  • Monitoring the large-scale magnetic field of AD Leo with SPIRou, ESPaDOnS, and Narval : Towards a magnetic polarity reversal?

    • Context: One clear manifestation of dynamo action on the Sun is the 22-yr magnetic cycle, exhibiting a polarity reversal and a periodic conversion between poloidal and toroidal fields. For M dwarfs, several authors claim evidence of activity cycles from photometry and analyses of spectroscopic indices, but no clear polarity reversal has been identified from spectropolarimetric observations. These stars are excellent laboratories to investigate dynamo-powered magnetic fields under different stellar interior conditions, that is partly or fully convective.Aims: Our aim is to monitor the evolution of the large-scale field of AD Leo, which has shown hints of a secular evolution from past dedicated spectropolarimetric campaigns. This is of central interest to inform distinct dynamo theories, contextualise the evolution of the solar magnetic field, and explain the variety of magnetic field geometries observed in the past.Methods: We analysed near-infrared spectropolarimetric observations of the active M dwarf AD Leo taken with SPIRou between 2019 and 2020 and archival optical data collected with ESPaDOnS and Narval between 2006 and 2019. We searched for long-term variability in the longitudinal field, the width of unpolarised Stokes profiles, the unsigned magnetic flux derived from Zeeman broadening, and the geometry of the large-scale magnetic field using both Zeeman-Doppler imaging and principal component analysis.Results: We found evidence of a long-term evolution of the magnetic field, featuring a decrease in axisymmetry (from 99% to 60%). This is accompanied by a weakening of the longitudinal field (-300 to -50 G) and a correlated increase in the unsigned magnetic flux (2.8-3.6 kG). Likewise, the width of the mean profile computed with selected near-infrared lines manifests a long-term evolution corresponding to field strength changes over the full time series, but does not exhibit modulation with the stellar rotation of AD Leo in individual epochs.Conclusions: The large-scale magnetic field of AD Leo manifested first hints of a polarity reversal in late 2020 in the form of a substantially increased dipole obliquity, while the topology remained predominantly poloidal and dipolar for 14 yr. This suggests that low-mass M dwarfs with a dipole-dominated magnetic field can undergo magnetic cycles.

  • GWAS Meta-Analysis of Suicide Attempt: Identification of 12 Genome-Wide Significant Loci and Implication of Genetic Risks for Specific Health Factors.

    • Suicidal behavior is heritable and is a major cause of death worldwide. Two large-scale genome-wide association studies (GWASs) recently discovered and cross-validated genome-wide significant (GWS) loci for suicide attempt (SA). The present study leveraged the genetic cohorts from both studies to conduct the largest GWAS meta-analysis of SA to date. Multi-ancestry and admixture-specific meta-analyses were conducted within groups of significant African, East Asian, and European ancestry admixtures.This study comprised 22 cohorts, including 43,871 SA cases and 915,025 ancestry-matched controls. Analytical methods across multi-ancestry and individual ancestry admixtures included inverse variance-weighted fixed-effects meta-analyses, followed by gene, gene-set, tissue-set, and drug-target enrichment, as well as summary-data-based Mendelian randomization with brain expression quantitative trait loci data, phenome-wide genetic correlation, and genetic causal proportion analyses.Multi-ancestry and European ancestry admixture GWAS meta-analyses identified 12 risk loci at p values <5×10-8. These loci were mostly intergenic and implicated DRD2, SLC6A9, FURIN, NLGN1, SOX5, PDE4B, and CACNG2. The multi-ancestry SNP-based heritability estimate of SA was 5.7% on the liability scale (SE=0.003, p=5.7×10-80). Significant brain tissue gene expression and drug set enrichment were observed. There was shared genetic variation of SA with attention deficit hyperactivity disorder, smoking, and risk tolerance after conditioning SA on both major depressive disorder and posttraumatic stress disorder. Genetic causal proportion analyses implicated shared genetic risk for specific health factors.This multi-ancestry analysis of suicide attempt identified several loci contributing to risk and establishes significant shared genetic covariation with clinical phenotypes. These findings provide insight into genetic factors associated with suicide attempt across ancestry admixture populations, in veteran and civilian populations, and in attempt versus death.

  • Pyxis : a ground-based demonstrator for formation-flying optical interferometry

    • In the past few years, there has been a resurgence in studies of space-based optical/ infrared interferometry, particularly with the vision to use the technique to discover and characterize temperate Earth-like exoplanets around solar analogs. One of the key technological leaps needed to make such a mission feasible is demonstrating that formation flying precision at the level needed for interferometry is possible. Here, we present Pyxis, a ground-based demonstrator for a future small satellite mission with the aim to demonstrate the precision metrology needed for spacebased interferometry. We describe the science potential of such a ground-based instrument and detail the various subsystems: three six-axis robots, a multi-stage metrology system, an integrated optics beam combiner, and the control systems required for the necessary precision and stability. We conclude by looking toward the next stage of Pyxis: a collection of small satellites in Earth orbit.

  • Immunomodulatory Nanoparticles for Modulating Arthritis Flares

    • Disease-modifying drugs have improved the treatment for autoimmune joint disorders, such as rheumatoid arthritis, but inflammatory flares are a common experience. This work reports the development and application of flare-modulating poly(lactic-co-glycolic acid)-poly(ethylene glycol)-maleimide (PLGA-PEG-MAL)-based nanoparticles conjugated with joint-relevant peptide antigens, aggrecan70-84 and type 2 bovine collagen256-270. Peptide-conjugated PLGA-PEG-MAL nanoparticles encapsulated calcitriol, which acted as an immunoregulatory agent, and were termed calcitriol-loaded nanoparticles (CLNP). CLNP had a ∼200 nm hydrodynamic diameter with a low polydispersity index. In vitro, CLNP induced phenotypic changes in bone marrow derived dendritic cells (DC), reducing the expression of costimulatory and major histocompatibility complex class II molecules, and proinflammatory cytokines. Bulk RNA sequencing of DC showed that CLNP enhanced expression of Ctla4, a gene associated with downregulation of immune responses. In vivo, CLNP accumulated in the proximal lymph nodes after intramuscular injection. Administration of CLNP was not associated with changes in peripheral blood cell numbers or cytokine levels. In the collagen-induced arthritis and SKG mouse models of autoimmune joint disorders, CLNP reduced clinical scores, prevented bone erosion, and preserved cartilage proteoglycan, as assessed by high-resolution microcomputed tomography and histomorphometry analysis. The disease protective effects were associated with increased CTLA-4 expression in joint-localized DC and CD4+ T cells but without generalized suppression of T cell-dependent immune response. The results support the potential of CLNP as modulators of disease flares in autoimmune arthropathies.

  • Diminishing benefits of urban living for children and adolescents' growth and development

    • Optimal growth and development in childhood and adolescence is crucial for lifelong health and well-being1–6. Here we used data from 2,325 population-based studies, with measurements of height and weight from 71 million participants, to report the height and body-mass index (BMI) of children and adolescents aged 5–19 years on the basis of rural and urban place of residence in 200 countries and territories from 1990 to 2020. In 1990, children and adolescents residing in cities were taller than their rural counterparts in all but a few high-income countries. By 2020, the urban height advantage became smaller in most countries, and in many high-income western countries it reversed into a small urban-based disadvantage. The exception was for boys in most countries in sub-Saharan Africa and in some countries in Oceania, south Asia and the region of central Asia, Middle East and north Africa. In these countries, successive cohorts of boys from rural places either did not gain height or possibly became shorter, and hence fell further behind their urban peers. The difference between the age-standardized mean BMI of children in urban and rural areas was <1.1 kg m–2 in the vast majority of countries. Within this small range, BMI increased slightly more in cities than in rural areas, except in south Asia, sub-Saharan Africa and some countries in central and eastern Europe. Our results show that in much of the world, the growth and developmental advantages of living in cities have diminished in the twenty-first century, whereas in much of sub-Saharan Africa they have amplified.

  • Statistical uncertainty analysis-based precipitation merging (SUPER) : A new framework for improved global precipitation estimation

    • Multi-source merging is an established tool for improving large-scale precipitation estimates. Existing merging frameworks typically use gauge-based precipitation error statistics and neglect the inter-dependence of various precipitation products. However, gauge-observation uncertainties at daily and sub-daily time scales can bias merging weights and yield sub-optimal precipitation estimates, particularly over data-sparse regions. Likewise, frameworks ignoring inter-product error cross-correlation will overfit precipitation observation noise. Here, a Statistical Uncertainty analysis-based Precipitation mERging framework (SUPER) is proposed for addressing these challenges. Specifically, a quadruple collocation analysis is employed to estimate precipitation error variances and covariances for commonly used precipitation products. These error estimates are subsequently used for merging all products via a least-squares minimization approach. In addition, false-alarm precipitation events are removed via a reference rain/no-rain time series estimated by a newly developed categorical variable merging method. As such, SUPER does not require any rain gauge observations to reduce daily random and rain/no-rain classification errors. Additionally, by considering precipitation product inter-dependency, SUPER avoids overfitting measurement noise present in multi-source precipitation products. Results show that the overall RMSE of SUPER-based precipitation is 3.35 mm/day and the daily correlation with gauge observations is 0.71 [−] – metrics that are generally superior to recent precipitation reanalyses and remote sensing products. In this way, we seek to propose a new framework for robustly generating global precipitation datasets that can improve land surface and hydrological modeling skill in data-sparse regions.

  • Combining hypothesis- and data-driven neuroscience modeling in FAIR workflows

    • Modeling in neuroscience occurs at the intersection of different points of view and approaches. Typically, hypothesis-driven modeling brings a question into focus so that a model is constructed to investigate a specific hypothesis about how the system works or why certain phenomena are observed. Data-driven modeling, on the other hand, follows a more unbiased approach, with model construction informed by the computationally intensive use of data. At the same time, researchers employ models at different biological scales and at different levels of abstraction. Combining these models while validating them against experimental data increases understanding of the multiscale brain. However, a lack of interoperability, transparency, and reusability of both models and the workflows used to construct them creates barriers for the integration of models representing different biological scales and built using different modeling philosophies. We argue that the same imperatives that drive resources and policy for data - such as the FAIR (Findable, Accessible, Interoperable, Reusable) principles - also support the integration of different modeling approaches. The FAIR principles require that data be shared in formats that are Findable, Accessible, Interoperable, and Reusable. Applying these principles to models and modeling workflows, as well as the data used to constrain and validate them, would allow researchers to find, reuse, question, validate, and extend published models, regardless of whether they are implemented phenomenologically or mechanistically, as a few equations or as a multiscale, hierarchical system. To illustrate these ideas, we use a classical synaptic plasticity model, the Bienenstock-Cooper-Munro rule, as an example due to its long history, different levels of abstraction, and implementation at many scales.
