  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Background The Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2017 comparative risk assessment (CRA) is a comprehensive approach to risk factor quantification that offers a useful tool for synthesising evidence on risks and risk-outcome associations. With each annual GBD study, we update the GBD CRA to incorporate improved methods, new risks and risk-outcome pairs, and new data on risk exposure levels and risk- outcome associations. Methods We used the CRA framework developed for previous iterations of GBD to estimate levels and trends in exposure, attributable deaths, and attributable disability-adjusted life-years (DALYs), by age group, sex, year, and location for 84 behavioural, environmental and occupational, and metabolic risks or groups of risks from 1990 to 2017. This study included 476 risk-outcome pairs that met the GBD study criteria for convincing or probable evidence of causation. We extracted relative risk and exposure estimates from 46 749 randomised controlled trials, cohort studies, household surveys, census data, satellite data, and other sources. We used statistical models to pool data, adjust for bias, and incorporate covariates. Using the counterfactual scenario of theoretical minimum risk exposure level (TMREL), we estimated the portion of deaths and DALYs that could be attributed to a given risk. We explored the relationship between development and risk exposure by modelling the relationship between the Socio-demographic Index (SDI) and risk-weighted exposure prevalence and estimated expected levels of exposure and risk-attributable burden by SDI. Finally, we explored temporal changes in risk-attributable DALYs by decomposing those changes into six main component drivers of change as follows: (1) population growth; (2) changes in population age structures; (3) changes in exposure to environmental and occupational risks; (4) changes in exposure to behavioural risks; (5) changes in exposure to metabolic risks; and (6) changes due to all other factors, approximated as the risk-deleted death and DALY rates, where the risk-deleted rate is the rate that would be observed had we reduced the exposure levels to the TMREL for all risk factors included in GBD 2017.

  • Delay-throughput tradeoffs for signalized networks with finite queue capacity

    • Network-level adaptive signal control is an effective way to reduce delay and increase network throughput. However, in the face of asymmetric exogenous demand, the increase of network performance via adaptive signal control alone is at the expense of service fairness (i.e., phase actuation fairness and network resource utilization fairness). In addition, for oversaturated networks, arbitrary adaptive signal control seems to have little effect on improving network performance. Therefore, under the assumption that the mean routing proportions/turn ratios of vehicles at intersections are fixed, this study investigates the problem of optimally allocating input rates to entry links and simultaneously finding a stabilizing signal control policy with phase fairness. We model the stochastic optimization problem of maximizing network throughput subject to network stability (i.e., all queue lengths have finite means) and average phase actuation constraints to bridge the gap between stochastic network stability control and convex optimization. Moreover, we further propose a micro-level joint admission and bounded signal control algorithm to achieve network stability and throughput optimization simultaneously. Joint control is implemented in a fully decomposed and distributed manner. For any arrival rate, joint control provably achieves network throughput within O(1/V) of optimality while trading off average delay with O(V), where V is an adjusted control parameter. Through a comparative simulation of a real network with 256 O-D pairs, the proposed joint control keeps network throughput at maximum, guarantees service fairness, and fully utilizes network capacity (i.e., increases network throughput by 17.54%).

  • Magneto-structural characterization of different kinds of magnetic nanoparticles

    • Using well-established measurement techniques like transmission electron microscopy (TEM), dynamic light scattering (DLS), small and wide angle X-ray scattering (SAXS, WAXS), susceptometry, and magnetorelaxometry, the distribution of the physical and magnetic size (magnetic moments) and magnetic anisotropy of a variety of structurally different magnetic nanoparticle samples (MNPs) is analyzed and compared. A term which accounts for the presence of weak magnetic areas (WMAs) within the MNPs was introduced to the widespread analysis model for M(H) data, enabling a consistent interpretation of the data in most of the systems. A comparison of the size distributions as obtained for the physical and the magnetic diameter suggests a multidomain structure for three single core systems under investigation, in all probability evoked by the presence of a wustite phase, as identified by WAXS. Analyzing the relationship d < dm < dc between the average single core diameter d, the effective magnetic (domain) size dm and the cluster diameter dc quantitatively, two qualitatively different magnetic structures in multicore MNP (MCMNP) systems were identified: (i) The magnetic moments of single cores within the MCMNP of fluidMAG tend to build flux closure structures, driven by dipole–dipole interaction. (ii) The magnetic behavior of Resovist® was attributed to the presence of domain sizes of about 12 nm within MCMNP, exceeding the single core diameters of 5 nm. Thereby, WAXS revealed a bimodal crystallite size distribution suggesting a crystallite merging process within the MCMNP. The value of the effective magnetic moment of these MCMNP could be explained within the presented “random moment cluster model” (RMCM). We conclude that the combination of physical and magnetic structure parameters obtained from complementary measurement methods allows a reliable assessment of the magnetic structure of single and multicore MNPs.

  • An Earth-sized Planet around an M5 Dwarf Star at 22 pc

    • We report on the discovery of an Earth-sized transiting planet (R p = 1.015 ± 0.051 R ⊕) in a P = 4.02 day orbit around K2-415 (EPIC 211414619), an M5V star at 22 pc. The planet candidate was first identified by analyzing the light-curve data obtained by the K2 mission, and it is here shown to exist in the most recent data from TESS. Combining the light curves with the data secured by our follow-up observations, including high-resolution imaging and near-infrared spectroscopy with IRD, we rule out false-positive scenarios, finding a low false-positive probability of 2 × 10−4. Based on IRD’s radial velocities of K2-415, which were sparsely taken over three years, we obtain a planet mass of 3.0 ± 2.7 M ⊕ (M p < 7.5 M ⊕ at 95% confidence) for K2-415b. Being one of the lowest-mass stars (≈0.16 M ⊙) known to host an Earth-sized transiting planet, K2-415 will be an interesting target for further follow-up observations, including additional radial velocity monitoring and transit spectroscopy.

    • Thermopile infrared detector is a kind of detector device mainly composed of thermocouple as the basic unit. Because of its simple principle, no need of cooling equipment, and other advantages, it has been widely used in various fields of production and life. However, the absorption rates of the materials in conventional thermopile devices are poor, and the majority of them are incompatible with microfabrication methods. In this work, a metal thermopile infrared detector with vertical graphene (VG) is designed and fabricated. The VG is grown via plasma enhanced chemical vapor deposition, and retained at the device’s thermal ends to provide the thermopile IR detector’s wideband and high response characteristics. The detector achieves a room temperature responsivity reaching a value as high as 1.53 V/W at 792 nm, which can increase the response results about 28 times and reduce the response time to 0.8 ms compared with the thermopile detector without VG. After systematically measuring the response results, it is finally found that there are three main mechanisms responsible for the response on the composite device. The first one is the response generated by the metal thermopile itself alone. The second one is the response increased eventually by the contribution of VG covered at the metal thermal junction that expands the temperature difference. The last one is the response generated by the temperature gradient existing inside the VG on the surface of the device after the absorption of heat. The portion of each partial response mechanism in the total response is also analyzed, providing a new reference direction for analyzing the response generation mechanism of thermopile detectors with other absorbing materials. The process is compatible with the microfabrication, while the device performance is enhanced and suitable for mass production. Furthermore, by utilizing the surface plasmon resonance to combine VG with metal nanoparticles, the material’ s light absorption is found to be enhanced significantly under the same conditions, and the resulting thermal voltage can be increased to 6 times. The results indicate that VG promises to possess practical applications, in many fields such as photoelectric sensing and power production devices. This technology provides a new method to manufacture high-performance thermopile infrared detectors and other sensor devices.

  • Durability of Slag-Cement Paste Containing Polyaluminum Chloride

    • The effect of polyaluminum chloride (PAC) on the durability, microstructure, and corrosion sensitivity of slag-cement pastes was investigated in this research. Various experimental tests were performed to assess the mechanical properties (compressive strength test), durability (rapid chloride migration test, water permeability test, capillary water absorption test, chloride binding test, accelerated carbonation test), microstructure (scanning electron microscopy, X-ray diffraction, mercury intrusion porosimetry), and corrosion sensitivity (accelerated corrosion sensitivity, initial chloride concentration). Experimental results showed that adding 1.5% PAC increased compressive strength by 77.0%, 53.8%, and 19.4% at 3, 7, and 28 days. Resistance to chloride migration, water permeation, capillary water absorption, and carbonation was improved. These results were consistent with the microstructural analysis, which showed that adding PAC facilitated the formation of Friedel's salt and development of pore structure, which was the key factor for the improved durability of the slag-cement paste. Adding 1.5% PAC had a limited influence on the corrosion sensitivity despite introducing the additional chloride ions into the system. These results promote the further practical application of PAC.
