  • Immunoreactive trypsin(Ogen) in the sera of children with recent-onset insulin-dependent diabetes and matched controls
  • 1990
  • Ingår i: Pancreas. - : Ovid Technologies (Wolters Kluwer Health). - 0885-3177. ; 5:3, s. 241-247
  • Tidskriftsartikel (refereegranskat)abstract
    • To evaluate the exocrine pancreatic function at the time of diagnosis of insulin-dependent diabetes mellitus, we determined immunoreactive an-odal and cathodal trypsin(ogen) levels in sera from almost all children (n = 375) 0-14 years of age in Sweden in whom diabetes developed during 1 year, and in sex-, age-, and geographically matched control subjects (n = 312). The median level of anodal trypsin(ogen) was 5 (quartile range, 3-7) µg/L in children with newly diagnosed diabetes, compared with a median level of 7 (quartile range, 4-8) µg/L in control subjects (p < 0.0001). Similarly, the median level of cathodal trypsin(ogen) was 8 (quartile range, 4-10) µg/L in children with diabetes, compared with a median level of 11 (quartile range, 7-15) µg/L in control subjects (p < 0.0001). The median of the individual ratios between cathodal and anodal trypsin(ogen) was 1.4 in the diabetic patients and 1.7 in the control children (p < 0.001). In a multivariate test, however, only the decrease in cathodal trypsin(ogen) concentration was associated with diabetes. The levels of trypsin(ogen)s did not correlate with levels of islet cell antibodies, present in 81% of the diabetic children. Several mechanisms may explain our findings, for example, similar pathogenetic factors may affect both the endocrine and exocrine pancreas simultaneously, a failing local trophic stimulation by insulin on the exocrine cells may decrease the trypsinogen production, and there may be an increased elimination of trypsin(ogen) because of higher filtration through the kidneys in the hyperglycemic state.

  • Evidence for iC3 generation during cardiopulmonary bypass as the result of blood-gas interaction.
  • 1993
  • Ingår i: Clinical and Experimental Immunology. - 0009-9104 .- 1365-2249. ; 91:3, s. 404-409
  • Tidskriftsartikel (refereegranskat)abstract
    • Earlier we have shown that iC3 is generated at the blood-gas interface in vitro and that the generation of this molecule is independent of complement activation and the composition of the gas. In order to investigate whether iC3 is also generated during cardiopulmonary bypass where blood comes into contact with oxygen bubbles, two bubble oxygenators were incubated at 37 degrees C with human heparinized blood. A continuous increase in the level of iC3 was shown in the oxygen-perfused bubble oxygenator (up to 100 nmol/l after 180 min) in contrast to the unbubbled control. Similarly, in plasma drawn from patients undergoing cardiopulmonary bypass using either bubble or membrane oxygenators, the levels of iC3 were shown to increase continuously during the operation. Furthermore, this form of C3 was found to be susceptible to cleavage by factor I. The formation of iC3 at the blood-gas interface in vivo could be a mechanism by which gas bubbles induce clinical manifestations associated with complement activation, e.g. during cardiopulmonary bypass, adult respiratory distress syndrome and decompression sickness.

  • Quantum interference effects in percolated metal networks
  • 1994
  • Ingår i: Physica B: Condensed Matter. - : Elsevier BV. - 0921-4526. ; , s. 1109-1110
  • Konferensbidrag (refereegranskat)abstract
    • Quantum corrections (QC) to the resistivity due to weak localization (WL) and electron-electron interaction (EEI) have been measured for percolated metal networks with percolation coherence length z comparable to the quantum length scales-the phase-breaking length L and the thermal diffusion length LT. Far from the percolation metal-insulator transition (MIT) the `macroscopic' disorder is averaged over the quantum lengths (L,LTz) and the QC are of the universal two-dimensional (2D) type with R=R(z), where R(z) is the resistance per square at scales larger than z. A crossover 2D1D in the dimensionality of the QC and the Nyquist phase-braking mechanism is observed near the MIT, where zL,LT

  • Study of Neutron Rich Neon Isotopes
  • 1992
  • Ingår i: Zeitschrift fur Physik A Hadrons and Nuclei. - 1431-5831 .- 0939-7922. ; 342:3, s. 303-307
  • Tidskriftsartikel (refereegranskat)abstract
    • The half-lives and P(n)-values of the neutron rich isotopes Ne-26-29 have been determined. The results are compared to shell-model calculations and good agreement is found except for Ne-29, where the half-life exceeds the predictions by more than an order of magnitude. This unexpectedly long half-life can be explained as due to a fp intruder configuration.

  • The cost of screening for hypercholesterolaemia- results from a clinical trial in Swedish primary health care
  • 1993
  • Ingår i: Scandinavian journal of clinical and laboratory investigation. - : Informa UK Ltd. - 1502-7686 .- 0036-5513. ; 53:7, s. 725-732
  • Tidskriftsartikel (refereegranskat)abstract
    • A convenience sample of the population was screened for inclusion in a study of the effectiveness of a lipid lowering programme. Included in the study were adults 30-59 years of age with a S-Cholesterol of 6.50 7.79 mmol 1_1 at randomization, plus two previous values ≥ 6.50 mmol 1_1 at screening and selection, with at least two other cardiovascular risk factors. In total 447 persons were screened and 37 were randomized into the lipid lowering programme. The mean cost per randomized person was estimated at about SEK 7500 (Swedish Crowns). An analysis of different inclusion criteria for treatment was also carried out. The cost of finding a patient to treat in the clinical trial was estimated to be more than three times as high with a total cholesterol cut-off point of 7.80 mmol 1−1 compared with a cut-off point of 5.20mmol 1−1.

  • Competitive elution of protein A fusion proteins allows specific recovery under mild conditions.
  • 1994
  • Ingår i: European Journal of Biochemistry. - : Wiley. - 0014-2956 .- 1432-1033. ; 224:1, s. 103-8
  • Tidskriftsartikel (refereegranskat)abstract
    • A novel system is described for mild elution of fusion proteins by competitive elution. The approach is based on displacement of immobilized fusions containing a monovalent IgG-binding staphylococcal protein A fragment (Z) from an IgG-affinity matrix by a divalent fragment fused to a serum-albumin-binding region derived from streptococcal protein G. Using real-time interaction analysis, the binding (K(aff)) to polyclonal human IgG was found to be 3.3 (+/- 0.4) x 10(8) M-1 for divalent ZZ and 2.0 (+/- 0.1) x 10(7) M-1 for monovalent Z. This more than tenfold difference in binding strength ensures a high efficiency in the elution step. The competitor protein can specifically be removed and recovered from the elution mixture by subsequent passage through a human serum albumin(HSA)-affinity column, leaving only the target fusion protein in the flow-through fraction. Here, we show that a recombinant Klenow fragment of DNA polymerase I expressed in Escherichia coli can be recovered with high yield, and retained activity, from a crude bacterial lysate by IgG-affinity chromatography using mild conditions during both binding and elution.
