  • Factors affecting peptide interactions with surface-bound microgels

    • Effects of electrostatics and peptide size on peptide interactions with surface-bound microgels were investigated with ellipsometry, confocal microscopy, and atomic force microscopy (AFM). Results show that binding of cationic poly-l-lysine (pLys) to anionic, covalently immobilized, poly(ethyl acrylate-co-methacrylic acid) microgels increased with increasing peptide net charge and microgel charge density. Furthermore, peptide release was facilitated by decreasing either microgel or peptide charge density. Analogously, increasing ionic strength facilitated peptide release for short peptides. As a result of peptide binding, the surface-bound microgels displayed pronounced deswelling and increased mechanical rigidity, the latter quantified by quantitative nanomechanical mapping. While short pLys was found to penetrate the entire microgel network and to result in almost complete charge neutralization, larger peptides were partially excluded from the microgel network, forming an outer peptide layer on the microgels. As a result of this difference, microgel flattening was more influenced by the lower Mw peptide than the higher. Peptide-induced deswelling was found to be lower for higher Mw pLys, the latter effect not observed for the corresponding microgels in the dispersed state. While the effects of electrostatics on peptide loading and release were similar to those observed for dispersed microgels, there were thus considerable effects of the underlying surface on peptide-induced microgel deswelling, which need to be considered in the design of surface-bound microgels as carriers of peptide loads, for example, in drug delivery or in functionalized biomaterials.

  • Climate mediates roles of pollinator species in plant–pollinator networks

    • Aim: Understanding how climate conditions influence plant–pollinator interactions at the global scale is crucial to understand how pollinator communities and ecosystem function respond to environmental change. Here, we investigate whether climate drives differences in network roles of the main insect pollinator orders: Diptera, Coleoptera, Lepidoptera and Hymenoptera. Location: Global. Time period: 1968–2020. Major taxa studied: Diptera, Coleoptera, Lepidoptera and Hymenoptera. Methods: We collated plant–pollinator networks from 26 countries and territories across the five main Köppen–Geiger climate zones. In total, we compiled data from 101 networks that included >1500 plant species from 167 families and >2800 pollinator species from 163 families. We assessed differences in the composition of plant–pollinator interactions among climate zones using a permutational ANOVA. We calculated standard network metrics for pollinator taxonomic groups and used Bayesian generalized mixed models to test whether climate zone influenced the proportion of pollinator network links and the level of pollinator generalism. Results: We found that climate is a strong driver of compositional dissimilarities between plant–pollinator interactions. Relative to other taxa, bees and flies made up the greatest proportion of network links across climate zones. When network size was accounted for, bees were the most generalist pollinator group in the tropics, whereas non-bee Hymenoptera were the most generalist in arid zones, and syrphid flies were the most generalist in polar networks. Main conclusions: We provide empirical evidence at the global scale that climate strongly influences the roles of different pollinator taxa within networks. Importantly, non-bee taxa, particularly flies, play central network roles across most climate zones, despite often being overlooked in pollination research and conservation. Our results identify the need for greater understanding of how global environmental change affects plant–pollinator interactions.

  • The HST See Change Program. I. Survey Design, Pipeline, and Supernova Discoveries

    • The See Change survey was designed to make z > 1 cosmological measurements by efficiently discovering high-redshift Type Ia supernovae (SNe Ia) and improving cluster mass measurements through weak lensing. This survey observed twelve galaxy clusters with the Hubble Space Telescope (HST) spanning the redshift range z = 1.13-1.75, discovering 57 likely transients and 27 likely SNe Ia at z similar to 0.8-2.3. As in similar previous surveys, this proved to be a highly efficient use of HST for supernova observations; the See Change survey additionally tested the feasibility of maintaining, or further increasing, the efficiency at yet higher redshifts, where we have less detailed information on the expected cluster masses and star formation rates. We find that the resulting number of SNe Ia per orbit is a factor of similar to 8 higher than for a field search, and 45% of our orbits contained an active SN Ia within 22 rest-frame days of peak, with one of the clusters by itself yielding 6 of the SNe Ia. We present the survey design, pipeline, and supernova discoveries. Novel features include fully blinded supernova searches, the first random forest candidate classifier for undersampled IR data (with a 50% detection threshold within 0.05 mag of human searchers), real-time forward-modeling photometry of candidates, and semi-automated photometric classifications and follow-up forecasts. We also describe the spectroscopic follow-up, instrumental in measuring host galaxy redshifts. The cosmology analysis of our sample will be presented in a companion paper.

    • A comprehensive database of paleoclimate records is needed to place recent warming into the longer-term context of natural climate variability. We present a global compilation of quality-controlled, published, temperature-sensitive proxy records extending back 12,000 years through the Holocene. Data were compiled from 679 sites where time series cover at least 4000 years, are resolved at sub-millennial scale (median spacing of 400 years or finer) and have at least one age control point every 3000 years, with cut-off values slackened in data-sparse regions. The data derive from lake sediment (51%), marine sediment (31%), peat (11%), glacier ice (3%), and other natural archives. The database contains 1319 records, including 157 from the Southern Hemisphere. The multi-proxy database comprises paleotemperature time series based on ecological assemblages, as well as biophysical and geochemical indicators that reflect mean annual or seasonal temperatures, as encoded in the database. This database can be used to reconstruct the spatiotemporal evolution of Holocene temperature at global to regional scales, and is publicly available in Linked Paleo Data (LiPD) format.

    • Agricultural intensification is a leading cause of global biodiversity loss, which can reduce the provisioning of ecosystem services in managed ecosystems. Organic farming and plant diversification are farm management schemes that may mitigate potential ecological harm by increasing species richness and boosting related ecosystem services to agroecosystems. What remains unclear is the extent to which farm management schemes affect biodiversity components other than species richness, and whether impacts differ across spatial scales and landscape contexts. Using a global metadataset, we quantified the effects of organic farming and plant diversification on abundance, local diversity (communities within fields), and regional diversity (communities across fields) of arthropod pollinators, predators, herbivores, and detritivores. Both organic farming and higher in-field plant diversity enhanced arthropod abundance, particularly for rare taxa. This resulted in increased richness but decreased evenness. While these responses were stronger at local relative to regional scales, richness and abundance increased at both scales, and richness on farms embedded in complex relative to simple landscapes. Overall, both organic farming and in-field plant diversification exerted the strongest effects on pollinators and predators, suggesting these management schemes can facilitate ecosystem service providers without augmenting herbivore (pest) populations. Our results suggest that organic farming and plant diversification promote diverse arthropod metacommunities that may provide temporal and spatial stability of ecosystem service provisioning. Conserving diverse plant and arthropod communities in farming systems therefore requires sustainable practices that operate both within fields and across landscapes.

    • Herein, electrostatic swelling transitions of poly (ethyl acrylate-co-methacrylic acid) microgels covalently bound to silica surfaces are investigated. Confined at a solid surface, microgel swelling is anisotropically hindered and the structure is flattened to an extent dictated by pH and microgel composition. Microgel deformation under applied load is also shown to depend on microgel charge density, with the highest deformation observed at intermediate charge densities. Two modes of microgel deformation under load were observed, one elastic and one viscoelastic, related to polymer strand deformation and displacement of trapped water, respectively. Results on polymer strand dynamics reveal that the microgels are highly dynamic, as the number of strand-tip interaction points increases 4-fold during a 10 s contact time. Furthermore, finite element modeling captures these effects qualitatively and shows that stress propagation in the microgel network decays locally at the rim of contact with a solid interface or close to the tip probe. Taken together, the results demonstrate a delicate interplay between the surface and microgel which determines the structure and nanomechanical properties of the latter and needs to be controlled in applications of systems such as pH-responsive surface coatings in biomaterials.

  • Dose dependence of mass and microcalcification detection in digital mammography: free response human observer studies.

    • The purpose of this study was to evaluate the effect of dose reduction in digital mammography on the detection of two lesion types-malignant masses and clusters of microcalcifications. Two free-response observer studies were performed-one for each lesion type. Ninety screening images were retrospectively selected; each image was originally acquired under automatic exposure conditions, corresponding to an average glandular dose of 1.3 mGy for a standard breast (50 mm compressed breast thickness with 50% glandularity). For each study, one to three simulated lesions were added to each of 40 images (abnormals) while 50 were kept without lesions (normals). Two levels of simulated system noise were added to the images yielding two new image sets, corresponding to simulated dose levels of 50% and 30% of the original images (100%). The manufacturer's standard display processing was subsequently applied to all images. Four radiologists experienced in mammography evaluated the images by searching for lesions and marking and assigning confidence levels to suspicious regions. The search data were analyzed using jackknife free-response (JA-FROC) methodology. For the detection of masses, the mean figure-of-merit (FOM) averaged over all readers was 0.74, 0.71, and 0.68 corresponding to dose levels of 100%, 50%, and 30%, respectively. These values were not statistically different from each other (F= 1.67, p=0.19) but showed a decreasing trend. In contrast, in the microcalcification study the mean FOM was 0.93, 0.67, and 0.38 for the same dose levels and these values were all significantly different from each other (F = 109.84, p < 0.0001). The results indicate that lowering the present dose level by a factor of two compromised the detection of microcalcifications but had a weaker effect on mass detection.
