  • Non-Structural Proteins of Arthropod-Borne Bunyaviruses: Roles and Functions

    • Viruses within the Bunyaviridae family are tri-segmented, negative-stranded RNA viruses. The family includes several emerging and re-emerging viruses of humans, animals and plants, such as Rift Valley fever virus, Crimean-Congo hemorrhagic fever virus, La Crosse virus, Schmallenberg virus and tomato spotted wilt virus. Many bunyaviruses are arthropod-borne, so-called arboviruses. Depending on the genus, bunyaviruses encode, in addition to the RNA-dependent RNA polymerase and the different structural proteins, one or several non-structural proteins. These non-structural proteins are not always essential for virus growth and replication but can play an important role in viral pathogenesis through their interaction with the host innate immune system. In this review, we will summarize current knowledge and understanding of insect-borne bunyavirus non-structural protein function(s) in vertebrate, plant and arthropod.

  • Borna disease virus infection in cats

    • Bornaviruses are known to cause neurological disorders in a number of animal species. Avian Bornavirus (ABV) causes proventricular dilatation disease (PDD) in birds and Borna disease virus (BDV) causes Borna disease in horses and sheep. BDV also causes staggering disease in cats, characterised by ataxia, behavioural changes and loss of postural reactions. BDV-infection markers in cats have been reported throughout the world. This review summarizes the current knowledge of Borna disease viruses in cats, including etiological agent, clinical signs, pathogenesis, epidemiology and diagnostics, with comparisons to Bornavirus infections in other species.

  • Challenges and opportunities in the implementation of new meat inspection systems in Europe

    • Background: The traditional meat inspection system is often found to be inefficient. Meat inspection in European countries is in a phase of modernisation to reflect improvements in livestock health and advances in understanding meat safety. The key point of progress is to replace the traditional meat inspection with risk-based meat inspection, including the elements of a meat safety assurance system (MSAS). Scope and approach: Modernisation of meat inspection was launched by the European Food Safety Authority's (EFSA) opinions and recommendations in the period 2011-2013 and consequent amendments to the EU meat inspection legislation in the period 2014-2019. For this study, the EU-funded RIBMINS COST Action conducted a comprehensive survey using an in-depth questionnaire to estimate the level of implementation of new risk-based meat inspection systems in Europe, stakeholders' confidence in the new systems and the main identified obstacles. Key findings and conclusions: The implementation of new meat inspection systems is still ongoing, as they have been fully implemented in just 61%, 42% and 38% of the surveyed countries in the pig, bovine, and poultry sectors, respectively. The main identified obstacles are existing trade agreements with 3rd countries, costs of implementation, inadequate food chain information and resistance from meat inspectors. Improvement of all components of the current meat inspection systems is a prerequisite for further modernisation.

  • Calicivirus Infection in Cats

    • Feline calicivirus (FCV) is a common pathogen in domestic cats that is highly contagious, resistant to many disinfectants and demonstrates a high genetic variability. FCV infection can lead to serious or even fatal diseases. In this review, the European Advisory Board on Cat Diseases (ABCD), a scientifically independent board of experts in feline medicine from 11 European countries, presents the current knowledge of FCV infection and fills gaps with expert opinions. FCV infections are particularly problematic in multicat environments. FCV-infected cats often show painful erosions in the mouth and mild upper respiratory disease and, particularly in kittens, even fatal pneumonia. However, infection can be associated with chronic gingivostomatitis. Rarely, highly virulent FCV variants can induce severe systemic disease with epizootic spread and high mortality. FCV can best be detected by reverse-transcriptase PCR. However, a negative result does not rule out FCV infection and healthy cats can test positive. All cats should be vaccinated against FCV (core vaccine); however, vaccination protects cats from disease but not from infection. Considering the high variability of FCV, changing to different vaccine strain(s) may be of benefit if disease occurs in fully vaccinated cats. Infection-induced immunity is not life-long and does not protect against all strains; therefore, vaccination of cats that have recovered from caliciviral disease is recommended.

  • A Global Systematic Review of Lead (Pb) Exposure and its Health Effects in Wild Mammals

    • Lead (Pb) is a toxic nonessential metal, known mainly for causing poisoning of humans and wild birds. However, little is known about Pb exposure and its associated health effects in wild mammals. We conducted a global systematic literature review to identify peer-reviewed studies published on Pb exposure in wild mammalian species and the health effects they identified. In total, 183 studies, conducted in 35 countries and published over 62 yr (1961-2022), were included in the review. Only 6% (11/183) of the studies were conducted in developing countries. Although 153 mammalian species were studied, most studies focused on species that are easy to access (i.e., hunted species and small mammals that are easy to trap). Therefore, carnivores and scavengers were less frequently studied than herbivores and omnivores. Despite all studies reporting Pb concentrations, only 45 (25%) studies investigated health effects and, of these 45 studies, only 28 (62%) found any health effect in 57 species. All health effects were negative and ranged from subclinical effects to fatality. Methodologies of Pb sampling and quantification and reporting of results varied widely across the studies, making both Pb concentrations and health effects difficult to compare and evaluate. Thus, there is a need for more research on Pb exposure and its health effects on wild mammals, especially as carnivores and scavengers could be used as sentinels for ecosystem health.

  • Structure-Functional Activity Relationship of beta-Glucans From the Perspective of Immunomodulation: A Mini-Review

    • beta-Glucans are a heterogeneous group of glucose polymers with a common structure comprising a main chain of beta-(1,3) and/or beta-(1,4)-glucopyranosyl units, along with side chains with various branches and lengths. beta-Glucans initiate immune responses via immune cells, which become activated by the binding of the polymer to specific receptors. However, beta-glucans from different sources also differ in their structure, conformation, physical properties, binding affinity to receptors, and thus biological functions. The mechanisms behind this are not fully understood. This mini-review provides a comprehensive and up-to-date commentary on the relationship between beta-glucans' structure and function in relation to their use for immunomodulation.

  • Food safety challenges and One Health within Europe

    • This review discusses food safety aspects of importance from a One Health perspective, focusing on Europe. Using examples of food pathogen/food commodity combinations, spread of antimicrobial resistance in the food web and the risk of transmission of zoonotic pathogens in a circular system, it demonstrates how different perspectives are interconnected. The chosen examples all show the complexity of the food system and the necessity of using a One Health approach. Food safety resources should be allocated where they contribute most One Health benefits. Data on occurrence and disease burden and knowledge of source attribution are crucial in assessing costs and benefits of control measures. Future achievements in food safety, public health and welfare will largely be based on how well politicians, researchers, industry, national agencies and other stakeholders manage to collaborate using the One Health approach. It can be concluded that closer cooperation between different disciplines is necessary to avoid silo thinking when addressing important food safety challenges. The importance of this is often mentioned, but more proof of concept is needed by the research community.

    • Arthropods, such as mosquitoes and ticks, are important vectors for different viruses (so called vector-borne viruses), some of which cause a significant number of human and animal deaths every year as well as affect public health worldwide. Dengue virus, yellow fever virus, chikungunya virus, Japanese encephalitis virus, tick-borne encephalitis virus and Zika virus are just a few examples of important vector-borne viruses. The majority of all vector-borne viruses have an RNA genome, which routinely undergo genetic modifications. The changes in the genome, apart from the environmental issues, can also influence the spread of viruses to new habitats and hosts and lead to the emergence of novel viruses, which may become a threat to public health. Therefore, it is important to investigate the viruses circulating in arthropod vectors to understand their diversity, host range and evolutionary history as well as to predict new emerging pathogens. The choice of detection method is important, as most of the methods can only detect viruses that have been previously well described. Viral metagenomics is a useful tool to simultaneously identify all the viruses present in a sample, including novel viruses. This review describes vector-borne viruses, their maintenance and emergence in nature, and detection using viral metagenomics.

    • This review addresses ways to prepare for and to mitigate effects of biohazards on primary production of crops and livestock. These biohazards can be natural or intentional introductions of pathogens, and they can cause major economic damage to farmers, the agricultural industry, society, and international trade. Agroterrorism is the intentional introduction of animal or plant pathogens into agricultural production systems with the intention to cause socioeconomic harm and generate public fear. Although few acts of agroterrorism are reported, the threat of agroterrorism in Europe is real. New concerns about threats arise from the rapid advancements in biotechnology and emerging technologies. FORSA, an analytical framework for risk and vulnerability analysis, was used to review how to prepare for and mitigate the possible effects of natural or intentional biohazards in agricultural production. Analyzing the effects of a biohazard event involves multiple scientific disciplines. A comprehensive analysis of biohazards therefore requires a systems approach. The preparedness and ability to manage events are strengthened by bolstered farm biosecurity, increased monitoring and laboratory capacity, improved inter-agency communication and resource allocation. The focus of this review is on Europe, but the insights gained have worldwide applications. The analytical framework used here is compared to other frameworks. With climate change, Covid-19 and the war in Ukraine, the supply chains are challenged, and we foresee increasing food prices associated with social tensions. Our food supply chain becomes more fragile with more unknowns, thereby increasing the needs for risk and vulnerability analyses, of which FORSA is one example.
