysis. All scaled-up resources are made available, while the performance of this enhanced tool is evaluated on two short datasets, achieving adequate results.

  • Reduced amplification by phi29 DNA polymerase in the presence of unbound oligos during reaction in RCA

    • Synthetic single-stranded oligonucleotides play crucial roles in DNA amplification reactions for various applications, such as serving as primers, enabling magnetic separation, and generating dsDNA for subsequent digestion. Typically, these oligos are added in excess to ensure rapid binding to their intended targets. However, while performing rolling circle amplification (RCA) using phi29 DNA polymerase, we observed a decrease in amplification efficiency when oligos were present in the reaction. This phenomenon was consistently observed in two separate laboratories, prompting this study to delve into the root causes responsible for the decline in RCA efficiency. The lowered efficiency was consistent regardless of the manufacturer or any mutations in the phi29 polymerase. We identified several variables that influenced RCA efficiency, mainly the length of the oligos used and the presence of modifications, particularly those obstructing 3’ end digestion. This strongly suggests that the exonuclease domain of phi29 DNA polymerase is responsible for the competition-based inhibition. Our investigation shows that even picomole quantities of oligos can significantly reduce total DNA production during the phi29 DNA polymerase-mediated amplification process. Conversely, the addition of oligos to the reaction did not impede the efficiency of Bst 3.0 polymerase, likely due to the lack of an exonuclease domain of said polymerase. While increasing the quantity of phi29 DNA polymerase in the reaction partially alleviated the adverse effects of excess oligos, we believe it is crucial to carefully optimize the oligo quantities to achieve maximum amplification of the desired targets.

  • Does parental or grandparental farm upbringing influence risk of asthma in offspring?

    • Background: Farm upbringing has been associated with lower risk of asthma, and also with methylation of asthma-related genes. As such, farm upbringing has the potential to transfer less asthma risk across generations. We aimed to study generational effects from parental farm upbringing on offspring asthma.Methods: Our study involved three generations: 5,759 participants from the ECRHS study (born 1945-71, denoted G1), their 9,991 parents (G0) and their 8,260 offspring (G2) participating in RHINESSA. Questionnaire data on upbringing and asthma were available for all generations; direct information for G1 and G2, and via G2 for G0. Parental and grandparental place of upbringing was categorised as (1) both parents from farm (2) mother from farm, father from village/city (3) father from farm, mother from village/city (4) both parents from village or one parent from village and one from city (5) both parents from city (ref.). Data was analysed in Cox regression with G2 age as time scale.Results: Parental farm upbringing was not related to offspring asthma when compared to city upbringing (HR 1.12, 95 % CI 0.74-1.69) Findings remained similar when stratified by offspring upbringing and asthma phenotypes. Quantitative bias analyses showed similar estimates for alternative data sources. Grandparental farm upbringing was not associated with offspring asthma in either the maternal (HR 1.05, 95% CI 0.67-1.65) or paternal line (HR 1.02, 95% CI 0.62-1.68).Conclusion: This multi-generation analysis suggests no evidence of an association between parental or grandparental farm upbringing and offspring asthma.

    • During traditional industrial processing of fruit and vegetable derived foods, consideration of the eventual retention, bio-accessibility or bio-availability of nutrients has hitherto been a secondary priority. Indeed standard processing of soups and sauces involves treating all ingredients in a similar way, usually by prolonged heat treatment and results in both lowered nutritional value and sensory quality of food products. Such products are typically structured using various additives including starches, gums and stabilizers, which consumers regard as unnatural, rather than exploiting the endogenous structuring potential of the fruit and vegetable ingredients as available at the farm gate. Recent studies undertaken within the EU sponsored 'Healthy Structuring' project have shown how to design an industrially manufactured product with good sensory qualities that is natural and nutritious. The strategy has been to use the inherent structural and nutritional properties of the raw material ingredients, on the one hand to ensure the textural quality of the final product without the use of artificial gums or stabilizers and, on the other hand to optimize the nutritional quality of the final product. This review briefly summarizes both the approach taken and the results obtained within the project.
