    • The two dimethyl sulfoxide solvated rhodium(III) compounds, [Rh(dmso-kappa O)(5)(dmso-kappa S)](CF(3)SO(3))(3) (1 & 1* at 298 K and 100 K, respectively) and [Rh(dmso-kappa O)(3)(dmso-kappa S)(2)Cl](CF(3)SO(3))(2) (2), crystallize with orthorhombic unit cells in the space group Pna2(1) (No. 33), Z = 4. In the [Rh(dmso)(6)](3+) complex with slightly distorted octahedral coordination geometry, the Rh-O bond distance is significantly longer with O trans to S, 2.143(6) angstrom (1) and 2.100(6) angstrom (1*), than the mean Rh-O bond distance with O trans to O, 2.019 angstrom (1) and 2.043 angstrom (1*). In the [RhCl(dmso)(5)](3+) complex, the mean Rh-O bond distance with O trans to S, 2.083 angstrom, is slightly longer than that for O trans to Cl, 2.067(4) angstrom, which is consistent with the trans influence DMSO-kappa S > Cl > DMSO-kappa O of the opposite ligands. Raman and IR absorption spectra were recorded and analyzed and a complete assignment of the vibrational bands was achieved with support by force field calculations. An increase in the Rh-O stretching vibrational frequency corresponded to a decreasing trans-influence from the opposite ligand. The Rh-O force constants obtained were correlated with the Rh-O bond lengths, also including previously obtained values for other M(dmso)(6)(3+) complexes with trivalent metal ions. An almost linear correlation was obtained for the MO stretching force constants vs. the reciprocal square of the MO bond lengths. The results show that the metal ion-oxygen bonding of dimethyl sulfoxide ligands is electrostatically dominated in those complexes and that the stretching force constants provide a useful measure of the relative trans-influence of the opposite ligands in hexa-coordinated Rh(III)-complexes.

  • Genetic dissection of quantitative traits in Scots pine

    • The phenotypic expressions of Scots pine, have been extensively studied over the years, but the gene regulation behind the traits has only just begun to be elucidated. The overall aim of this thesis was to start dissecting the genetics behind a number of adaptive traits in Scots pine and examine how they are influenced by relatedness using different molecular tools. In a full-sib family of Scots pine the genetic variation in autumn frost hardiness and height growth was revelead by measuring open-pollinated offspring. A significant genetic variation for both traits was found, a prerequisite to identify quantitative trait loci (QTL). A conceptual statistical model in the Bayesian framework for identifying QTLs in dynamic traits i.e. traits that vary over time, was developed and applied in a QTL study based on a full-sib family of 250 trees. A set of 160 AFLPs were utilized. QTLs for three latent traits were identified: one for the slope (growth) and two for the quadratic term (growth cessation). As a tool to identify candidate genes for the study of quantitative adaptive traits, gene regulation under continuous red (cR) and far-red (cFR) light was studied in hypocotyls from open-pollinated seeds from a natural population in northern Sweden using microarray technology. The gene expression patterns for the light response pathway in Scots pine under cFR show clear differences from those of angiosperms, wherein we observed up-regulation of cryptochrome1. This gene has, therefore, become a strong candidate gene that deserves further studies to elucidate the genetics behind Scots pine adaptation. Not knowing the genetic relationship and inbreeding of trees, and how it influences the phenotypic expression, can lead to over- or underestimation of additive genetic values resulting in biased heritability estimates. A natural population of Scots pine, earlier identified as being highly inbreed, was used to investigate the influence of inbreeding by the correlation between heterozygosity and propotion of sound seed (PSS), average seed weight and proportion of rare alleles (PRA). Heterozygosity fitness correlation (HFC) was found positive for PSS and negative for PRA most likely due to recessive deleterious alleles purged in homozygotes. The study provided evidence that, as predicted by theory, inbreeding enhances HFC in a species with high outbreeding rate and high number of lethal equivalents as Scots pine.

    • Associations of salicylate-like phenolic glycosides (PGs) with biological activity have been reported in Salix and Populus trees, but only for a few compounds, and in relation to a limited number of herbivores. By considering the full diversity of PGs, we may improve our ability to recognize genotypes or chemotype groups and enhance our understanding of their ecological function. Here, we present a fast and efficient general method for salicylate determination in leaves of Eurasian aspen that uses ultra-high performance liquid chromatography-electrospray ionization/time-of-flight mass spectrometry (UHPLC-ESI/TOFMS). The time required for the liquid chromatography separations was 13.5 min per sample, compared to around 60 min per sample for most HPLC protocols. In leaf samples from identical P. tremula genotypes with diverse propagation and treatment histories, we identified nine PGs. We found the compound-specific mass chromatograms to be more informative than the UV-visible chromatograms for compound identification and when quantitating samples with large variability in PG content. Signature compounds previously reported for P. tremoloides (tremulacin, tremuloidin, salicin, and salicortin) always were present, and five PGs (2'-O-cinnamoyl-salicortin, 2'-O-acetyl-salicortin, 2'-O-acetyl-salicin, acetyl-tremulacin, and salicyloyl-salicin) were detected for the first time in P. tremula. By using information about the formic acid adduct that appeared for PGs in the LTQ-Orbitrap MS environment, novel compounds like acetyl-tremulacin could be tentatively identified without the use of standards. The novel PGs were consistently either present in genotypes regardless of propagation and damage treatment or were not detectable. In some genotypes, concentrations of 2'-O-acetyl-salicortin and 2'-O-cinnamoyl-salicortin were similar to levels of biologically active PGs in other Salicaceous trees. Our study suggests that we may expect a wide variation in PG content in aspen populations which is of interest both for studies of interactions with herbivores and for mapping population structure.

    • In this study, different growth conditions of Anthracophyllum discolor Sp4 including the effect of agitation, additions of lignocellulosic support, inducer and surfactant were evaluated on the MnP production in Kirk medium using a culture system made up of the tubes containing the glass bead. The highest MnP production (1,354 U/L on day 13) was obtained when the medium was supplemented with wheat grain and 0.25 mM MnSO 4 as inducer, under static conditions at 30°C. Two isoenzymes were purified (35 and 38 kDa respectively). MnP presented a maximal activity in the pH range between 4.5 and 5.5, a relatively high temperature tolerance (50°C) and a high catalytic activity for 2,6-dimethoxyphenol and hydrogen peroxide.

    • Manganese peroxidase (MnP) produced by Anthracophyllum discolor, a Chilean white rot fungus, was immobilized on nanoclay obtained from volcanic soil and its ability to degrade polycyclic aromatic hydrocarbons (PAHs) compared with the free enzyme was evaluated. At the same time, nanoclay characterization was performed.Nanoclay characterization by transmission electronic microscopy showed a particle average size smaller than 100nm. The isoelectric points (IEP) of nanoclay and MnP from A. discolor were 7.0 and 3.7, respectively, as determined by micro electrophoresis migration and preparative isoelectric focusing. Results indicated that 75% of the enzyme was immobilized on the nanoclay through physical adsorption. As compared to the free enzyme, immobilized MnP from A. discolor achieved an improved stability to temperature and pH. The activation energy (Ea) value for immobilized MnP (51.9kJmol -1) was higher than that of the free MnP (34.4kJmol -1).The immobilized enzyme was able to degrade pyrene (>86%), anthracene (>65%), alone or in mixture, and to a less extent fluoranthene (<15.2%) and phenanthrene (<8.6%). Compared to free MnP from A. discolor, the enzyme immobilized on nanoclay enhanced the enzymatic transformation of anthracene in soil.Overall results indicate that nanoclay, a carrier of natural origin, is a suitable support material for MnP immobilization. In addition, immobilized MnP shows an increased stability to high temperature, pH and time storage, as well as an enhanced PAHs degradation efficiency in soil. All these characteristics may suggest the possible use of nanoclay-immobilized MnP from A. discolor as a valuable option for in situ bioremediation purposes. © 2010 Elsevier Ltd.

    • The degradation of three- and four-ring polycyclic aromatic hydrocarbons (PAHs) in Kirk medium by Anthracophyllum discolor, a white-rot fungus isolated from the forest of southern Chile, was evaluated. In addition, the removal efficiency of three-, four- and five-ring PAHs in contaminated soil bioaugmented with A. discolor in the absence and presence of indigenous soil microorganisms was investigated. Production of lignin-degrading enzymes and PAH mineralization in the soil were also determined. A. discolor was able to degrade PAHs in Kirk medium with the highest removal occurring in a PAH mixture, suggesting synergistic effects between PAHs or possible cometabolism. A high removal capability for phenanthrene (62%), anthracene (73%), fluoranthene (54%), pyrene (60%) and benzo(a)pyrene (75%) was observed in autoclaved soil inoculated with A. discolor in the absence of indigenous microorganisms, associated with the production of manganese peroxidase (MnP). The metabolites found in the PAH degradation were anthraquinone, phthalic acid, 4-hydroxy-9-fluorenone, 9-fluorenone and 4,5-dihydropyrene. A. discolor was able to mineralize 9% of the phenanthrene. In non-autoclaved soil, the inoculation with A. discolor did not improve the removal efficiency of PAHs. Suitable conditions must be found to promote a successful fungal bioaugmentation in non-autoclaved soils. © 2010 Elsevier B.V.
