  • Panning for gold, but finding helium: Discovery of the ultra-stripped supernova SN 2019wxt from gravitational-wave follow-up observations

    • We present the results from multi-wavelength observations of a transient discovered during an intensive follow-up campaign of S191213g, a gravitational wave (GW) event reported by the LIGO-Virgo Collaboration as a possible binary neutron star merger in a low latency search. This search yielded SN 2019wxt, a young transient in a galaxy whose sky position (in the 80% GW contour) and distance (∼150 Mpc) were plausibly compatible with the localisation uncertainty of the GW event. Initially, the transienta's tightly constrained age, its relatively faint peak magnitude (Mi ∼ -16.7 mag), and the r-band decline rate of ∼1 mag per 5 days appeared suggestive of a compact binary merger. However, SN 2019wxt spectroscopically resembled a type Ib supernova, and analysis of the optical-near-infrared evolution rapidly led to the conclusion that while it could not be associated with S191213g, it nevertheless represented an extreme outcome of stellar evolution. By modelling the light curve, we estimated an ejecta mass of only ∼0.1 M·, with 56Ni comprising ∼20% of this. We were broadly able to reproduce its spectral evolution with a composition dominated by helium and oxygen, with trace amounts of calcium. We considered various progenitor channels that could give rise to the observed properties of SN 2019wxt and concluded that an ultra-stripped origin in a binary system is the most likely explanation. Disentangling genuine electromagnetic counterparts to GW events from transients such as SN 2019wxt soon after discovery is challenging: in a bid to characterise this level of contamination, we estimated the rate of events with a volumetric rate density comparable to that of SN 2019wxt and found that around one such event per week can occur within the typical GW localisation area of O4 alerts out to a luminosity distance of 500 Mpc, beyond which it would become fainter than the typical depth of current electromagnetic follow-up campaigns.

  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Consensus-based technical recommendations for clinical translation of renal T1 and T2 mapping MRI

    • To develop technical recommendations on the acquisition and post-processing of renal longitudinal (T1) and transverse (T2) relaxation time mapping. A multidisciplinary panel consisting of 18 experts in the field of renal T1 and T2 mapping participated in a consensus project, which was initiated by the European Cooperation in Science and Technology Action PARENCHIMA CA16103. Consensus recommendations were formulated using a two-step modified Delphi method. The first survey consisted of 56 items on T1 mapping, of which 4 reached the pre-defined consensus threshold of 75% or higher. The second survey was expanded to include both T1 and T2 mapping, and consisted of 54 items of which 32 reached consensus. Recommendations based were formulated on hardware, patient preparation, acquisition, analysis and reporting. Consensus-based technical recommendations for renal T1 and T2 mapping were formulated. However, there was considerable lack of consensus for renal T1 and particularly renal T2 mapping, to some extent surprising considering the long history of relaxometry in MRI, highlighting key knowledge gaps that require further work. This paper should be regarded as a first step in a long-term evidence-based iterative process towards ever increasing harmonization of scan protocols across sites, to ultimately facilitate clinical implementation.

  • Exploring variability in methods and data sensitivity in carbon footprints of feed ingredients

    • Purpose: Production of feed is an important contributor to life cycle greenhouse gas emissions, or carbon footprints (CFPs), of livestock products. Consequences of methodological choices and data sensitivity on CFPs of feed ingredients were explored to improve comparison and interpretation of CFP studies. Methods and data for emissions from cultivation and processing, land use (LU), and land use change (LUC) were analyzed. Method: For six ingredients (maize, wheat, palm kernel expeller, rapeseed meal, soybean meal, and beet pulp), CFPs resulting from a single change in methods and data were compared with a reference CFP, i.e., based on IPCC Tier 1 methods, and data from literature. Results and discussion: Results show that using more detailed methods to compute N2O emissions from cultivation hardly affected reference CFPs, except for methods to determine leaching (contributing to indirect N2O emissions) in which the influence is about -7 to +12 %. Overall, CFPs appeared most sensitive to changes in crop yield and applied synthetic fertilizer N. The inclusion of LULUC emissions can change CFPs considerably, i.e., up to 877 %. The level of LUC emissions per feed ingredient highly depends on the method chosen, as well as on assumptions on area of LUC, C stock levels (mainly aboveground C and soil C), and amortization period. Conclusions: We concluded that variability in methods and data can significantly affect CFPs of feed ingredients and hence CFPs of livestock products. Transparency in methods and data is therefore required. For harmonization, focus should be on methods to calculate leaching and emissions from LULUC. It is important to consider LUC in CFP studies of food, feed, and bioenergy products.

  • Hansenula polymorpha cells lacking the ER-localized peroxins Pex23 or Pex29 show defects in mitochondrial function and morphology

    • Pex23 family proteins localize to the endoplasmic reticulum and play a role in peroxisome and lipid body formation. The yeast Hansenula polymorpha contains four members: Pex23, Pex24, Pex29 and Pex32. We previously showed that loss of Pex24 or Pex32 results in severe peroxisomal defects, caused by reduced peroxisomeendoplasmic reticulum contact sites. We now analyzed the effect of the absence of all four Pex23 family proteins on other cell organelles. Vacuoles were normal in all four deletion strains. The number of lipid droplets was reduced in pex23 and pex29 , but not in pex24 and pex32 cells, indicating that peroxisome and lipid droplet formation require different Pex23 family proteins in H. polymorpha . In pex23 and pex29 cells mitochondria were fragmented and clustered accompanied by reduced levels of the fusion protein Fzo1. Deletion of DNM1 suppressed the morphological phenotype of pex23 and pex29 cells, suggesting that mitochondrial fusion is affected. pex23 and pex29 cells showed retarded growth and reduced mitochondrial activities. The growth defect was partially suppressed by DNM1 deletion as well as by an artificial mitochondrion-endoplasmic reticulum tether. Hence, the absence of Pex23 family proteins may influence mitochondrion-endoplasmic reticulum contact sites.

  • Livestock and the Environment: What Have We Learned in the Past Decade?

    • The livestock and environment nexus has been the subject of considerable research in the past decade. With a more prosperous and urbanized population projected to grow significantly in the coming decades comes a gargantuan appetite for livestock products. There is growing concern about how to accommodate this increase in demand with a low environmental footprint and without eroding the economic, social, and cultural benefits that livestock provide. Most of the effort has focused on sustainably intensifying livestock systems. Two things have characterized the research on livestock and the environment in the past decade: the development of increasingly disaggregated and sophisticated methods for assessing different types of environmental impacts (climate, water, nutrient cycles, biodiversity, land degradation, deforestation, etc.) and a focus on examining the technical potential of many options for reducing the environmental footprint of livestock systems. However, the economic or sociocultural feasibility of these options is seldom considered. Now is the time to move this agenda from knowledge to action, toward realizable goals. This will require a better understanding of incentives and constraints for farmers to adopt new practices and the design of novel policies to support transformative changes in the livestock sector. It will also require novel forms of engagement, interaction, and consensus building among stakeholders with enormously diverse objectives. Additionally, we have come to realize that managing the demand trajectories of livestock products must be part of the solution space, and this is an increasingly important research area for simultaneously achieving positive health and environmental outcomes.

    • Comprehensive but interpretable assessment of the environmental performance of diets involves choosing a set of appropriate indicators. Current knowledge and data gaps on the origin of dietary foodstuffs restrict use of indicators relying on site-specific information. This Personal View summarises commonly used indicators for assessing the environmental performance of diets, briefly outlines their benefits and drawbacks, and provides recommendations on indicator choices for actors across multiple fields involved in activities that include the environmental assessment of diets. We then provide recommendations on indicator choices for actors across multiple fields involved in activities that use environmental assessments, such as health and nutrition experts, policy makers, decision makers, and private-sector and public-sector sustainability officers. We recommend that environmental assessment of diets should include indicators for at least the five following areas: climate change, biosphere integrity, blue water consumption, novel entities, and impacts on natural resources (especially wild fish stocks), to capture important environmental trade-offs. If more indicators can be handled in the assessment, indicators to capture impacts related to land use quantity and quality and green water consumption should be used. For ambitious assessments, indicators related to biogeochemical flows, stratospheric ozone depletion, and energy use can be added.
