  • Toward an operational anthropogenic CO2 emissions monitoring and verification support capacity

    • Under the Paris Agreement (PA), progress of emission reduction efforts is tracked on the basis of regular updates to national greenhouse gas (GHG) inventories, referred to as bottom-up estimates. However, only top-down atmospheric measurements can provide observation-based evidence of emission trends. Today, there is no internationally agreed, operational capacity to monitor anthropogenic GHG emission trends using atmospheric measurements to complement national bottom-up inventories. The European Commission (EC), the European Space Agency, the European Centre for Medium-Range Weather Forecasts, the European Organisation for the Exploitation of Meteorological Satellites, and international experts are joining forces to develop such an operational capacity for monitoring anthropogenic CO2 emissions as a new CO2 service under the EC's Copernicus program. Design studies have been used to translate identified needs into defined requirements and functionalities of this anthropogenic CO2 emissions Monitoring and Verification Support (CO2MVS) capacity. It adopts a holistic view and includes components such as atmospheric spaceborne and in situ measurements, bottom-up CO2 emission maps, improved modeling of the carbon cycle, an operational data-assimilation system integrating top-down and bottom-up information, and a policy-relevant decision support tool. The CO2MVS capacity with operational capabilities by 2026 is expected to visualize regular updates of global CO2 emissions, likely at 0.05° x 0.05°. This will complement the PA's enhanced transparency framework, providing actionable information on anthropogenic CO2 emissions that are the main driver of climate change. This information will be available to all stakeholders, including governments and citizens, allowing them to reflect on trends and effectiveness of reduction measures. The new EC gave the green light to pass the CO2MVS from exploratory to implementing phase.

  • KIF1A variants are a frequent cause of autosomal dominant hereditary spastic paraplegia

    • Variants in the KIF1A gene can cause autosomal recessive spastic paraplegia 30, autosomal recessive hereditary sensory neuropathy, or autosomal (de novo) dominant mental retardation type 9. More recently, variants in KIF1A have also been described in a few cases with autosomal dominant spastic paraplegia. Here, we describe 20 KIF1A variants in 24 patients from a clinical exome sequencing cohort of 347 individuals with a mostly ‘pure’ spastic paraplegia. In these patients, spastic paraplegia was slowly progressive and mostly pure, but with a highly variable disease onset (0–57 years). Segregation analyses showed a de novo occurrence in seven cases, and a dominant inheritance pattern in 11 families. The motor domain of KIF1A is a hotspot for disease causing variants in autosomal dominant spastic paraplegia, similar to mental retardation type 9 and recessive spastic paraplegia type 30. However, unlike these allelic disorders, dominant spastic paraplegia was also caused by loss-of-function variants outside this domain in six families. Finally, three missense variants were outside the motor domain and need further characterization. In conclusion, KIF1A variants are a frequent cause of autosomal dominant spastic paraplegia in our cohort (6–7%). The identification of KIF1A loss-of-function variants suggests haploinsufficiency as a possible mechanism in autosomal dominant spastic paraplegia. © 2019, The Author(s).

  • Comparison of 6-mm and 11-mm dental implants in the posterior region supporting fixed dental prostheses: 5-year results of an open multicenter randomized controlled trial

    • Objective The aim of this multicenter, randomized controlled trial was to compare the clinical and radiographic outcomes of 6-mm or 11-mm implants, placed in the posterior maxilla and mandible, during a 5-year follow-up period. Materials and methods Ninety-five patients with adequate bone height for 11-mm implants, were randomly allocated to a 6-mm group (test group with short implants) or an 11-mm group (control group with standard-length implants). Two or three implants of the same length were placed in each patient and after 6 weeks loaded with a splinted provisional restoration. This was followed by definitive splinted restoration 6 months after implant placement. Clinical and radiographic parameters, including the occurrence of complications were recorded. Results A total of 49 patients were enrolled to receive 6-mm implants (n = 108) and 46 patients to receive 11-mm implants (n = 101). Three implants (two of 6 mm and one of 11 mm in length) were lost before loading and one 6-mm implant after 15 months of function, and one 11-mm implant was lost during the first year of function. The 5-year survival rates were 96.0% and 98.9% in the 6-mm and 11-mm group, respectively. The mean marginal bone level changes 5 years post-loading were 0.01 +/- 0.45 mm (bone gain) in the 6-mm group and -0.12 +/- 0.93 mm (bone loss) in the 11-mm group (p = .7670). Clinical parameters, including plaque, bleeding on probing and pocket probing depth were not significantly different between the groups, and also technical complications were low. Conclusion The clinical and radiographic outcomes of 6-mm short and 11-mm standard-length implants were not different during a 5-year evaluation period.

    • A challenge describes both opportunities and barriers, and an enabler provides means to manage achallenge.The framework for the deliverable is based upon identifying process facilitators (e.g.workshops) and discursive abilities/devices providing enablers that partners used in order to make senseacross organizations and professional communities (developers and users) as a means to develop anddeploy digital technologies and services.IVL lead the work and edited the deliverable. Gothenburg city (CGEA), city of Amersfoort (COA) andBarcelona waste and wastewater company (BCASA) provided experiences as stakeholder and dataprovider, through user involvement.IVL Swedish Environmental Institute, Eurecat (EUT), Talkpool (TP),Civity (CIV) provided experiences from development and implementation work (focusing on stakeholderengagement and user involvement). They all contributed to the deliverable through meetings and writingone section each. Analysing the same processes from different points of view provided interesting insightsinto the development process and implications for the coming phases of the project.The Gothenburg section expresses how insights from cooperation issues from before the project, wereturned into a well-designed collaboration process (process facilitator) between developers andusers/stakeholders. In Gothenburg, engaging with various departments within the municipality and withexternal stakeholders has been a continuing learning process for IVL (case study leader).IVL graduallypresented various opportunities within the project, both in Amersfoort and Barcelona, which has beenvery much appreciated by the stakeholders. In this way, the engagement process has also enabledstakeholders to envision a number of opportunities that the new technologies empower that can becontinued beyond SCOREwater.Moreover, due to a fruitful collaboration between developers and stakeholders, both Gothenburg andBarcelona cities now envision even more opportunities and ambitions, some of which might be realizedwithin the project, some outside.Both Amersfoort and Barcelona also analyse how insights from different organizational issues influencedtheir design of process facilitators and discursive abilities.The Amersfoort section shows the influencefrom a) the different organizational structures and motives between the municipality, the for-profitcompanies and citizen volunteers and b) the differences in work processes between their policy makersand data analysts, people that seemed to be previously unfamiliar with working together.The first issuewas addressed through finding common ground in the objectives (added value as a discursive device) andthe second through designing a common process – going from simple to more complex hypothesis.In the Barcelona case, BCASA realized a) that they needed to “translate” their needs and concepts withregard to wastewater maintenance to other Catalan partners and b) that they needed to involve andengage both workers and managers at several departments for the SCOREwater project so that theyunderstand.see and value the benefits it provide and therefore engage in and support the SCOREwaterproject. EUT, TP and CIV expressed fewer specific challenges than the case studies, using familiar andproved process facilitators and discursive devices (CRISP-DM business modelling and user stories).

    • DNA methylation profiles of aggressive behavior may capture lifetime cumulative effects of genetic, stochastic, and environmental influences associated with aggression. Here, we report the first large meta-analysis of epigenome-wide association studies (EWAS) of aggressive behavior (N = 15,324 participants). In peripheral blood samples of 14,434 participants from 18 cohorts with mean ages ranging from 7 to 68 years, 13 methylation sites were significantly associated with aggression (alpha = 1.2 × 10−7; Bonferroni correction). In cord blood samples of 2425 children from five cohorts with aggression assessed at mean ages ranging from 4 to 7 years, 83% of these sites showed the same direction of association with childhood aggression (r = 0.74, p = 0.006) but no epigenome-wide significant sites were found. Top-sites (48 at a false discovery rate of 5% in the peripheral blood meta-analysis or in a combined meta-analysis of peripheral blood and cord blood) have been associated with chemical exposures, smoking, cognition, metabolic traits, and genetic variation (mQTLs). Three genes whose expression levels were associated with top-sites were previously linked to schizophrenia and general risk tolerance. At six CpGs, DNA methylation variation in blood mirrors variation in the brain. On average 44% (range = 3–82%) of the aggression–methylation association was explained by current and former smoking and BMI. These findings point at loci that are sensitive to chemical exposures with potential implications for neuronal functions. We hope these results to be a starting point for studies leading to applications as peripheral biomarkers and to reveal causal relationships with aggression and related traits.
