  • Reconstructing the Younger Dryas ice dammed lake in the Baltic Basin: Bathymetry, area and volume

    • A digital 3D-reconstruction of the Baltic Ice Lake's (BIL) configuration during the termination of the Younger Dryas cold phase (ca. 11700 cal. yr BP) was compiled using a combined bathymetric-topographic Digital Terrain Model (DTM), Scandinavian ice sheet limits, Baltic Sea Holocene bottom sediment thickness information, and a paleoshoreline database maintained at the Lund University. The bathymetric-topographic DTM, assembled from publicly available data sets, has a resolution of 500 X 500 m on Lambert Azimuthal Equal Area projection allowing area and volume calculations of the BIL to be made with an unprecedented accuracy. When the damming Scandinavian ice sheet margin eventually retreated north of Mount Billingen, the high point in terrain of Southern central Sweden bordering to lower terrain further to the north, the BIL was catastrophically drained resulting in a 25 m drop of the lake level. With our digital reconstruction, we estimate that approximately 7800 km(3) of water drained during this event and that the ice dammed lake area was reduced by ca. 18%. Building on previous results suggesting drainage over 1 to 2 years, our lake volume calculations imply that the freshwater flux to the contemporaneous sea in the west was between about 0.12 and 0.25 Sv. The BIL reconstruction provides new detailed information on the paleogeography in the area of southern Scandinavia, both before and after the drainage event, with implications for interpretations of geological records concerning the post-glacial environmental development.     • In chemical imaging spectra are acquired over a surface with one spectrum for each pixel of the image. The obtained spectra usually carry a mixture of chemical and physical information. One may view the properties that vary over the image, the mean spectral magnitude from separate wavelength intervals, or better, PCA scores may be shown as images.In this way a multitude of images are compressed to a few images that in the PCA case are representative for the main variation in the sample images. These images may be viewed manually and deductions as to e.g. differences in homogeneity can be made. At an increased rate of samples, the observer will have difficulties coping with the repetitive work and different observers will most likely have slightly different interpretations. In order to automate the process of estimation of e.g. homogeneity and particle density, image filters can be used to calculate a small set of texture descriptors for each image. Calculations based on the 2D versions of the discrete wavelet transform (DWT) using Daubechies 14 and the dual tree complex wavelet transform (DT-CWT) using near-symmetric 13, 19 tap filters in combination with q-shift 14, 14 tap filters were evaluated for this purpose.The aim with this work is to evaluate texture descriptors based on a combination of 2D-wavelet filters and energy, i.e. l(1)-norm, calculations for each wavelet scale. These descriptors are then used as observations for overview in e.g. PCA. In this way the texture differences can be ranked by ordinary use of PCA or PLS.This method is tested on multivariate near infrared images of pharmaceutical tablets. Score images are selected to represent variations of the aggregate density and sizes in the compressed tablets. Images are shifted and rotated to compare shift and rotational independence of the texture descriptors.

  • Investigation of interface properties of Ni/Cu multilayers by high kinetic energy photoelectron spectroscopy

    • High kinetic-energy photoelectron spectroscopy (HIKE) or hard x-ray photoelectron spectroscopy has been used to investigate the alloying of Ni/Cu (100) multilayers. Relative intensities of the corelevels and their chemical shifts derived from binding energy changes are shown to give precise information on physicochemical properties and quality of the buried layers. Interface roughening, including kinetic properties such as the rate of alloying, and temperature effects on the processes can be analyzed quantitatively. Using HIKE, we have been able to precisely follow the deterioration of the multilayer structure at the atomic scale and observe the diffusion of the capping layer into the multilayer structure which in turn is found to lead to a segregation in the ternary system. This is of great importance for future research on multilayered systems of this kind. Our experimental data are supplemented by first-principles theoretical calculations of the core-level shifts for a ternary alloy to allow for modeling of the influence of capping materials on the chemical shifts.

  • Activity of lactoperoxidase when adsorbed on protein layers

    • Lactoperoxidase (LPO) is an enzyme, which is used as an antimicrobial agent in a number of applications, e.g., food technology. In the majority of applications LPO is added to a homogeneous product phase or immobilised on product surface. In the latter case, however, the measurements of LPO activity are seldom reported. In this paper we have assessed LPO enzymatic activity on bare and protein modified gold surfaces by means of electrochemistry. It was found that LPO rapidly adsorbs to bare gold surfaces resulting in an amount of LPO adsorbed of 2.9 mg/m2. A lower amount of adsorbed LPO is obtained if the gold surface is exposed to bovine serum albumin, bovine or human mucin prior to LPO adsorption. The enzymatic activity of the adsorbed enzyme is in general preserved at the experimental conditions and varies only moderately when comparing bare gold and gold surface pretreated with the selected proteins. The measurement of LPO specific activity, however, indicate that it is about 1.5 times higher if LPO is adsorbed on gold surfaces containing a small amount of preadsorbed mucin in comparison to the LPO directly adsorbed on bare gold.

    • The valence electronic structure of a series of molecular films containing ruthenium polypyridine complexes has been investigated by photoelectron spectroscopy (PES) at high kinetic energy (HIKE) using hard X-ray. The experiment shows the possibility to experimentally probe the metal contribution to the valence spectra in a bulk sensitive mode. Specifically to directly follow the Ru 4d contribution to the highest occupied molecular orbitals of such complexes. The experimental spectra are accurately modeled by DFT calculations only if a crystal structure environment is taken into account showing the importance of intermolecular interaction for modeling the electronic structure of such complexes.

  • Quasispecies dynamics and molecular evolution of human norovirus capsid P region during chronic infection

    • In this novel study, we have for the first time identified evolutionarily conserved capsid residues in an individual chronically infected with norovirus (GGII.3). From 2000 to 2003, a total of 147 P1-1 and P2 capsid sequences were sequenced and investigated for evolutionarily conserved and functionally important residues by the evolutionary trace (ET) algorithm. The ET algorithm revealed more absolutely conserved residues (ACR) in the P1-1 domain (47/53, 88 %) as compared with the P2 domain (86/133, 64 %). The capsid P1-1 and P2 domains evolved in time-dependent manner, with a distinct break point observed between autumn/winter of year 2000 (isolates P1, P3 and P5) and spring to autumn of year 2001 (isolates P11, P13 and P15), which presumably coincided with a change of clinical symptoms. Furthermore, the ET analysis revealed a similar receptor-binding pattern as reported for Norwalk and VA387 strains, with the CS-4 and CS-5 patch (Norwalk strain) including residues 329 and 377 and residues 306 and 310, respectively, all being ACR in all partitions. Most interesting was that residues 343, 344, 345, 374, 390 and 391 of the proposed receptor A and B trisaccharide binding site (VA387 strain) within the P2 domain remained ACR in all partitions, presumably because there was no selective advantage to alter the histo blood group antigens (HBGA) receptor binding specificity. In conclusion, this study provides novel insights to the evolutionary process of norovirus during chronic infection.

    • Abstract In situ ellipsometry was used to study layer-by-layer film formation on hydrophilic and hydrophobized silica surfaces by alternating sequential adsorption of human mucin MUC5B and cationic proteins lysozyme, lactoferrin, lactoperoxidase or histatin 5, respectively. The stability of the multilayers was investigated by addition of sodium dodecyl sulfate solution (SDS). Atomic force microscopy was employed to investigate morphological structures on the surfaces during the layer-by-layer film build-up. It was clearly shown that, on both hydrophilic and hydrophobized silica, only MUC5B and lactoperoxidase showed the ability for multilayer formation, resulting in an approximately linear increase in adsorbed amount and film thickness with each deposition cycle. The net increase in amounts per cycle was larger on the hydrophilic silica. Further, MUC5B needs to be adsorbed first on the hydrophilic substrates to obtain this fast build-up behavior. Generally, addition of SDS solution showed that a large fraction of the adsorbed film could be desorbed. However, films on the hydrophobized silica were more resistant to surfactant elution. In conclusion, MUC5B–cationic protein multilayers can be formed on hydrophilic and hydrophobized silica, depending on the choice of the cationic protein as well as in which order the build-up is started on hydrophilic silica. Additionally, SDS disrupts the layer-by-layer film formed by MUC5B and lactoperoxidase.
