- Colonization of the human stomach by Helicobacter pylori is associated with the development of gastritis, duodenal ulcer, mucosa-associated lymphoid tissue (MALT) lymphoma, and gastric cancer. H. pylori-antigen-binding single-chain variable fragments (ScFv) were derived from murine hybridomas producing monoclonal antibodies and expressed as a g3p-fusion protein on a filamentous M13 phage. The recombinant ScFv-phage reacted specifically with a 30-kDa monomeric protein of a H. pylori surface antigen preparation and by means of immunofluorescence microscopy the phage was shown to bind to both the spiral and coccoid forms of the bacterium. In vitro, the recombinant phage exhibited a bacteriocidal effect and inhibited specifically the growth of all the six strains of H. pylori tested. When H. pylori was pretreated with the phage 10 min before oral inoculation of mice, the colonization of the mouse stomachs by the bacterium was significantly reduced (P<0.01). The results suggest that genetic engineering may be used to generate therapy-effective phages.

- Using the ion-selective electrode method with a concentrated electrolyte solution added continuously, the mean activity coefficients of HCl in the HCl + NH4Cl + H2O system were experimentally measured at 298.15 and 313.15 K and at five molality fractions of NH4Cl (y2 = mNH4Cl/(mHCl + mNH4Cl) from 0.1 to 0.9. The measurements were made by an electrochemical cell using a H glass ion-selective electrode and a chloride solid-state ion-selective electrode. It was found that the influence of NH4+ on the H glass ion-selective electrode could be neglected up to 1.3 molkg-1, and this pair of ion-selective electrodes was suitable for determining the activity coefficients of HCl in the system. A new set of Pitzer mixing parameters, correlated from the experimental results, was used to calculate the activity coefficients for HCl in the system from 293.15 to 313.15 K up to 3.0 molkg-1.

- Using ion-selective electrode(s) (ISE) the activity coefficients of NaCl in the system NaCl-NH4Cl-H2O at 10, 25, and 40°C were measured by a computer-controlled automatic titration system. The ionic strength fractions of NH4Cl were 0.2, 0.4, 0.6, and 0.8, respectively. It was found that the influence of the NH4+ cation on the Na glass ISE was small enough to be neglected up to 3.0 mol-kg-1. The Pitzer equation was adopted to calculate the activity coefficients of NaCl in this system and compared with the experimental data. Comparison of results indicates that the Pitzer parameters correlated from solubility data are suitable for calculating the activity coefficients for this system within the saturated solutions.

- Insulin-like growth factor (IGF) I is an important regulator of both skeletal growth and adult bone metabolism. To better understand the relative importance of systemic IGF-I versus locally expressed IGF-I we have developed a transgenic mouse model with inducible specific IGF-I gene inactivation in the liver (LI-IGF-I-/-). These mice are growing normally up to 12 weeks of age but have a disturbed carbohydrate and lipid metabolism. In this study, the long-term effects of liver-specific IGF-I inactivation on skeletal growth and adult bone metabolism were investigated. The adult (week 8-55) axial skeletal growth was decreased by 24% in the LI-IGF-I-/- mice whereas no major reduction of the adult appendicular skeletal growth was seen. The cortical cross-sectional bone area, as measured in the middiaphyseal region of the long bones, was decreased in old LI-IGF-I-/- mice. This reduction in the amount of cortical bone was caused mainly by decreased periosteal circumference and was associated with a weaker bone determined by a decrease in ultimate load. In contrast, the amount of trabecular bone was not decreased in the LI-IGF-I-/- mice. DNA microarray analysis of 30-week-old LI-IGF-I-/- and control mice indicated that only four genes were regulated in bone whereas approximately 40 genes were regulated in the liver, supporting the hypothesis that liver-derived IGF-I is of minor importance for adult bone metabolism. In summary, liver-derived IGF-I exerts a small but significant effect on cortical periosteal bone growth and on adult axial skeletal growth while it is not required for the maintenance of the trabecular bone in adult mice.

- This aim of this investigation is to study the effect of the presence of Na2SO4 deposits and water vapor on the oxidation of an air plasma-sprayed TBC composed of a partially stabilized ZrO2 top coat with an underlying NiCoCrAlY bond coat. XRD and SEM/EDX were used to analyze the changes in the coatings after oxidation at 1000degreesC for 72 hours in dry or humidified (containing 50% H2O) O-2 atmosphere, with or without Na2SO4 deposited on the top coat. When oxidized in oxygen, bond coat oxide regions, consisting of almost pure aluminum oxide, were formed at the top coat / bond coat and bond coat / substrate interfaces and in the bond coat around the splat lines. When water vapor was present, the top coat / bond coat interface oxide was marginally thicker and included small regions with more chromium, cobalt and nickel. The addition of the salt deposits resulted in some destabilization in the outermost regions of the top coat. The salt deposits also caused the formation of thicker oxide comprising two regions, though this formation was observed only at the top coat / bond coat interface. The first type was a thin inner (i.e. bordering the bond coat) oxide that was mostly aluminum oxide. The other was a much thicker oxide containing higher levels of chromium, cobalt and nickel along with aluminum. In this oxide region, the aluminum level was higher when the chromium level was lower and vice versa.

- IGF-I has been suggested to be of importance for cardiovascular structure and function, but the relative role of locally produced and liver-derived endocrine IGF-I remains unclear. Using the Cre-LoxP recombination system, we have previously created transgenic mice with a liver-specific, inducible IGF-I knockout (LI-IGF-I-/-). To examine the role of liver-derived IGF-I in cardiovascular physiology, liver-derived IGF-I was inactivated at 4 wk of age, resulting in a 79% reduction of serum IGF-I levels. At 4 months of age, systolic blood pressure (BP) was increased in LI-IGF-I-/- mice. Echocardiography showed increased posterior wall thickness in combination with decreased stroke volume and cardiac output, whereas other systolic variables were unchanged, suggesting that these cardiac effects were secondary to increased peripheral resistance. Acute nitric oxide-synthase inhibition increased systolic BP more in LI-IGF-I-/- mice than in control mice. LI-IGF-I-/- mice showed impaired acetylcholine-induced vasorelaxation in mesenteric resistance vessels and increased levels of endothelin-1 mRNA in aorta. Thus, the increased peripheral resistance in LI-IGF-I-/- mice might be attributable to endothelial dysfunction associated with increased expression of endothelin-1 and impaired vasorelaxation of resistance vessels. In conclusion, our findings suggest that liver-derived IGF-I is involved in the regulation of BP in mice.
