    • Biochar, a promising carbon-rich and carbon-negative material, can control water pollution, harness the synergy of sustainable development goals, and achieve circular economy. This study examined the performance feasibility of treating fluoride-contaminated surface and groundwater using raw and modified biochar synthesized from agricultural waste rice husk as problem-fixing renewable carbon-neutral material. Physicochemical characterizations of raw/modified biochars were investigated using FESEM-EDAX, FTIR, XRD, BET, CHSN, VSM, pHpzc, Zeta potential, and particle size analysis were analyzed to identify the surface morphology, functional groups, structural, and electrokinetic behavior. In fluoride (Fˉ) cycling, performance feasibility was tested at various governing factors, contact time (0–120 min), initial Fˉ levels (10–50 mg L−1), biochar dose (0.1–0.5 g L−1), pH (2–9), salt strengths (0–50 mM), temperatures (301–328 K), and various co-occurring ions. Results revealed that activated magnetic biochar (AMB) possessed higher adsorption capacity than raw biochar (RB) and activated biochar (AB) at pH 7. The results indicated that maximum Fˉ removal (98.13%) was achieved using AMB at pH 7 for 10 mg L−1. Electrostatic attraction, ion exchange, pore fillings, and surface complexation govern Fˉ removal mechanisms. Pseudo-second-order and Freundlich were the best fit kinetic and isotherm for Fˉ sorption, respectively. Increased biochar dose drives an increase in active sites due to Fˉ level gradient and mass transfer between biochar-fluoride interactions, which reported maximum mass transfer for AMB than RB and AB. Fluoride adsorption using AMB could be described through chemisorption processes at room temperature (301 K), though endothermic sorption follows the physisorption process. Fluoride removal efficiency reduced, from 67.70% to 53.23%, with increased salt concentrations from 0 to 50 mM NaCl solutions, respectively, due to increased hydrodynamic diameter. Biochar was used to treat natural fluoride-contaminated surface and groundwater in real-world problem-solving measures, showed removal efficiency of 91.20% and 95.61%, respectively, for 10 mg L−1 Fˉ contamination, and has been performed multiple times after systematic adsorption-desorption experiments. Lastly, techno-economic analysis was analyzed for biochar synthesis and Fˉ treatment performance costs. Overall, our results revealed worth output and concluded with recommendations for future research on Fˉ adsorption using biochar.

    • This research article presents a comprehensive investigation into the corrosion inhibition performance of a 1,2,3-triazole-based hybrid (TCA) with incorporated triazole, chalcone, and amide moieties. The amalgamation of these pharmacologically active moieties synergistically enhances the ability of the TCA hybrid to form protective films on metal surfaces. The study employs a combination of experimental and theoretical methods to assess the corrosion inhibition properties. Multiple sophisticated techniques like weight loss, EIS, PDP, SEM, EDX, and UV–Vis spectroscopy reveals TCA's exceptional inhibitory efficiency (∼99 % at 200 ppm) for mild steel (MS). PDP analysis suggests a mixed-type inhibition mechanism. The experimental findings are further supported by the DFT calculations by exploring the interactions between the inhibitor and mild steel (MS). The remarkable corrosion inhibition efficacy of TCA along with its eco-friendly behavior aligns the present work with the growing global emphasis on sustainability.

  • A non-classical route of efficient plant uptake verified with fluorescent nanoparticles and root adhesion forces investigated using AFM

    • Classical plant uptake is limited to hydrophilic or water-dispersible material. Therefore, in order to test the uptake behaviour of hydrophobic particles, here, we tested the fate of hydrophobic particles (oleylamine coated Cu2-xSe NPs (CS@OA)) in comparison to hydrophilic particles (chitosan-coated Cu2-xSe NPs (CS@CH)) by treatment on the plant roots. Surprisingly, hydrophobic CS@OA NPs have been found to be ~ 1.3 times more efficient than hydrophilic CS@CH NPs in tomato plant root penetration. An atomic force microscopy (AFM) adhesion force experiment confirms that hydrophobic NPs experience non-spontaneous yet energetically favorable root trapping and penetration. Further, a relative difference in the hydrophobic vs. hydrophilic NPs movement from roots to shoots has been observed and found related to the change in protein corona as identified by two dimensional-polyacrylamide gel electrophoresis (2D-PAGE) analysis. Finally, the toxicity assays at the give concentration showed that Cu2-xSe NPs lead to non-significant toxicity as compared to control. This technology may find an advantage in fertilizer application.

    • In the present investigation with seeds of two important medicinal plants viz. Bixa orellana L.(Annatto) and Clitoria ternatea L., optimal conditions for seed germination in Annatto were determined by subjecting the seeds to varied concentration of plant growth regulators earlier but hardseededness could not be broken. Thereafter, acid scarification (AS) with H2SO4 was done. Freshly harvested seeds (FH) and one year old (OYO) seeds were taken for the study. The study on effectivity of sulfuric acid at elevated moisture content (60 %) has been amply demonstratedin enhancing germination and reducing MGT in B. orellana. In C. ternatea, a 3×4×3 factorial experiment was set up to test the effects of moisture content with three levels of moisture of 12%, 10% and 5%, storage duration at 4 levels of 3, 6, 9 and 12 months and storage temperature at 3 levels of 30ºC, 20ºC and -20ºC on the seed germination. All pretreatments significantly reduced the percentage of hard seeds as compared to non treated control seeds in Annatto. The percentage of hard seed decreased gradually as the pretreatment duration increased for all samples. The final germination percentage of all accessions, was significantly increased by the pretreatments. The total increase on average was 43.5 percent point for all samples (from 36.7 % of control to 80.2 % of pretreatments) in Annatto. The study suggests that the seeds of C. ternatea may possess physiological dormancy and seeds are moisture sensitive and therefore, altering moisture level is an effective method for breaking its physiological dormancy.

    • It is estimated that 350 million individuals worldwide suffer from rare diseases, which are predominantly caused by mutation in a single gene(1). The current molecular diagnostic rate is estimated at 50%, with whole-exome sequencing (WES) among the most successful approaches(2-5). For patients in whom WES is uninformative, RNA sequencing (RNA-seq) has shown diagnostic utility in specific tissues and diseases(6-8). This includes muscle biopsies from patients with undiagnosed rare muscle disorders(6,9), and cultured fibroblasts from patients with mitochondrial disorders(7). However, for many individuals, biopsies are not performed for clinical care, and tissues are difficult to access. We sought to assess the utility of RNA-seq from blood as a diagnostic tool for rare diseases of different pathophysiologies. We generated whole-blood RNA-seq from 94 individuals with undiagnosed rare diseases spanning 16 diverse disease categories. We developed a robust approach to compare data from these individuals with large sets of RNA-seq data for controls (n = 1,594 unrelated controls and n = 49 family members) and demonstrated the impacts of expression, splicing, gene and variant filtering strategies on disease gene identification. Across our cohort, we observed that RNA-seq yields a 7.5% diagnostic rate, and an additional 16.7% with improved candidate gene resolution.

    • Background: Each year, 2.2 million intrapartum-related deaths (intrapartum stillbirths and first day neonatal deaths) occur worldwide with 99% of them taking place in low- and middle-income countries. Despite the accelerated increase in the proportion of deliveries taking place in health facilities in these settings, the stillborn and neonatal mortality rates have not reduced proportionately. Poor quality of care in health facilities is attributed to two-thirds of these deaths. Improving quality of care during the intrapartum period needs investments in evidence-based interventions. We aim to evaluate the quality improvement packageScaling Up Safer Bundle Through Quality Improvement in Nepal (SUSTAIN)on intrapartum care and intrapartum-related mortality in public hospitals of Nepal.Methods: We will conduct a stepped wedge cluster randomized controlled trial in eight public hospitals with each having least 3000 deliveries a year. Each hospital will represent a cluster with an intervention transition period of 2months in each. With a level of significance of 95%, the statistical power of 90% and an intra-cluster correlation of 0.00015, a study period of 19months should detect at least a 15% change in intrapartum-related mortality. Quality improvement training, mentoring, systematic feedback, and a continuous improvement cycle will be instituted based on bottleneck analyses in each hospital. All concerned health workers will be trained on standard basic neonatal resuscitation and essential newborn care. Portable fetal heart monitors (Moyo (R)) and neonatal heart rate monitors (Neobeat (R)) will be introduced in the hospitals to identify fetal distress during labor and to improve neonatal resuscitation. Independent research teams will collect data in each hospital on intervention inputs, processes, and outcomes by reviewing records and carrying out observations and interviews. The dose-response effect will be evaluated through process evaluations.Discussion: With the global momentum to improve quality of intrapartum care, better understanding of QI package within a health facility context is important. The proposed package is based on experiences from a similar previous scale-up trial carried out in Nepal. The proposed evaluation will provide evidence on QI package and technology for implementation and scale up in similar settings.

  • Correlation of Dengue and Meteorological Factors in Bangladesh : A Public Health Concern

    • Dengue virus (DENV) is an enveloped, single-stranded RNA virus, a member of the Flaviviridae family (which causes Dengue fever), and an arthropod-transmitted human viral infection. Bangladesh is well known for having some of Asia’s most vulnerable Dengue outbreaks, with climate change, its location, and it’s dense population serving as the main contributors. For speculation about DENV outbreak characteristics, it is crucial to determine how meteorological factors correlate with the number of cases. This study used five time series models to observe the trend and forecast Dengue cases. Current data-based research has also applied four statistical models to test the relationship between Dengue-positive cases and meteorological parameters. Datasets were used from NASA for meteorological parameters, and daily DENV cases were obtained from the Directorate General of Health Service (DGHS) open-access websites. During the study period, the mean of DENV cases was 882.26 ± 3993.18, ranging between a minimum of 0 to a maximum of 52,636 daily confirmed cases. The Spearman’s rank correlation coefficient between climatic variables and Dengue incidence indicated that no substantial relationship exists between daily Dengue cases and wind speed, temperature, and surface pressure (Spearman’s rho; r = −0.007, p > 0.05; r = 0.085, p > 0.05; and r = −0.086, p > 0.05, respectively). Still, a significant relationship exists between daily Dengue cases and dew point, relative humidity, and rainfall (r = 0.158, p < 0.05; r = 0.175, p < 0.05; and r = 0.138, p < 0.05, respectively). Using the ARIMAX and GA models, the relationship for Dengue cases with wind speed is −666.50 [95% CI: −1711.86 to 378.86] and −953.05 [−2403.46 to 497.36], respectively. A similar negative relation between Dengue cases and wind speed was also determined in the GLM model (IRR = 0.98). Dew point and surface pressure also represented a negative correlation in both ARIMAX and GA models, respectively, but the GLM model showed a positive association. Additionally, temperature and relative humidity showed a positive correlation with Dengue cases (105.71 and 57.39, respectively, in the ARIMAX, 633.86, and 200.03 in the GA model). In contrast, both temperature and relative humidity showed negative relation with Dengue cases in the GLM model. In the Poisson regression model, windspeed has a substantial significant negative connection with Dengue cases in all seasons. Temperature and rainfall are significantly and positively associated with Dengue cases in all seasons. The association between meteorological factors and recent outbreak data is the first study where we are aware of the use of maximum time series models in Bangladesh. Taking comprehensive measures against DENV outbreaks in the future can be possible through these findings, which can help fellow researchers and policymakers.
