  • Factor VIII gene inversions in severe hemophilia A : Results of an international consortium study

    • Twenty-two molecular diagnostic laboratories from 14 countries participated in a consortium study to estimate the impact of Factor VIII gene inversions in severe hemophilia A. A total of 2,093 patients with severe hemophilia A were studied; of those, 740 (35%) had a type 1 (distal) factor VIII inversion, and 140 (7%) showed a type 2 (proximal) inversion. In 25 cases, the molecular analysis showed additional abnormal or polymorphic patterns. Ninety-eight percent of 532 mothers of patients with inversions were carriers of the abnormal factor VIII gene; when only mothers of nonfamilial cases were studied, 9 de novo inversions in maternal germ cells ware observed among 225 cases (≃ 1 de novo maternal origin of the inversion in 25 mothers of sporadic cases). When the maternal grandparental origin was examined, the inversions occurred de novo in male germ cells in 69 cases and female germ cells in 1 case. The presence of factor VIII inversions is not a major predisposing factor for the development of factor VIII inhibitors; however, slightly more patients with severe hemophilia A and factor VIII inversions develop inhibitors (130 of 642 [20%]) than patients with severe hemophilia A without inversions (131 of 821 [16%]).

    • Cities currently harbour more than half of the world's human population and continued urban expansion replaces natural landscapes and increases habitat fragmentation. The impacts of urbanisation on biodiversity have been extensively studied in some parts of the world, but there is limited information from South Asia, despite the rapid expansion of cities in the region. Here, we present the results of monthly surveys of butterflies in three urban parks in Dhaka city, Bangladesh, over a 3-year period (January 2014 to December 2016). We recorded 45% (137 of the 305 species) of the country's butterfly richness, and 40% of the species detected are listed as nationally threatened. However, butterfly species richness declined rapidly in the three study areas over the 3-year period, and the decline appeared to be more severe among threatened species. We developed linear mixed effect models to assess the relationship between climatic variables and butterfly species richness. Overall, species richness was positively associated with maximum temperature and negatively with mean relative humidity and saturation deficit. Our results demonstrate the importance of urban green spaces for nationally threatened butterflies. With rapidly declining urban green spaces in Dhaka and other South Asian cities, we are likely to lose refuges for threatened fauna. There is an urgent need to understand urban biodiversity dynamics in the region, and for proactive management of urban green spaces to protect butterflies in South Asia.

  • Pegylated bis-indolyl polyurethane dendrimer : Empty drug carrier with prominent anticancer activity

    • Blocked isocyanate-terminated, bioactive bis-indole based polyurethane dendrimers of generation G0 – G3 were subjected to urethane interchange reaction with poly(ethylene glycol) monomethyl ether; the reactions yielded corresponding PEGylated dendrimers. The structures of the PEGylated dendrimers were confirmed using spectroscopic and SEC-MALS techniques. The PDI of the polymers were found between 1.24 and 1.49 and these values were on par with 1.46 of poly(ethylene glycol) monomethyl ether used. All the PEGylated dendrimers were found to have a prominent cytotoxicity in human breast (MDA MB-231) and lung (A549) cancer cells. Based on the MTT, AO/EtBr staining and ROS assay results, PEGylated G2 and G3 dendrimers were further subjected to determination of apoptosis using protein markers; the pro-apoptotic markers BaX and caspase-3 were found up regulated and the anti-apoptotic marker Bcl-2 was down regulated. The results confirmed that the PEGylated bis-indolyl G3 polyurethane dendrimer – an empty drug carrier – itself had strong tendency to activate apoptosis type cell death in human cancer cells.

  • Elimination of 50% Iodine and Excellent Performance of Dye-Sensitized Solar Cell Enabled by TEMPO Radical Dendrimer-Iodide Dual Redox Systems

    • Detrimental effect of counter electrode corrosion, electrolyte leakage problem, and sublimation of iodine ions affect the performance and stability of dye-sensitized solar cells (DSSCs) based on a liquid redox electrolyte system. TEMPO/TEMPO+ is the most desirable one to replace the existing redox couples in liquid electrolyte owing to its rapid and reversible one electron kinetics, high diffusion coefficient, and electrochemical stability. In the present study, a fifth generation polyurethane dendrimer end-capped with 2,2,6,6-tetramethylpiperidin-1-oxyl (TEMPO) was prepared and employed as an active redox mediator to replace up to 75% of iodine ions in the standard liquid iodide electrolyte system to elucidate the effect of a quick self-exchange reaction in DSSCs. In this concern, quasi-gel type electrolytes were prepared by incorporating different amounts of a newly prepared TEMPO radical dendrimer to a liquid I-/I3- redox system and were used as a dual redox mediator for DSSCs. Blending of 50 wt % of I-/I3- electrolyte with 50 wt % of the radical dendrimer improved the device performance (η) unprecedentedly from 6.56 to 9.54% upon illumination with 1 sun intensity. Thorough electrochemical characterization of cells was carried out and all such parameters authenticated this remarkable enhancement. Stability of the device was also found to be good, which retained its >80% efficiency after 30 days.

  • Strong regional influence of climatic forcing datasets on global crop model ensembles

    • We present results from the Agricultural Model Intercomparison and Improvement Project (AgMIP) Global Gridded Crop Model Intercomparison (GGCMI) Phase I, which aligned 14 global gridded crop models (GGCMs) and 11 climatic forcing datasets (CFDs) in order to understand how the selection of climate data affects simulated historical crop productivity of maize, wheat, rice and soybean. Results show that CFDs demonstrate mean biases and differences in the probability of extreme events, with larger uncertainty around extreme precipitation and in regions where observational data for climate and crop systems are scarce. Countries where simulations correlate highly with reported FAO national production anomalies tend to have high correlations across most CFDs, whose influence we isolate using multi-GGCM ensembles for each CFD. Correlations compare favorably with the climate signal detected in other studies, although production in many countries is not primarily climate-limited (particularly for rice). Bias-adjusted CFDs most often were among the highest model-observation correlations, although all CFDs produced the highest correlation in at least one top-producing country. Analysis of larger multi-CFD-multi-GGCM ensembles (up to 91 members) shows benefits over the use of smaller subset of models in some regions and farming systems, although bigger is not always better. Our analysis suggests that global assessments should prioritize ensembles based on multiple crop models over multiple CFDs as long as a top-performing CFD is utilized for the focus region.
