  • The role of learning in transdisciplinary research: moving from a normative concept to an analytical tool through a practice-based approach

    • Transdisciplinary (TD) research is an example of a participatory research approach that has been developed to address the complexity of societal problems through the exchange of knowledge and expertise across diverse groups of societal actors. The concept of knowledge exchange is central to the ability of TD research to produce usable knowledge. There is, however, limited theoretical attention to the processes that enable knowledge exchange, namely learning. In this article, we analyze the "transferability" of knowledge generated in TD research settings from a practice-based approach. In this approach, learning and knowing are seen as situated in social practices, in meaning making processes where the involved participants make sense of what they do and why they do it. We describe and analyze three TD projects, and discuss the role of practitioners' perspectives in the interpretation of the tasks and realization of TD, and in the consequences this has for the organization of the research process and the usability of its results. The analysis shows that while the project teams were given the same task and framework, they did not understand or enact TD in a similar fashion. The three projects created different goals and organizations. They also resulted in different challenges, which could be identified and analyzed by the use of a practice-based approach to learning. In the conclusions, we identify aspects for both practice and research that are important for creating sufficient conditions for learning in TD research processes so that they can better promote contributions to societal change.

    • We consider TCP performance for cloud services in 5G cellular networks and show that edge clouds offer the highest throughput, but that further improvements still are possible. To this end we devise a concept where edge clouds connected directly to the radio access network can increase their transmission rates by relying on the built in re-transmissions (through quality of service features) and on the built in user fairness (through per-user buffers and scheduler policies). We use numerical examples from measurements in real networks to demonstrate that the concept is efficient and significantly improves the effective throughput beyond what placement alone can do, and we also outline possible further improvements.

    • We consider bottlenecks of TCP throughput in scenarios with edge clouds and 5G cellular networks. By numerical examples from measurements in real networks, we show that edge clouds indeed improve throughput but that several, non-negligible bottlenecks remain. We therefore devise a concept where edge clouds connected directly to the radio access network can increase their transmission rates by relying on built in retransmissions (through quality of service features) and on the built in user fairness (through per-user buffers and scheduler policies). We then return to the numerical examples and show that our solution provides substantial gains and we conclude by identifying and discussing the remaining bottlenecks and the potential of an improved protocol.

    • Intranasal oxytocin (OT) has previously been found to increase spirituality, an effect moderated by OT-related genotypes. This pre-registered study sought to conceptually replicate and extend those findings. Using a single dose of intranasal OT vs placebo (PL), we investigated experimental treatment effects, and moderation by OT-related genotypes on spirituality, mystical experiences, and the sensed presence of a sentient being. A more exploratory aim was to test for interactions between treatment and the personality disposition absorption on these spirituality-related outcomes. A priming plus sensory deprivation procedure that has facilitated spiritual experiences in previous studies was used. The sample (N = 116) contained both sexes and was drawn from a relatively secular context. Results failed to conceptually replicate both the main effects of treatment and the treatment by genotype interactions on spirituality. Similarly, there were no such effects on mystical experiences or sensed presence. However, the data suggested an interaction between treatment and absorption. Relative to PL, OT seemed to enhance spiritual experiences in participants scoring low in absorption and dampen spirituality in participants scoring high in absorption.

  • The role of ghrelin signalling for sexual behaviour in male mice.

    • Ghrelin, a gut-brain signal, is well known to regulate energy homeostasis, food intake and appetite foremost via hypothalamic ghrelin receptors (GHS-R1A). In addition, ghrelin activates the reward systems in the brain, namely the mesolimbic dopamine system, and regulates thereby the rewarding properties of addictive drugs as well as of palatable foods. Given that the mesolimbic dopamine system mandates the reinforcing properties of addictive drugs and natural rewards, such as sexual behaviour, we hypothesize that ghrelin plays an important role for male sexual behaviour, a subject for the present studies. Herein we show that ghrelin treatment increases, whereas pharmacological suppression (using the GHSR-1A antagonist JMV2959) or genetic deletion of the GHS-R1A in male mice decreases the sexual motivation for as well as sexual behaviour with female mice in oestrus. Pre-treatment with L-dopa (a dopamine precursor) prior to treatment with JMV2959 significantly increased the preference for female mouse compared with vehicle treatment. On the contrary, treatment with 5-hydroxythyptohan (a precursor for serotonin) prior to treatment with JMV2959 decreased the sexual motivation compared to vehicle. In separate experiments, we show that ghrelin and GHS-R1A antagonism do not affect the time spent over female bedding as measured in the androgen-dependent bedding test. Collectively, these data show that the hunger hormone ghrelin and its receptor are required for normal sexual behaviour in male mice and that the effects of the ghrelin signalling system on sexual behaviour involve dopamine neurotransmission.

  • Polymorphisms in genes in the androgen pathway and risk of Barrett's esophagus and esophageal adenocarcinoma

    • The strong male predominance in Barrett's esophagus (BE) and esophageal adenocarcinoma (EAC) remains inadequately explained, but sex hormones might be involved. We hypothesized that single nucleotide polymorphisms (SNPs) in the androgen pathway influence risk of developing BE and EAC. This genetic-epidemiological analysis included 14 studies from Australia, Europe and North America. Polymorphisms in 16 genes coding for the androgen pathway were analyzed using a gene-based approach: versatile gene-based test association study. This method evaluates associations between a trait and all SNPs within a specific gene rather than each SNP marker individually as in a conventional GWAS. The data were stratified for sex, body-mass index, waist-to-hip ratio, tobacco smoking and gastroesophageal reflux status. Included were data from 1,508 EAC patients, 2,383 BE patients and 2,170 control participants. SNPs within the gene CYP17A1 were associated with risk of BE in the sexes combined (p=0.002) and in males (p=0.003), but not in females separately (p=0.3). This association was found in tobacco smokers (p=0.003) and in BE patients without reflux (p=0.004), but not in nonsmokers (p=0.2) or those with reflux (p=0.036). SNPs within JMJD1C were associated with risk of EAC in females (p=0.001). However, none of these associations replicated in a subsequent sample. Fourteen other genes studied did not reach statistically significant levels of association with BE, EAC or the combination of BE and EAC, after correcting for the number of genes included in the analysis. In conclusion, genetic variants in the androgen-related genes CYP17A1 and JMJD1C might be associated with risk of BE and EAC, respectively, but replication data with larger sample sizes are needed.

  • Female Mice Lacking Estrogen Receptor-alpha in Hypothalamic Proopiomelanocortin (POMC) Neurons Display Enhanced Estrogenic Response on Cortical Bone Mass

    • Estrogens are important regulators of bone mass and their effects are mainly mediated via estrogen receptor(ER)alpha. Central ER alpha exerts an inhibitory role on bone mass. ER alpha is highly expressed in the arcuate (ARC) and the ventromedial (VMN) nuclei in the hypothalamus. To test whether ER alpha in proopiomelanocortin (POMC) neurons, located in ARC, is involved in the regulation of bone mass, we used mice lacking ER alpha expression specifically in POMC neurons (POMC-ER alpha(-/-)). Female POMC-ER alpha(-/-) and control mice were ovariectomized (OVX) and treated with vehicle or estradiol (0.5 mu g/d) for 6 weeks. As expected, estradiol treatment increased the cortical bone thickness in femur, the cortical bone mechanical strength in tibia and the trabecular bone volume fraction in both femur and vertebrae in OVX control mice. Importantly, the estrogenic responses were substantially increased in OVX POMC-ER alpha(-/-) mice compared with the estrogenic responses in OVX control mice for cortical bone thickness (+ 126 +/- 34%, P < .01) and mechanical strength (+ 193 +/- 38%, P <.01). To test whether ER alpha in VMN is involved in the regulation of bone mass, ER alpha was silenced using an adeno-associated viral vector. Silencing of ER alpha in hypothalamic VMN resulted in unchanged bone mass. In conclusion, mice lacking ER alpha in POMC neurons display enhanced estrogenic response on cortical bone mass and mechanical strength. We propose that the balance between inhibitory effects of central ER alpha activity in hypothalamic POMC neuronsin ARC and stimulatory peripheral ER alpha-mediated effects in bone determines cortical bone mass in female mice.

    • Female sexual desire and arousal problems have been shown to have a heritable component of moderate size. Previous molecular genetic studies on sexual desire have mainly focused on genes associated with neurotransmitters such as dopamine and serotonin. Nevertheless, there is reason to believe that hormones with more specific functions concerning sexuality could have an impact on sexual desire and arousal.

  • Adaptive management? Observations of knowledge coordination in the communication practice of Swedish game management

    • Modern management of natural resources is guided by the normative theory of adaptive management (AM). Behind this theory lies a strong, albeit implicit, expectation that organisations aiming for AM have the capacity to communicate in a way that facilitates the required coordination of the knowledge perspectives involved. The aim of this article is to discuss the extent to which the communication practice of Swedish game management organisations facilitates coordination of knowledge corresponding to AM. Based on operationalizations of communicative rationality and agonistic pluralism, we use the concepts 'discursive closure' and 'discursive opening' to investigate how the coordination of knowledge is carried out through communication in relatively recently established organisations, the Swedish Game Management Delegations (GMDs). We analyse four communication episodes from GMD meetings and notice that multiple perspectives were expressed (discourse openings) but were not evaluated in a communicative rational way before being closed. The consequences of these closures were that knowledge perspectives with potential relevance, but with unclear validity for game management, were not elaborated upon, in terms of their truth, intelligibility, legitimacy or sincerity, which inhibited AM. The concepts of discursive closure and discursive opening proved useful for investigating communicative capacity. An important question which needs to be addressed to improve communicative capacity for AM is whether it would be practically possible to keep to the agenda and rules of the GMD meetings and still admit discursive openings about differences in perspectives.
