  • Clinical Manifestations, Immunological Characteristics and Genetic Analysis of Patients with Hyper-Immunoglobulin M Syndrome in Iran

    • <b><i>Background:</i></b> Hyper-immunoglobulin M (HIGM) syndrome is a rare heterogeneous group of primary immunodeficiency disorders characterized by low or absent serum levels of IgG and IgA along with normal or elevated serum levels of IgM. <b><i>Methods:</i></b> Clinical and immunological data were collected from the 75 patients’ medical records diagnosed in Children’s Medical Center affiliated to Tehran University Medical Sciences and other Universities of Medical Sciences in Iran. Among 75 selected patients, 48 patients (64%) were analyzed genetically using targeted and whole-exome sequencing. <b><i>Results:</i></b> The ratio of male to female was 2.9:1. The median age at the onset of the disease, time of diagnosis, and diagnostic delay were 10.5, 50, and 24 months, respectively. Pneumonia and lower respiratory tract infections (61.3%) were the most common complications. Responsible genes were identified in 35 patients (72.9%) out 48 ge<i>netically</i> analyzed patients. <i>Cluster of differentiation 40 ligand</i> gene was the most mutated gene observed in 24 patients (68.5%) followed by <i>activation-induced cytidine deaminase</i> gene in 7 patients, <i>lipopolysaccharide-responsive and beige-like anchor</i> (1 patient), <i>nuclear factor-kappa-B essential modulator</i> (1 patient), <i>phosphoinositide-3-kinase regulatory subunit 1</i> (1 patient), and <i>nuclear factor kappa B subunit 1</i> (1 patient) genes. Nineteen (25.3%) patients died during the study period, and pneumonia was the major cause of death occurred in 6 (31.6%) patients. <b><i>Conclusion:</i></b> Physicians in our country should carefully pay attention to respiratory tract infections and pneumonia, particularly in patients with a positive family history. Further investigations are required for detection of new genes and pathways resulting in HIGM phenotype.

    • This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come.

  • Conversion of Glycerol to 3-Hydroxypropanoic Acid by Genetically Engineered Bacillus subtilis

    • 3-Hydroxypropanoic acid (3-HP) is an important biomass-derivable platform chemical that can be converted into a number of industrially relevant compounds. There have been several attempts to produce 3-HP from renewable sources in cell factories, focusing mainly on Escherichia coli, Klebsiella pneumoniae, and Saccharomyces cerevisiae. Despite the significant progress made in this field, commercially exploitable large-scale production of 3-HP in microbial strains has still not been achieved. In this study, we investigated the potential of Bacillus subtilis as a microbial platform for bioconversion of glycerol into 3-HP. Our recombinant B. subtilis strains overexpress the two-step heterologous pathway containing glycerol dehydratase and aldehyde dehydrogenase from K. pneumoniae. Genetic engineering, driven by in silico optimization, and optimization of cultivation conditions resulted in a 3-HP titer of 10 g/L, in a standard batch cultivation. Our findings provide the first report of successful introduction of the biosynthetic pathway for conversion of glycerol into 3-HP in B. subtilis. With this relatively high titer in batch, and the robustness of B. subtilis in high density fermentation conditions, we expect that our production strains may constitute a solid basis for commercial production of 3-HP.

  • Directional Modulation Via Symbol-Level Precoding : A Way to Enhance Security

    • Wireless communication provides a wide coverage at the cost of exposing information to unintended users. As an information-theoretic paradigm, secrecy rate derives bounds for secure transmission when the channel to the eavesdropper is known. However, such bounds are shown to be restrictive in practice and may require exploitation of specialized coding schemes. In this paper, we employ the concept of directional modulation and follow a signal processing approach to enhance the security of multiuser multi-input multioutput (MIMO) communication systems when a multiantenna eavesdropper is present. Security enhancement is accomplished by increasing the symbol error rate at the eavesdropper. Unlike the information-theoretic secrecy rate paradigm, we assume that the legitimate transmitter is not aware of its channel to the eavesdropper, which is a more realistic assumption. We examine the applicability of MIMO receiving algorithms at the eavesdropper. Using the channel knowledge and the intended symbols for the users, we design security enhancing symbol-level precoders for different transmitter and eavesdropper antenna configurations. We transform each design problem to a linearly constrained quadratic program and propose two solutions, namely the iterative algorithm and one based on nonnegative least squares, at each scenario for a computationally efficient modulation. Simulation results verify the analysis and show that the designed precoders outperform the benchmark scheme in terms of both power efficiency and security enhancement.

  • Secrecy Analysis on Network Coding in Bidirectional Multibeam Satellite Communications

    • Network coding is an efficient means to improve the spectrum efficiency of satellite communications. However, its resilience to eavesdropping attacks is not well understood. This paper studies the confidentiality issue in a bidirectional satellite network consisting of two mobile users who want to exchange message via a multibeam satellite using the XOR network coding protocol. We aim to maximize the sum secrecy rate by designing the optimal beamforming vector along with optimizing the return and forward link time allocation. The problem is nonconvex, and we find its optimal solution using semidefinite programming together with a 1-D search. For comparison, we also solve the sum secrecy rate maximization problem for a conventional reference scheme without using network coding. Simulation results using realistic system parameters demonstrate that the bidirectional scheme using network coding provides considerably higher secrecy rate compared with that of the conventional scheme.
