  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Cancer Genome Atlas (TCGA) Research Network has profiled and analyzed large numbers of human tumors to discover molecular aberrations at the DNA, RNA, protein and epigenetic levels. The resulting rich data provide a major opportunity to develop an integrated picture of commonalities, differences and emergent themes across tumor lineages. The Pan-Cancer initiative compares the first 12 tumor types profiled by TCGA. Analysis of the molecular aberrations and their functional roles across tumor types will teach us how to extend therapies effective in one cancer type to others with a similar genomic profile. © 2013 Nature America, Inc.   • DIII-D research advancing the physics basis for optimizing the tokamak approach to fusion energy

    • DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I p steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at ∼8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I p beam injection was coupled to a radiative divertor. The hybrid core scenario was achieved directly, without the need for anomalous current diffusion, using off-axis current drive actuators. Also, a controller to assess proximity to stability limits and regulate β N in the ITER baseline scenario, based on plasma response to probing 3D fields, was demonstrated. Finally, innovative tokamak operation using a negative triangularity shape showed many attractive features for future pilot plant operation.

  • Dynamic Response of 6MW Spar Type Floating Offshore Wind Turbine by Experiment and Numerical Analyses

    • The floating offshore wind turbine (FOWT) is widely used for harvesting marine wind energy. Its dynamic responses under offshore wind and wave environment provide essential reference for the design and installation. In this study, the dynamic responses of a 6MW Spar type FOWT designed for the water depth of 100 m are investigated by means of the wave tank experiment and numerical analysis. A scaled model is manufactured for the experiment at a ratio of 65.3, while the numerical model is constructed on the open-source platform FAST (Fatigue, Aerodynamics, Structures, and Turbulence). Still water tests, wind-induced only tests, wave-induced only tests and combined wind-wave-current tests are all conducted experimentally and numerically. The accuracy of the experimental set-up as well as the loading generation has been verified. Surge, pitch and heave motions are selected to analyze and the numerical results agree well with the experimental values. Even though results obtained by using the FOWT calculation model established in FAST software show some deviations from the test results, the trends are always consistent. Both experimental and numerical studies demonstrate that they are reliable for the designed 6MW Spar type FOWT.

    • A prognostics and health management (PHM) system with prediction at its core optimizes the durability of the proton exchange membrane fuel cell (PEMFC). However, the aging behavior model has some uncertainty due to limited knowledge, affecting the predictive performance in remaining useful life (RUL) prediction. To address this issue, an RUL prediction method based on the Bayesian framework considering uncertainty quantification on the full-time scale is proposed. Firstly, the state of health (SOH) of the PEMFC is estimated, and the behavior of uncertainty is quantified. Afterwards, a long short-term memory (LSTM) neural network is employed to make a prediction for its behavior. Finally, the RUL of PEMFC is predicted based on historical SOH and the predicted behavior of uncertainty. Validation indicates that the proposed method can make a long-term prediction and provide RUL prediction with high accuracy. Under the dynamic operating condition, in terms of long-term prediction, compared to unscented Kalman filter, adaptive unscented Kalman filter, double-input-echo-state-network and bidirectional LSTM, the proposed method decreases the error by 88.12%, 41.99%, 13.82% and 3.21%, respectively. And under the dynamic operating condition, the proposed method shows good stability. Moreover, the robustness of this method has also been verified.

  • A Dynamic Transformation Car-Following Model for the Prediction of the Traffic Flow Oscillation

    • Car-following (CF) behavior is a fundamental of traffic flow modeling; it can be used for the virtual testing of connected and automated vehicles and the simulation of various types of traffic flow, such as free flow and traffic oscillation. Although existing CF models can replicate the free flow well, they are incapable of simulating complicated traffic oscillation, and it is difficult to strike a balance between accuracy and efficiency. This article investigates the error variation when the traffic oscillation is simulated by the intelligent driver model (IDM). Then, it divides the traffic oscillation into four phases (coasting, deceleration, acceleration, and stationary) by using the space headway of multiple steps. To simulate traffic oscillation between multiple human-driven vehicles, a dynamic transformation CF model is proposed, which includes the long-time prediction submodel [modified sequence-to-sequence (Seq2seq)] model, short-time prediction submodel (Transformer), and their dynamic transformation strategy]. The first submodel is utilized to simulate the coasting and stationary phases, while the second submodel is utilized to simulate the acceleration and deceleration phases. The results of experiments indicated that compared to K-nearest neighbors, IDM, and Seq2seq CF models, the dynamic transformation CF model reduces the trajectory error by 60.79–66.69% in microscopic traffic flow simulations, 7.71–29.91% in mesoscopic traffic flow simulations, and 1.59–18.26% in macroscopic traffic flow simulations. Moreover, the runtime of the dynamic transformation CF model (Inference) decreased by 14.43–66.17% when simulating the large-scale traffic flow.

  • Chip-scale solar thermal electrical power generation

    • There is an urgent need for alternative compact technologies that can derive and store energy from the sun, especially the large amount of solar heat that is not effectively used for power generation. Here, we report a combination of solution- and neat-film-based molecular solar thermal (MOST) systems, where solar energy can be stored as chemical energy and released as heat, with microfabricated thermoelectric generators to produce electricity when solar radiation is not available. The photophysical properties of two MOST couples are characterized both in liquid with a catalytical cycling setup and in a phase-interconvertible neat film. Their suitable photophysical properties let us combine them individually with a microelectromechanical ultrathin thermoelectric chip to use the stored solar energy for electrical power generation. The generator can produce, as a proof of concept, a power output of up to 0.1 nW (power output per unit volume up to 1.3 W m−3). Our results demonstrate that such a molecular thermal power generation system has a high potential to store and transfer solar power into electricity and is thus potentially independent of geographical restrictions.

    • After our recent successful demonstration of high brightness white light emitting diodes (HB-LEDs) based on high temperature grown n-ZnO nanowires on different p-type semiconductors, we present here LEDs fabricated on n-ZnO nano-wires and p-type organic semiconductors. By employing a low temperature chemical growth (≤ 90 °C) approach for ZnO synthesis combined together with organic p-type semiconductors, we demonstrate high quality LEDs fabricated on a variety of different substrates. The substrates include transparent glass, plastic, and conventional Si. Different multi-layers of p-type organic semiconductors with or without electron blocking layers have been demonstrated and characterized. The investigated p-type organic semiconductors include PEDOT:PSS, which was used as a anode in combination with other p-type polymers. Some of the heterojunction diodes also contain an electron blocking polymer sandwiched between the p-type polymer and the n-ZnO nano-wire. The insertion of electron blocking layer is necessary to engineer the device for the desired emission. Structural and electrical results will be presented. The preliminary I-V characteristics of the organic-inorganic hybrid heterojunction diodes show good rectifying properties. Finally we also present our findings on the origin of the green luminescence band which is responsible of the white light emission in ZnO is discussed.
