  • LOW GENETIC-VARIATION IN SWEDISH POPULATIONS OF THE RARE SPECIES VICIA-PISIFORMIS (FABACEAE) REVEALED WITH RFLP (RDNA) AND RAPD

    • Nine Swedish populations, 1-5 individuals/population, and one cultivated individual of the rare species Vicia pisiformis were investigated for genetic variation. In hybridizations with two rDNA probes using 8 restriction enzymes, only two individuals belonging to one population were polymorphic. A map of the rDNA gene cluster was constructed for four of the restriction enzymes used. Two of the polymorphic sites were mapped and were found to be located outside regions coding for rRNA, presumably caused by single point mutations or small deletions. The repeat length of the rDNA region was c. 10,000 bp, which corresponds well with the size found for other species belonging to Fabaceae. No length polymorphism was found in the intergenic spacer, contrary to the situation found for most other plant species investigated for rDNA variation. The haplotype diversity for the species (Hsp Shannon) was very low (0.055). Within-population values (Hpop) was 0 for all populations except the variable one, which had 0.301. PCR amplification with 6 random primers also revealed very low levels of genetic diversity. A polymorphism was observed in a limited number of individuals for four populations. Hsp was 0.065 and HpopBAR was 0.050. The average D value (Wetton) for the PCR haplotypes was 0.99.

  • Glucocorticoid receptor messenger ribonucleic acid in different regions of human adipose tissue

    • The expression of glucocorticoid receptor (GR) messenger RNA (mRNA) was investigated in sc adipose tissue and isolated adipocytes from the abdominal and gluteal regions in men and women using a human GR complementary RNA probe. GR mRNA levels were 2-fold higher in female than in male abdominal tissue or adipocytes, whereas in gluteal tissue or adipocytes no sex differences were observed. GR mRNA levels in female abdominal adipocytes were 50% higher than in corresponding female gluteal adipocytes; the opposite was observed corresponding in males. Northern blot analysis of total cellular RNA isolated from abdominal and gluteal adipocytes showed hybridization of the human GR probe to an RNA species of approximately 7.1 kilobases in both regions. No sex or regional differences in GR mRNA stability were observed. The human metallothionein II (hMTII) mRNA, which is regulated by glucocorticoids at the transcriptional level, showed an opposite sex and regional pattern as GR mRNA. However, in gluteal adipose tissue no sex differences were observed in hMTII mRNA levels. The expression of beta-actin mRNA, which is not regulated by glucocorticoids, showed no sex or regional variation. By immunocytochemistry, using an anti-GR-monoclonal antibody, cytoplasmic as well as nuclear staining for GR was demonstrated in both sexes and both regions. In conclusion, variations in GR mRNA levels between sexes and body regions may explain the well known sex and tissue differences in effects of glucocorticoids on human adipose tissue.

  • Deuteron breakup by 1.15 GeV protons and excitation of the Δ isobar

    • The pd→npp reaction has been measured using circulating 1.15 GeV protons, a gas jet target and scintillation detectors selecting scattered protons with energy above 30 MeV. The distribution of the two-proton invariant mass has a maximum at 2.13 ± 0.01 GeV/c2 and a width of 130 ± 10 MeV/c2 (FWHM). We see no evidence of narrow (Γ < 30 MeV/c2) peaks in the phase-space region selected. The mass of X, excited in the pN→nX elementary process, is 40 MeV/c2 smaller than the mass of a real Δ isobar.

    • The pd → npp charge exchange reaction has been studied at 1.15 GeV incident proton energy in a selected kinematical region. The distributions of two-proton invariant mass Mpp, of angle of a single proton and the differential cross section dσ/dt of the (p, n) reaction are presented and compared with model predictions corrected for the acceptance of the experimental apparatus. We see no evidence of narrow (Γ < 30 MeV/c2) peaks in the Mpp interval 2.1-2.3 GeV/c2. The production of an intermediate Δ isobar in the primary (p, n) charge exchange and its subsequent absorption in a final state ΔN → pp process together with the background from quasi-free charge exchange scattering reasonably reproduces the general trends of the data.

    • Barbiturates are used clinically as anaesthetics and to reduce raised intracranial pressure. One side effect is hypotension, usually ascribed to a depression of cardiac contractility, while their effects on the resistance vessels are more controversial: both vasodilation and vasoconstriction have been described. This study analyzes the effects of thiopental on basal vascular tone in the cat skeletal muscle. We found that total resistance increased by almost 20% at low (50 mumol/l) and decreased down to about 50% of control at high (350 mumol/l) plasma concentrations of thiopental. The vasoconstriction dominated in the large arterioles (i.d. greater than 25 microns) and the vasodilation in the small arterioles (i.d. less than 25 microns). A dose-dependent inhibition of myogenic vascular reactivity (here defined as the maximum resistance increase to a transient rise in transmural pressure) coincided with the vasodilation. Autoregulation of blood flow was depressed by thiopental. During vasoconstriction there was a net transcapillary fluid absorption and during vasodilation a net fluid filtration. The fluid movements could be ascribed to variations in capillary hydrostatic pressure. If applicable to the cerebral circulation these results suggest that thiopental at high plasma concentrations might induce, instead of reduce, interstitial brain oedema.
