  • Consequences of eliminating adenosine A(1) receptors in mice

    • The second coding exon of the adenosine A, receptor gene was eliminated by homologous recombination. The phenotype of mice (mixed C57B6/129OlaHsd background) was studied, using siblings from matings of heterozygous mice. Among the offspring the ratio between+/+, +/-and -/-animals was 1:2:1. Over the first half-year-at least-growth and viability were the same in all genotypes. Binding of A(1) ligands was eliminated in-/-mice and halved in+/-mice. Blood pressure was increased in-/-mice and this was paralleled by an increase in plasma renin. Heart rate was unaffected, as was contractility. Furthermore, the response of the perfused heart to ischemia was similar in+/+and -/-hearts. However, remote preconditioning was eliminated in-/-mouse hearts. Tubuloglomerular feedback in the kidney was also lost in-/-mice. The analgesic response to a non-selective adenosing receptor agonist was lost in-/-mice, which also showed hyperalgesia in the tail-flick test. There was a slight hypoactivity in-/-mice, but responses to caffeine were essentially normal. The inhibition of excitatory neurotransmission in hippocampus by adenosine was lost in-/-mice and reduced in+/-mice. Responses to ATP were affected similarly. Hypoxic depression of synaptic transmission was essentially eliminated in hippocampus and hypoxic decrease in spinal respiratory neuron firing was markedly reduced. These results show that adenosine A, receptors play a physiologically important role in the kidney, spinal cord, and hippocampus and that they are critically important in the adaptive responses to hypoxia.     • The European Prospective Investigation into Cancer and Nutrition (EPIC), which covers a large cohort of half a million men and women from 23 European centres in 10 Western European countries, was designed to study the relationship between diet and the risk of chronic diseases, particularly cancer. Information on usual individual dietary intake was assessed using different validated dietary assessment methods across participating countries. In order to adjust for possible systematic over- or underestimation in dietary intake measurements and correct for attenuation bias in relative risk estimates, a calibration approach was developed. This approach involved an additional dietary assessment common across study populations to re-express individual dietary intakes according to the same reference scale. A single 24-hour diet recall was therefore collected, as the EPIC reference calibration method, from a stratified random sample of 36 900 subjects from the entire EPIC cohort, using a software program (EPIC-SOFT) specifically designed to standardise the dietary measurements across study populations. This paper describes the design and populations of the calibration sub-studies set up in the EPIC centres. In addition, to assess whether the calibration sub-samples were representative of the entire group of EPIC cohorts, a series of subjects' characteristics known possibly to influence dietary intakes was compared in both population groups. This was the first time that calibration sub-studies had been set up in a large multi-centre European study. These studies showed that, despite certain inherent methodological and logistic constraints, a study design such as this one works relatively well in practice. The average response in the calibration study was 78.3% and ranged from 46.5% to 92.5%. The calibration population differed slightly from the overall cohort but the differences were small for most characteristics and centres. The overall results suggest that, after adjustment for age, dietary intakes estimated from calibration samples can reasonably be interpreted as representative of the main cohorts in most of the EPIC centres.

  • Nucleon-induced reactions at intermediate energies : New data at 96 MeV and theoretical status

    • Double-differential cross sections for light charged particle production (up to A=4) were measured in 96 MeV neutron-induced reactions, at the TSL Laboratory Cyclotron in Uppsala (Sweden). Measurements for three targets, Fe, Pb, and U, were performed using two independent devices, SCANDAL and MEDLEY. The data were recorded with low-energy thresholds and for a wide angular range (20°–160°). The normalization procedure used to extract the cross sections is based on the np elastic scattering reaction that we measured and for which we present experimental results. A good control of the systematic uncertainties affecting the results is achieved. Calculations using the exciton model are reported. Two different theoretical approaches proposed to improve its predictive power regarding the complex particle emission are tested. The capabilities of each approach is illustrated by comparison with the 96 MeV data that we measured, and with other experimental results available in the literature.

  • The solution thermolysis approach to molybdenum(V) alkoxides : synthesis, solid state and solution structures of the bimetallic alkoxides of molybdenum(V) and niobium(V), tantalum(V) and tungsten(VI)

    • No complex formation can be observed between molybdenum(VI) oxoalkoxides and the alkoxides of niobium(V) or tantalum(V) at room temperature. The bimetallic derivatives of molybdenum(V), Mo4M2O8((OPr)-Pr-i)(14), where M=Nb 1 and Ta 2, were instead isolated on cooling from the solutions of the isopropoxides in toluene subjected to a short-time reflux. The X-ray single crystal study showed both 1 and 2 to be built of ((PrO)-Pr-i)(3)M(mu-(OPr)-Pr-i)(3)MoO(mu-O)(2)MoO(mu-(OPr)-Pr-i)(2)MoO(mu-O)(2)MoO(mu-(OPr)-Pr-i)(3)M((OPr)-Pr-i)(3) non-linear chain molecules with 2 Mo-Mo bonds (2.5836(8) Angstrom) and short but non-bonding Mo-M distances (3.1791(8) Angstrom for 1 and 3.1746(8) Angstrom for 2). According to NMR and EXAFS data this structure becomes very fluxional or might even be partially broken into homometallic components in hydrocarbon solutions. The oxidation of 2 with traces of oxygen leads to the formation of Mo3Ta2O8((OPr)-Pr-i)(10) 3. Compound 3 can be isolated in a pure form from the reaction of MoO((OPr)-Pr-i)(4) with Ta((OPr)-Pr-i)(4)(OMe) 6: the presence of methoxide ligands leads to the formation of additional oxoligands via non-reductive thermolysis leading to the formation of a (CH3)(2)C(OMe)(2) ketal as organic byproduct. The molecules of 3 are 5-member rings with a MoO(mu-O)(2)MoO fragment in the basis (Mo-Mo 2.5730(13) Angstrom), coupled to two (mu-(OPr)-Pr-i)(2)Ta((OPr)-Pr-i)(3) fragments that are joined together by an oxomolybdate ligand (mu-O)(2)MoO2. According to NMR-spectroscopic data the aggregate is preserved and rigid in solution. Mo4Ta4O16((OPr)-Pr-i)(12) 4 was found to be one of the products of complete oxidation of 2 (and 3) on prolonged contact with dry oxygen. The thermal treatment of the solutions of MoO((OPr)-Pr-i)(4) and WO((OPr)-Pr-i)(4) in toluene yields (Mo4O8)-O-V(Mo,W)O-VI(2)2((OPr)-Pr-i)(12) 5 with a molecular structure very close to its homometallic analog Mo6O10((OPr)-Pr-i)(12). The complete X-ray single crystal study was carried out for the sample of 5 with (Mo4O8)-O-V(Mo0.45W0.55)O-VI(2)2((OPr)-Pr-i)(12) composition.

    • Exposure to quartz induces pulmonary inflammation and development of fibrosis. In order to study the fibrosing process, we investigated morphology function and phenotype of alveolar (AMs) and interstitial (IMs) macrophages at an early stage of fibrosis in rats. Rats were exposed by intratracheal instillations of 10 mg quartz (n = 8) or saline (n = 8) and studied 3 months later. AMs were obtained by bronchoalveolar lavage and IMs by mechanical fragmentation, followed by enzymatic digestion of lung tissue. Histology revealed subacute silicosis, with early focal fibrosis and alveolar lipoproteinosis. AM quartz exposure increased phagocytic activity and expression of major histocompatibility complex (MHC) Ia antigens, the latter being associated with cellular antigen presenting capacity. IM had an even more pronounced expression of MHC than AM after quartz exposure. Both macrophage fractions had a higher expression of OX-42 (complement receptor 3, CR3) than controls, but the increase in the IM fraction might be explained by the remaining AM in the IM fraction. Exposed AM adhered less to extracellular matrix components (vitronectin and fibronectin) than controls. In contrast, the adhesion of IM to vitronectin increased after exposure. Besides increased adhesion, the effects on IM were scarce. Our results therefore do not support the hypothesis that IM has a key role in the process of inflammation, including fibrosis.

The cohort has been positioned as a national and international resource for scientific research.
