  • Reproducibility in DNA flow cytometric analysis of breast cancer: comparison of 12 laboratories' results for 67 sample homogenates

    • Flow cytometric (FCM) DNA analysis yields information on ploidy status and the S-phase fraction (SPF), variables of prognostic importance in breast cancer. The clinical value of the SPF is currently being evaluated in prospective randomized trials. The widespread use of FCM DNA analysis emphasizes the importance of reproducibility (both intra- and interlaboratory). In this study, 67 nuclear suspensions of breast cancer samples were analyzed by 12 laboratories routinely performing FCM DNA analysis in breast cancer. No general guidelines were imposed; each laboratory used its own standard protocols. For DNA ploidy status (diploid vs. non-diploid), agreement was complete for 79% (53/67) of the samples, compared with 64% (43/67) of samples when tetraploidy was considered [i.e., euploid (diploid+tetraploid) vs. aneuploid (the remaining non-diploid)]. For the SPF, pairwise comparison of the results of all 12 laboratories yielded a mean Spearman's rank correlation of 0.78 (range: 0.54-0.93). For those 39 samples being categorized in low or high SPF by all laboratories, all agreed in 14 samples (36%). Similar patterns were obtained with kappa measures, agreement being good for ploidy status (diploid vs. non-diploid; overall kappa = 0.87 and 0.74 for euploid vs. aneuploid), but moderate for the SPF [overall kappa = 0.47 (for low SPF vs. high SPF vs. "no SPF reported")]. Discrepancies were chiefly attributable to differences in the categorization of the S-phase values, rather than in FCM procedures, other critical differences being in the detection and interpretation of near-diploid and small non-diploid cell populations, the definition of tetraploidy, and the choice and execution of the method used for S-phase estimation. Based on the observations of this study, detailed guidelines for FCM analysis and interpretation of data are proposed in the Appendix. Some issues remain, however, e.g., to standardize a method for S-phase calculation and tetraploid definition.

    • In this study a mathematical model, based on Navier Stokes equations, was developed and validated against experimental data. This model predicts the pressure changes in the spinal canal as a function of the volume change inside the canal during neck bending in the x-z (sagittal) plane. Another aim of the study was to investigate pressure phenomena and ganglion injuries at static neck extension loading and dynamic neck extension trauma with a head-restraint present. Experiments on pigs were conducted. Preliminary results indicate that ganglion injuries, as well as pressure transients inside the spinal canal, seem to correlate to the phase shift when the neck passes an s-shape (or maximal retraction) during the rearward motion of the head. That is, when the upper neck quickly changes from a flexion to an extension shape. Static loading of the neck resulted in no signs of injuries to the ganglia. A possible candidate for a neck injury criterion is presented, based on the relative acceleration between the top and the bottom of the cervical spine. A tolerance level based on the pig tests is also discussed.

    • p-phenylenediamine derivatives are widely used in industry and in cosmetics, and several of them are well-known sensitizers. One group of allergenic p-phenylenediamine derivatives are used as colour film developers. Cross-reactivity between the colour film developers has been reported. In this paper, an occupational facial dermatitis due to colour film developers is described. The patient reacted to colour film developers (CD-1, CD-2, CD-3, and CD-4), but not to other p-phenylenediamine derivatives tested. He also showed allergic reactions to Disperse Blue 106 and Disperse Blue 124 and to Disperse Red 17, but not to Disperse Orange 3. The activation of the colour film developers by oxidation at physiological pH was analysed with chemical methods, and the mechanism responsible for the concomitant reactivities to the colour film developers and the disperse dyes at a molecular level is discussed.

  • Extraction of mercaptobenzothiazole compounds from rubber products

    • At evaluation of contact dermatitis caused by solid material, patch testing is usually performed with the material as such and with extracts of it. In this study, optimization of the extraction technique monitored by quantitative HPLC analysis of the extracted haptens is described for mercaptobenzothiazole derivatives. Several solvents with different properties are included. Acetone has traditionally been a solvent widely used for the extraction of organic haptens from solid products. However, acetone and other ketones are not inert solvents. The rubber accelerators 2-(4-morpholinyl mercapto) benzothiazole (MMBT) and N-cyclohexyl-2-benzothiazyl sulfenamide (CBS) react with acetone, yielding 2 new compounds, which were isolated and characterised by NMR and MS. For the extraction of solid rubber products, methyl tert-butyl ether (MTBE) was found to be a more suitable solvent which is unreactive to most common haptens.

Mehrere dieser Symptomekönnen möglicherweise durch Verletzungenim Bereich der Nervenwurzeln der HWS erklärt werden. Die Hypothese wurde aufgestellt,daû während der Extensions- und Flexionsbewegungendes Halses, aufgrund vonhämodynamischen Auswirkungen, ¾nderungendes inneren Volumens des Halswirbelkanalskurzzeitige Druckveränderungenim ZNS verursachen und daû dadurch Gewebeschäden durch die mechanische Belastungder Nervenwurzel entstehen. Um dieHypothese zu überprüfen, wurden anästhesierteSchweine einem experimentellenSchleudertrauma mit Extensions-, FlexionsundSeitneigungsbewegungen ausgesetzt.Die traumatische Belastung wurde unterhalbeiner Stufe gehalten, bei der eine Halswirbelfrakturstattfinden könnte. Während derBelastung wurde der Pulsdruck im Halswirbelkanalgemessen. In diesem Zusammenhangergaben sich Hinweise einer Dysfunktionder Zellmembran bei Nervenzellkörpernder Spinalganglien. Die ganglionären Verletzungenkönnen möglicherweise einige dermit Weichteilverletzungen des Halses nachAutounfällen einhergehenden Symptomeerklären. Um die Situation bei einem Auffahrunfallzu simulieren, wurde der Kopf desSchweines rückwärts vom Torso weggezogen.Dabei wurde festgestellt, daû die ganglionären Verletzungen zu einem sehr frühenZeitpunkt während der Halsbewegungenstattfinden und zwar in der Phase, in der dieBewegung von der Retraktion zur Extensionwechselt. Bei einer ähnlichen, jedoch statischenBelastung des Halses wurden keineganglionären Verletzungen bei den Schweinenfestgestellt. Dies ist ein Hinweis darauf,daû diese Verletzungen durch dynamischeFaktoren verursacht werden und bietet somitweitere Unterstützung für die Druckhypothese.Auf der Basis eines theoretischen Modellswurden Kriterien für Halsverletzungen(neck injury criterion = NIC) entwickelt. Esweist darauf hin, daû das Risiko von ganglionären Verletzungen am Punkt der maximalenHalsretraktion durch die unterschiedlichehorizontale Beschleunigung und Geschwindigkeitzwischen Kopf und oberemTorso bestimmt wird.
