    • We introduce the 4-metre Multi-Object Spectroscopic Telescope (4MOST), a new high-multiplex, wide-field spectroscopic survey facility under development for the four-metre-class Visible and Infrared Survey Telescope for Astronomy (VISTA) at Paranal. Its key specifications are: a large field of view (FoV) of 4.2 square degrees and a high multiplex capability, with 1624 fibres feeding two low-resolution spectrographs (R = λ/Δλ ~ 6500), and 812 fibres transferring light to the high-resolution spectrograph (R ~ 20 000). After a description of the instrument and its expected performance, a short overview is given of its operational scheme and planned 4MOST Consortium science; these aspects are covered in more detail in other articles in this edition of The Messenger. Finally, the processes, schedules, and policies concerning the selection of ESO Community Surveys are presented, commencing with a singular opportunity to submit Letters of Intent for Public Surveys during the first five years of 4MOST operations.

  • Sloan Digital Sky Survey IV : Mapping the Milky Way, Nearby Galaxies, and the Distant Universe

    • We describe the Sloan Digital Sky Survey IV (SDSS-IV), a project encompassing three major spectroscopic programs. The Apache Point Observatory Galactic Evolution Experiment 2 (APOGEE-2) is observing hundreds of thousands of Milky Way stars at high resolution and. high signal-to-noise ratios in the near-infrared. The Mapping Nearby Galaxies at Apache Point Observatory (MaNGA) survey is obtaining spatially resolved spectroscopy for thousands of nearby galaxies (median z similar to 0.03). The extended Baryon Oscillation Spectroscopic Survey (eBOSS) is mapping the galaxy, quasar, and neutral gas distributions between z similar to 0.6 and 3.5 to constrain cosmology using baryon acoustic oscillations, redshift space distortions, and the shape of the power spectrum. Within eBOSS, we are conducting two major subprograms: the SPectroscopic IDentification of eROSITA Sources (SPIDERS), investigating X-ray AGNs. and galaxies in X-ray clusters, and the Time Domain Spectroscopic Survey (TDSS), obtaining spectra of variable sources. All programs use the 2.5 m Sloan Foundation Telescope at the. Apache Point Observatory; observations there began in Summer 2014. APOGEE-2 also operates a second near-infrared spectrograph at the 2.5 m du Pont Telescope at Las Campanas Observatory, with observations beginning in early 2017. Observations at both facilities are scheduled to continue through 2020. In keeping with previous SDSS policy, SDSS-IV provides regularly scheduled public data releases; the first one, Data Release 13, was made available in 2016 July.

    • We report calculations of spectroscopic accuracy for 513 states in the B-like ions with Z = 24-30. The energy levels and transition rates are calculated using the second-order many-body perturbation theory, while multiconfiguration Dirac-Hartree-Fock calculations are performed to support the former results. The present two sets of results agree excellently with each other, and can be used as benchmarks in the future. The electron-impact excitation (EIE) collision strengths are calculated based on independent processes and isolated resonances approximation using the distorted wave method. Based on the present atomic data, the spectral lines of these ions are simulated using a collisional radiative model. The amount of energy, transition data, and EIE collision strengths of high accuracy are significantly increased for several B-like ions of astrophysical interest, where experimental data are still very scarce. The results can be used in line identification, plasma modeling, and diagnostics of astrophysical plasmas.

    • Biological membranes do not only occur as planar bilayer structures, but depending on the lipid composition, can also curve into intriguing three-dimensional structures. In order to fully understand the biological implications as well as to reveal the full potential for applications, e.g. for drug delivery and other biomedical devices, of such structures, well-defined model systems are required. Here, we discuss the formation of lipid non-lamellar liquid crystalline (LC) surface layers spin-coated from the constituting lipids followed by hydration of the lipid layer. We demonstrate that hybrid lipid polymer films can be formed with different properties compared with the neat lipid LC layers. The nanostructure and morphologies of the lipid films formed reflect those in the bulk. Most notably, mixed lipid layers, which are composed of glycerol monooleate and diglycerol monooleate with poly(N-isopropylacrylamide) nanogels, can form films of reverse cubic phases that are capable of responding to temperature stimulus. Owing to the presence of the nanogel particles, changing the temperature not only regulates the hydration of the cubic phase lipid films, but also the lateral organization of the lipid domains within the lipid self-assembled film. This opens up the possibility for new nanostructured materials based on lipid-polymer responsive layers.

    • The multiconfiguration Dirac-Hartree-Fock (MCDHF) and relativistic configuration interaction (RCI) methods were used to compute excitation energies and transition data for the 147 lowest states of the even 3s(3)p(4), 3s(2)3p2(3)d, 3p(4)3d, 3s(3)p(2)3d(2) configurations and for the 124 lowest states of the odd 3s23p3, 3p5, 3s3p33d, 3s(2)3p(3)d2, 3p(3)3d(2) configurations for the P-like ions: As XIX, Kr XXII, Sr XXIV, Zr XXVI, Mo XXVIII, and W LX. E1 transition rates and weighted oscillator strengths among these states are given. Valence-valence, core-valence and core-core electron correlation effects are included. Computed excitation energies and transition data are compared with the NIST recommended values and experimental or theoretical results of other authors.     • BACKGROUND: Subendothelial deposited low-density lipoprotein particles are a known inflammatory factor in atherosclerosis. However, the causal components derived from low-density lipoprotein are still poorly defined. Apolipoprotein B100 (ApoB100) is the unexchangeable protein component of low-density lipoprotein, and the progression of atherosclerosis is associated with immune responses to ApoB100-derived peptides. In this study, we analyzed the proinflammatory activity of ApoB100 peptides in atherosclerosis. METHODS AND RESULTS: By screening a peptide library of ApoB100, we identified a distinct native peptide referred to as ApoB100 danger-associated signal 1 (ApoBDS-1), which shows sequence-specific bioactivity in stimulation of interleukin-8, CCL2, and interleukin-6. ApoBDS-1 activates mitogen-activated protein kinase and calcium signaling, thereby effecting the expression of interleukin-8 in innate immune cells. Ex vivo stimulation of carotid plaques with ApoBDS-1 enhances interleukin-8 and prostaglandin E release. Furthermore, we demonstrated that ApoBDS-1-positive peptide fragments are present in atherosclerotic lesions using immunoassays and that low-molecular-weight fractions isolated from plaque show ApoBDS-1 activity inducing interleukin-8 production. CONCLUSIONS: Our data show that ApoBDS-1 is a previously unrecognized peptide with robust proinflammatory activity, contributing to the disease-promoting effects of low-density lipoprotein in the pathogenesis of atherosclerosis.

    • Large-scale multiconfiguration Dirac-Hartree-Fock calculations are provided for the n <= 5 states in C-like ions from O iii to Mg vii. Electron correlation effects are accounted for by using large configuration state function expansions, built from sets of orbitals with principal quantum numbers n <= 10. An accurate and complete data set of excitation energies, wavelengths, radiative transition parameters, and lifetimes is offered for the 156 (196, 215, 272, 318) lowest states of the 2s (2)2p (2), 2s2p (3), 2p (4), 2s (2)2p3s, 2s (2)2p3p, 2s (2)2p3d, 2s2p (2)3s, 2s2p (2)3p, 2s2p (2)3d, 2p (3)3s, 2p (3)3p, 2p (3)3d, 2s (2)2p4s, 2s (2)2p4p, 2s (2)2p4d, 2s (2)2p4f, 2s2p (2)4s, 2s2p (2)4p, 2s2p (2)4d, 2s2p (2)4f, 2s (2)2p5s, 2s (2)2p5p, 2s (2)2p5d, 2s (2)2p5f, and 2s (2)2p5g configurations in O iii (F iv, Ne v, Na vi, Mg vii). By comparing available experimental wavelengths with the MCDHF results, the previous line identifications for the n = 5, 4, 3 -> n = 2 transitions of Na vi in the X-ray and EUV wavelength range are revised. For several previous identifications discrepancies are found, and tentative new (or revised) identifications are proposed. A consistent atomic data set including both energy and transition data with spectroscopic accuracy is provided for the lowest hundreds of states for C-like ions from O iii to Mg vii.

    • Aims. The Lande g-factor is an important parameter in astrophysical spectropolarimetry, used to characterize the response of a line to a given value of the magnetic field. The purpose of this paper is to present accurate Lande g-factors for states in B II, C I-IV, Al I-II, Si I-IV, P II, S II, Cl III, Ar IV, Ca I, Ti II, Zr III, and Sn II.Methods. The multiconfiguration Dirac-Hartree-Fock and relativistic configuration interaction methods, which are implemented in the general-purpose relativistic atomic structure package GRASP2K, are employed in the present work to compute the Lande g-factors for states in B II, C I-IV, Al I-II, Si I-IV, P II, S II, Cl III, Ar IV, Ca I, Ti II, Zr III, and Sn II. The accuracy of the wave functions for the states, and thus the accuracy of the resulting Lande g-factors, is evaluated by comparing the computed excitation energies and energy separations with the National Institute of Standards and Technology (NIST) recommended data.Results. All excitation energies are in very good agreement with the NIST values except for Ti II, which has an average difference of 1.06%. The average uncertainty of the energy separations is well below 1% except for the even states of Al I; odd states of Si I, Ca I, Ti II, Zr III; and even states of Sn II for which the relative differences range between 1% and 2%. Comparisons of the computed Lande g-factors are made with available NIST data and experimental values. Analysing the LS-composition of the wave functions, we quantify the departures from LS-coupling and summarize the states for which there is a difference of more than 10% between the computed Lande g-factor and the Lande g-factor in pure LS-coupling. Finally, we compare the computed Lande g-factors with values from the Kurucz database.

  • Targeted inhibition of ERα signaling and PIP5K1α/Akt pathways in castration‐resistant prostate cancer

    • Selective ERα modulator, tamoxifen, is well tolerated in a heavily pretreated castration‐resistant prostate cancer (PCa) patient cohort. However, its targeted gene network and whether expression of intratumor ERα due to androgen‐deprivation therapy (ADT) may play a role in PCa progression is unknown. In this study, we examined the inhibitory effect of tamoxifen on castration‐resistant PCa in vitro and in vivo. We found that tamoxifen is a potent compound that induced a high degree of apoptosis and significantly suppressed growth of xenograft tumors in mice, at a degree comparable to ISA‐2011B, an inhibitor of PIP5K1α that acts upstream of PI3K/AKT survival signaling pathway. Moreover, depletion of tumor‐associated macrophages using clodronate in combination with tamoxifen increased inhibitory effect of tamoxifen on aggressive prostate tumors. We showed that both tamoxifen and ISA‐2011B exert their on‐target effects on prostate cancer cells by targeting cyclin D1 and PIP5K1α/AKT network and the interlinked estrogen signaling. Combination treatment using tamoxifen together with ISA‐2011B resulted in tumor regression and had superior inhibitory effect compared with that of tamoxifen or ISA‐2011B alone. We have identified sets of genes that are specifically targeted by tamoxifen, ISA‐2011B or combination of both agents by RNA‐seq. We discovered that alterations in unique gene signatures, in particular estrogen‐related marker genes are associated with poor patient disease‐free survival. We further showed that ERα interacted with PIP5K1α through formation of protein complexes in the nucleus, suggesting a functional link. Our finding is the first to suggest a new therapeutic potential to inhibit or utilize the mechanisms related to ERα, PIP5K1α/AKT network and MMP9/VEGF signaling axis, providing a strategy to treat castration‐resistant ER‐positive subtype of prostate cancer tumors with metastatic potential.
