  • Biomass offsets little or none of permafrost carbon release from soils, streams, and wildfire : an expert assessment

    • As the permafrost region warms, its large organic carbon pool will be increasingly vulnerable to decomposition, combustion, and hydrologic export. Models predict that some portion of this release will be offset by increased production of Arctic and boreal biomass; however, the lack of robust estimates of net carbon balance increases the risk of further overshooting international emissions targets. Precise empirical or model-based assessments of the critical factors driving carbon balance are unlikely in the near future, so to address this gap, we present estimates from 98 permafrost-region experts of the response of biomass, wildfire, and hydrologic carbon flux to climate change. Results suggest that contrary to model projections, total permafrost-region biomass could decrease due to water stress and disturbance, factors that are not adequately incorporated in current models. Assessments indicate that end-of-the-century organic carbon release from Arctic rivers and collapsing coastlines could increase by 75% while carbon loss via burning could increase four-fold. Experts identified water balance, shifts in vegetation community, and permafrost degradation as the key sources of uncertainty in predicting future system response. In combination with previous findings, results suggest the permafrost region will become a carbon source to the atmosphere by 2100 regardless of warming scenario but that 65%-85% of permafrost carbon release can still be avoided if human emissions are actively reduced.

  • A metabolite roadmap of the wood-forming tissue in Populus tremula

    • Wood, or secondary xylem, is the product of xylogenesis, a developmental process that begins with the proliferation of cambial derivatives and ends with mature xylem fibers and vessels with lignified secondary cell walls. Fully mature xylem has undergone a series of cellular processes, including cell division, cell expansion, secondary wall formation, lignification and programmed cell death. A complex network of interactions between transcriptional regulators and signal transduction pathways controls wood formation. However, the role of metabolites during this developmental process has not been comprehensively characterized. To evaluate the role of metabolites during wood formation, we performed a high spatial resolution metabolomics study of the wood-forming zone of Populus tremula, including laser dissected aspen ray and fiber cells. We show that metabolites show specific patterns within the wood-forming zone, following the differentiation process from cell division to cell death. The data from profiled laser dissected aspen ray and fiber cells suggests that these two cell types host distinctly different metabolic processes. Furthermore, by integrating previously published transcriptomic and proteomic profiles generated from the same trees, we provide an integrative picture of molecular processes, for example, deamination of phenylalanine during lignification is of critical importance for nitrogen metabolism during wood formation.

  • The sedimentary and remote-sensing reflection of biomass burning in Europe

    • Aim: We provide the first European-scale geospatial training set relating the charcoal signal in surface lake sediments to fire parameters (number, intensity and area) recorded by satellite moderate resolution imaging spectroradiometer (MODIS) sensors. Our calibration is intended for quantitative reconstructions of key fire-regime parameters by using sediment sequences of microscopic (MIC from pollen slides, particles 10-500 mu m) and macroscopic charcoal (MAC from sieves, particles > 100 mu m). Location: North-south and east-west transects across Europe, covering the mediterranean, temperate, alpine, boreal and steppe biomes. Time period: Lake sediments and MODIS active fire and burned area products were collected for the years 2012-2015. Methods: Cylinder sediment traps were installed in lakes to annually collect charcoal particles in sediments. We quantitatively assessed the relationships between MIC and MAC influx (particles/cm(2)/year) and the MODIS-derived products to identify source areas of charcoal and the extent to which lake-sediment charcoal is linked to fire parameters across the continent. Results: Source area of sedimentary charcoal was estimated to a 40-km radius around sites for both MIC and MAC particles. Fires occurred in grasslands and in forests, with grass morphotypes of MAC accurately reflecting the burned fuel-type. Despite the lack of local fires around the sites, MAC influx levels reached those reported for local fires. Both MIC and MAC showed strong and highly significant relationships with the MODIS-derived fire parameters, as well as with climatic variation along a latitudinal temperature gradient. Main conclusions: MIC and MAC are suited to quantitatively reconstructing fire number and fire intensity on a regional scale. However, burned area may only be estimated using MAC. Local fires may be identified by using several lines of evidence, e.g. analysis of large particles (> 600 mu m), magnetic susceptibility and sedimentological data. Our results offer new insights and applications to quantitatively reconstruct fires and to interpret available sedimentary charcoal records.

  • Pedunculate Oaks (Quercus robur L.) Differing in Vitality as Reservoirs for Fungal Biodiversity

    • Ecological significance of trees growing in urban and peri-urban settings is likely to increase in future land-use regimes, calling for better understanding of their role as potential reservoirs or stepping stones for associated biodiversity. We studied the diversity of fungal endophytes in woody tissues of asymptomatic even aged pedunculate oak trees, growing as amenity trees in a peri-urban setting. The trees were classified into three groups according to their phenotypic vitality (high, medium, and low). Endophytes were cultured on potato dextrose media from surface sterilized twigs and DNA sequencing was performed to reveal the taxonomic identity of the morphotypes. In xylem tissues, the frequency and diversity of endophytes was highest in oak trees showing reduced vitality. This difference was not found for bark samples, in which the endophyte infections were more frequent and communities more diverse than in xylem. In general, most taxa were shared across the samples with few morphotypes being recovered in unique samples. Leaf phenolic profiles were found to accurately classify the trees according to their phenotypic vitality. Our results confirm that xylem is more selective substrate for endophytes than bark and that endophyte assemblages in xylem are correlated to the degree of host vitality. Thus, high vitality of trees may be associated with reduced habitat quality to wood-associated endophytes.

  • Actin is bundled in activation-tagged tobacco mutants that tolerate aluminum

    • A panel of aluminum-tolerant (AlRes) mutants was isolated by protoplast-based T-DNA activation tagging in the tobacco cultivar SR1. The mutants fell into two phenotypic classes: a minority of the mutants were fertile and developed similarly to the wild type (type I), the majority was male-sterile and grew as semi-dwarfs (type II). These traits, along with the aluminum tolerance, were inherited in a monogenic dominant manner. Both types of mutants were characterized by excessive bundling of actin microfilaments and by a strongly increased abundance of actin, a phenotype that could be partially phenocopied in the wild type by treatment with aluminum chloride. The actin bundles could be dissociated into finer strands by addition of exogenous auxin in both types of mutants. However, actin microfilaments and leaf expansion were sensitive to blockers of actin assembly in the wild type and in the mutants of type I, whereas they were more tolerant in the mutants of type II. The mutants of type II displayed a hypertrophic development of vasculature, manifest in form of supernumerary leaf veins and extended xylem layers in stems and petioles. Whereas mutants of type I were characterized by a normal, but aluminum-tolerant polar auxin-transport, auxin-transport was strongly promoted in the mutants of type II. The phenotype of these mutants is discussed in terms of reduced endocytosis leading, concomitantly with aluminum tolerance, to changes in polar auxin transport.

  • Barium as a Potential Indicator of Phosphorus in Agricultural Runoff

    • In many catchments, anthropogenic input of contaminants, and in particular phosphorus (P), into surface water is a mixture of agricultural and sewage runoff. Knowledge about the relative contribution from each of these sources is vital for mitigation of major environmental problems such as eutrophication. In this study, we investigated whether the distribution of trace elements in surface waters can be used to trace the contamination source. Water from three groups of streams was investigated: streams influenced only by agricultural runoff, streams influenced mainly by sewage runoff, and reference streams. Samples were collected at different flow regimes and times of year and analyzed for 62 elements using ICP-MS. Our results show that there are significant differences between the anthropogenic sources affecting the streams in terms of total element composition and individual elements, indicating that the method has the potential to trace anthropogenic impact on surface waters. The elements that show significant differences between sources are strontium (p < 0.001), calcium (p < 0.004), potassium (p < 0.001), magnesium (p < 0.001), boron (p < 0.001), rhodium (p = 0.001), and barium (p < 0.001). According to this study, barium shows the greatest potential as a tracer for an individual source of anthropogenic input to surface waters. We observed a strong relationship between barium and total P in the investigated samples (R-2 = 0.78), which could potentially be used to apportion anthropogenic sources of P and thereby facilitate targeting of mitigation practices.

  • Identification and quantification of organic phosphorus forms in soils from fertility experiments

    • The effects of soil type, crop rotation, fertilizer type and application rate on the composition of organic phosphorus (P) compounds in soils from four sites in a Swedish long-term fertilizer experiment were investigated with 31P-NMR. Soil textures investigated were loamy sand, sandy loam, silty clay loam and clay. Phosphorus has been added to the soils since the 1950s and 1960s at four different rates in the form of either mineral fertilizer or a combination of manure and mineral fertilizer. Results show that in soils receiving no P addition, most of the soil P was present in the form of phosphate monoesters (6070%, depending on soil type). However, a P addition equivalent to the amount of P removed annually by harvest altered this relationship so that the soils were dominated by orthophosphate instead. This trend became more obvious with increasing P addition. At the greatest P application rate, orthophosphate comprised 70% or more of the total extracted P in all the soils. These changes in the soil were due entirely to increase in orthophosphate, because the amounts of monoesters did not change with increasing P additions. This was true both for mineral fertilizer and the combination of manure and mineral fertilizer P. Soil type and crop rotation did not influence the results. The results indicate that there is no apparent build-up of organic P in the soils, but that P addition mainly affects the orthophosphate amounts in the soils regardless of form or amount of fertilizer.

  • Widespread Unquantified Conversion of Old Boreal Forests to Plantations

    • Across the boreal biome, clear-cutting of old, previously non clear-cut forests with high naturalness followed by tree planting or seeding is a major land use change. However, how much previously uncut forest has been converted to plantations remains unquantified. We combine Swedish national databases on clear-cuts and forest inventories to show that at least 19% of all clear-cuts since 2003 have occurred in old forests that were most likely not previously cut and planted or seeded. Old forests have been cut and lost at a steady rate of ∼1.4% per year for the same period, and at this rate they will disappear by the 2070s. There is further evidence that this type of unreported forest conversion is occurring across much of the world's boreal forest.

    • Spent grains are one of the lignocellulosic biomasses available in abundance, discarded by breweries as waste. The brewing process generates around 25–30% of waste in different forms and spent grains alone account for 80–85% of that waste, resulting in a significant global waste volume. Despite containing essential nutrients, i.e., carbohydrates, fibers, proteins, fatty acids, lipids, minerals, and vitamins, efficient and economically viable valorization of these grains is lacking. Microbial fermentation enables the valorization of spent grain biomass into numerous commercially valuable products used in energy, food, healthcare, and biomaterials. However, the process still needs more investigation to overcome challenges, such as transportation, cost-effective pretreatment, and fermentation strategy. to lower the product cost and to achieve market feasibility and customer affordability. This review summarizes the potential of spent grains valorization via microbial fermentation and associated challenges.
