  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Artificial intelligence-assisted point-of-care testing system for ultrafast and quantitative detection of drug-resistant bacteria

    • As one of the major causes of antimicrobial resistance, beta-lactamase develops rapidly among bacteria. Detection of beta-lactamase in an efficient and low-cost point-of-care testing (POCT) way is urgently needed. However, due to the volatile environmental factors, the quantitative measurement of current POCT is often inaccurate. Herein, we demonstrate an artificial intelligence (AI)-assisted mobile health system that consists of a paper-based beta-lactamase fluorogenic probe analytical device and a smartphone-based AI cloud. An ultrafast broad-spectrum fluorogenic probe (B1) that could respond to beta-lactamase within 20 s was first synthesized, and the detection limit was determined to be 0.13 nmol/L. Meanwhile, a three-dimensional microfluidic paper-based analytical device was fabricated for integration of B1. Also, a smartphone-based AI cloud was developed to correct errors automatically and output results intelligently. This smart system could calibrate the temperature and pH in the beta-lactamase level detection in complex samples and mice infected with various bacteria, which shows the problem-solving ability in interdisciplinary research, and demonstrates potential clinical benefits.

  • Exosome-like nanoplatform modified with targeting ligand improves anti-cancer and anti-inflammation effects of imperialine

    • Currently, most anti-cancer therapies are still haunted by serious and deleterious adverse effects. Here, we report a highly biocompatible tumor cell-targeting delivery systems utilizing exosome-like vesicles (ELVs) that delivers a low-toxicity anti-cancer agent imperialine against non-small cell lung cancer (NSCLC). First, we introduced a novel micelle-aided method to efficiently load imperialine into intact ELVs. Then, integrin alpha 3 beta 1-binding octapeptide cNGQGEQc was modified onto ELV platform for tumor targeting as integrin alpha 3 beta 1 is overexpressed on NSCLC cells. This system not only significantly improved imperialine tumor accumulation and retention, but also had extremely low systemic toxicity both in vitro and in vivo. Our discoveries offer new ways to utilize ELV more efficiently for both drug loading and targeting. The solid pharmacokinetics improvement and extraordinary safety of this system also highlight possibilities of alternative long course cancer therapies using similar strategies.

    • Developing new probes for visualizing H2S in the endoplasmic reticulum (ER) is of great significance in physiological and pathological fields in that the probe can antagonise ER stress. Herein, we try to explore efficient reporting fluorophores based on three pyrimidine derivatives (L1, L2 and L3), and eventually, a novel probe WH2S was fabricated by using emissive pyrimidine derivative (L1) as the reporting fluomphore. Upon the addition of H2S, the probe processed a thiolytic cleavage to regenerate L1, delivering a remarkable fluorescence enhancement. Probe WH2S presents a perfect selectivity, high sensitivity and low detection limit (3.81 mu M) towards H2S in the buffer media. Most importantly, fluorescence microscopy experiments demonstrate that WH2S can precisely accumulate in ER and detect exogenous/endogenous H2S at cellular level. These results imply that probe WH2S possesses great potentiality in tracking target H2S in complicated and changeable living systems.

  • Non-linear impact of natural resources, green financing, and energy transition on sustainable environment: A way out for common prosperity in NORDIC countries

    • In a couple of decades, the immense interest of scholars and policymakers is to identify the factors that support environmental sustainability without sacrificing economic well-being. Nordic countries with huge natural resource reserves are known as the top countries of the "European GDP per capita league table." However, with the lofty climate targets, these countries are still responsible for environmental impairments through their incessant contribution to the global energy supply network. Therefore, to defeat this predicament, the study investigates the role of natural resources (NTR), green finance (GFN), green energy (GEC), and economic growth (GDP) in Nordic countries from 1990 to 2018. The study employed the novel quantile-based econometrics approach of "Method of the Moments Quantile Regression" (MMQR), which provides the direction and magni-tude of the asymmetric association of NTR, GFN, GEC, and GDP with the ecological footprint. The results of this test revealed that the NTR and GDP have a significantly positive influence, while GEN and GEC have significantly negative associations with an ecological footprint across all quantiles. This has implied that green finance and green energy work as the solution, whereas natural resources and economic growth are key drivers of envi-ronmental degradation. Further, the "Augmented Mean Group" (AMG) and "Common Correlated Effect Mean Group" (CCEMG) are used as the robustness check, which endorsed the same association of NTR, GFN, GEC, and GDP with the EFP suggested by MMQR. Based on these findings, the study suggests policy implications for controlling the ecological footprint in Nordic countries.

    • It is still a challenge to obtain two-photon excited fluorescent bioimaging probes with intense emission, high photo-stability and low cytotoxicity. In the present work, four Zn(II)-coordinated complexes (1-4) constructed from two novel D-A and D-p-A ligands (L-1 and L-2) are investigated both experimentally and theoretically, aiming to explore efficient two-photon probes for bioimaging. Molecular geometry optimization used for theoretical calculations is achieved using the crystallographic data. Notably, the results indicate that complexes 1 and 2 display enhanced two-photon absorption (2PA) cross sections compared to their corresponding D-A ligand (L1). Furthermore, it was found that complex 1 has the advantages of moderate 2PA cross section in the near-infrared region, longer fluorescence lifetime, higher quantum yield, good biocompatibility and enhanced two-photon excited fluorescence. Therefore, complex 1 is evaluated as a bioimaging probe for in vitro imaging of HepG2 cells, in which it is observed under a two-photon scanning microscope that complex 1 exhibits effective co-staining with endoplasmic reticulum (ER) and nuclear membrane; as well as for in vivo imaging of zebrafish larva, in which it is observed that complex 1 exhibits specificity in the intestinal system.

    • A series of novel D-pi-A type two-photon absorption (2PA) imidazole-pyrimidine derivatives (EX-1 similar to EX-4) have been synthesized and characterized, with EX-1 was crystallography confirmed. Based on systematic photophysical investigations, the structure property relationships can be drawn as follows: (1) Both theoretical and experimental studies indicated that the different donor groups have large influences on the optical properties. (2) The 2PA cross-section values (sigma) were obtained both by Z-Scan and two photon excited fluorescence (2PEF) measurements. 2PA cross sections show an increasing trend with increasing electron-donating strength and the number of branches. (3) Comprehensively considered the optical performance, molecular volume, cytotoxicity and solubility, EX-1 and EX-2 were identified to be the best candidates for living cells (HepG2) imaging. Moreover, the 2PA excitable features of EX-1 and EX-2 are capable of imaging in fresh mouses liver tissues with a depth of ca. 70 mu m. (C) 2015 Elsevier Ltd. All rights reserved.
