  • Surface-adsorbed fibrinogen and fibrin may activate the contact activation system

    • INTRODUCTION: This study was designed to investigate whether fibrinogen, soluble desAA-fibrin, and insoluble desAABB-fibrin are able to induce clotting by triggering the plasma contact activation system when adsorbed to polystyrene. MATERIALS AND METHODS: The above-mentioned substances were individually prepared on polystyrene meshwork squares, and then exposed to a purified FXII solution or non-calcium containing plasma (citrated and dialyzed normal pooled plasma) in polystyrene cuvettes coated with surface-immobilized heparin, to completely block contact activation and the coagulation mechanism that might be induced by the cuvette surfaces. Sodium glass beads were used as the reference material. RESULTS: On exposure to purified FXII solution and plasma, all the tested materials adsorbed and activated FXII to varying degrees. This activation led to the formation of FXIa in the exposed plasma, with the highest activation occurring upon exposure to glass, desAA-fibrin and desAABB-fibrin and the lowest upon exposure to fibrinogen-adsorbed or unmodified polystyrene meshwork squares. Following recalcification, in cuvettes with surface-immobilized heparin, a spectrophotometric assay showed that the surface-exposed plasma aliquots clotted within 5 min after contact with glass, within 10 to 15 min after contact with the two forms of fibrin, and somewhat longer after contact with adsorbed fibrinogen. The longest lag phase, close to 20 min, occurred in plasma exposed to unmodified polystyrene meshwork. Whole blood deposited in surface heparinized cuvettes directly from the cubital vein did not clot during the observation time (2 h). CONCLUSIONS: These results indicate that domains induced by conformational changes in adsorbed fibrinogen and fibrin are capable of activating adsorbed proenzymes and that various forms of fibrin are considerably stronger activators of the contact activation system than are adsorbed fibrinogen or a polystyrene meshwork. The delayed coagulation in plasma exposed to the unmodified polystyrene meshwork can be explained by a two-step process: first, adsorption of fibrinogen, and second, activation of FXII. Under our experimental conditions, the adsorption and activation of FXII on fibrinogen and fibrin seems to be an important mechanism for triggering coagulation.

    • Aim. Calcium phosphate deposition in blood vessels is correlated to increased mortality risk. In this study, the formation of solid calcium phosphate in an in vitro solution mimicking the inorganic part of blood serum was studied. Methods. The precipitates formed were analyzed using several experimental techniques, including infrared spectroscopy, Raman spectroscopy, X-ray energy dispersive spectroscopy, chemical analysis of combustion gases, thermogravimetric analysis, as well as transmission electron microscopy and scanning electron microscopy. Results. The results indicate a microscopically amorphous but nano-crystalline material with an overall apatite structure. A plausible stoichiometry was determined to Ca-5(PO4)(3)(HCO3)center dot 4H(2)O with an estimated solubility constant of 6.10(-39) (mol/L)(9). Bicarbonate in the water solution was shown to be essential for the precipitation, giving implications for in vitro studies. Conclusions. The calcium phosphate formed in this study shows many similarities to pathological calcium phosphates regarding composition, morphology and crystallinity.

    • The angiogenesis inhibitor histidine-rich glycoprotein (HRG) constitutes one of several examples of molecules regulating both angiogenesis and hemostasis. The antiangiogenic properties of HRG are mediated via its proteolytically released histidine- and proline-rich (His/Pro-rich) domain.Using a combination of immunohistochemistry and massspectrometry, we here provide biochemical evidence for thepresence of a proteolytic peptide, corresponding to the antiangiogenic domain of HRG, in vivo in human tissue. This finding supports a role for HRG as an endogenous regulator of angiogenesis. Interestingly, the His/Pro-rich peptide bound to the vessel wall in tissue from cancer patients but not to the vasculature in tissue from healthy persons.Moreover, the His/Pro-rich peptide was found in close association with platelets. Relesate from in vitro–activated platelets promoted binding of the His/Pro-rich domain of HRG to endothelial cells, an effect mediated by Zn2+.Previous studies have shown that zinc-dependent bindingof the His/Pro-rich domain of HRG to heparan sulfate on endothelial cells is required for inhibition of angiogenesis.We describe a novel mechanism to increase the local concentration and activity of an angiogenesis inhibitor,which may reflect a host response to counteract angiogenesis during pathologic conditions. Our finding that tumor angiogenesis is elevated in HRG-deficient mice supports this conclusion.

  • Delayed harvest of reed canary grass translocates more nutrients in rhizomes

    • Two field experiments, one in large plots and the other in small framed plots, were conducted in Umea°, northern Sweden. The objectives were (1) to examine the seasonal patterns of rhizome growth and nutrient dynamics of the energy crop reed canary grass (Phalaris arundinacea L.) in ley I and II, and (2) to evaluate the roles of soil type (mineral vs. organic), fertilisation level (0, 50, and 100 kg N ha-1), and season/harvest time (Oct-96, May-97, and Aug-97) on the rhizome growth and nutrient dynamics by means of a factorially designed experiment. The general pattern of rhizome growth was that biomass was low in June during initiation of shoot growth, but increased steadily during the growing season, reached a peak in late autumn, and remained high until next spring. The N and P accumulation in rhizomes followed a similar pattern. During ley years I and II, reed canary grass rhizome growth was less dependent on soil type, and more dependent on fertilisation and season, with fertilisation being the most important predictor of growth. The season/harvest time, followed by soil type, was the most important factor for both concentrations and therefore total uptake of N, P, and K in rhizomes. Soil type affected N content in rhizomes significantly, and also interacted with season and enhanced the effect on N, P, and K content in rhizomes. The seasonal dynamics of the nutrient content in rhizomes indicate a remobilisation of the nutrients from rhizomes to the regrowth of shoots and roots in spring and relocation/storage from aboveground shoots to rhizomes during late summer and autumn. The results of this study suggest that delaying the harvest to later than October would result in considerably more energy and nutrient resources being translocated from aboveground shoots to rhizomes for growth in the next season
