  • Ecology and sociotechnical systems research – motivations for theoretical and methodological integration across fields

    • Currently, we are witnessing a number of global trends that do not promise well for the future. Accelerating climate change, loss of biodiversity, chemical pollution, disappearance of natural forest and degradation of fishing grounds and agricultural lands are just a few of the serious environmental problems that threaten the functional and structural integrity of ecosystems, to an extent that also human societies risk collapse. The scale of human impact is now such that scholars suggest that we live in the Anthropocene. The trends are driven by several linked factors, which are not easily disentangled into manageable specific problems to be solved by specific policies. More than ever, interdisciplinary and transdisciplinary collaborations are needed in order to address these urgent challenges. The objective of this paper is to argue for the importance of research on socio-technical-ecological systems (STES) rather than social-ecological (SES) and sociotechnical systems (STS) separately. Hence, we address researchers in both the social-ecological and sociotechnical fields. We organize the argument around six reasons why “technology” should be integrated into SES studies. We call these reasons: (1) the interface and mediation aspect, (2) ambivalence, (3) the agency aspect, (4) the question of scale, (5) the question of governance and politics, and (6) the question of epistemology and framing. We also highlight potential conceptual conflicts and mistranslations. Our discussion is primarily a theoretical argument, exemplified with empirical examples.Among the conceptual challenges, we note that SES scholars, if they consider technology in their analyses, generally treat it as an exogenous factor or as a passive background element. Similarly, STS scholars tend to neglect ecological dynamics and refer to the ecological domain mainly in terms of inputs and outputs, e.g. natural resources, environmental and health problems caused by human activities. In light of the discussion, we conclude that the importance of collaborating across the two fields goes beyond each field adding pieces together. We argue that integration and translation across these domains will lead to qualitative change in the theoretical and methodological approaches of both fields; and that technology, society and ecology should be given symmetric analytical attention.

  • Is it possible to avoid bad impacts by using good fuel ethanol?

    • Much of the global production of biofuels is considered to be non-sustainable. Brazilian sugarcane ethanol, on the other hand, is normally judged to be “good”. Swedes are anxious only to use fuel ethanol with the best climate characteristics in a life-cycle perspective, and the bulk of ethanol used in Sweden comes from Brazil. The Swedish Environmental Protection Agency has identified some crucial issues which often are left out from discussions. These might be of extra importance for the Swedish ethanol use: - Might Swedish demand for good ethanol indirectly raise the demand for “bad” ethanol, such as US maize ethanol with fossil energy input? Or is it possible to encourage the production of exclusively “good” ethanol by choosing such (certified) ethanol? This depends on how the international market for fuel ethanol works. - To what extent does increased Swedish, or European, demand encourage the long-term supply of ethanol? What supply elasticities are there in Brazil and globally? If increased European use only means that we take hold of a fixed supply, the climate benefit compared to fossil fuels will not occur. The analyses are further complicated by the fact that there might be land-use competition between fuel, feedstuffs and food. When available land becomes more limited, increased production might necessitate breaking new soil, which could lead to emissions of climate-changing gases elsewhere. Consequently it is not only the fuel market itself that needs to be analysed.

  • Comparison of four DNA extraction methods for comprehensive assessment of 16S rRNA bacterial diversity in marine biofilms using high-throughput sequencing

    • High-throughput DNA sequencing technologies are increasingly used for the metagenomic characterization of microbial biodiversity. However, basic issues, such as the choice of an appropriate DNA extraction method, are still not resolved for non-model microbial communities. This study evaluates four commonly used DNA extraction methods for marine periphyton biofilms in terms of DNA yield, efficiency, purity, integrity and resulting 16S rRNA bacterial diversity. Among the tested methods, the Plant DNAzol® Reagent (PlantDNAzol) and the Fast DNATM SPIN Kit for Soil (FastDNA Soil) methods were best suited to extract high quantities of DNA (77 - 130 μg g wet wt-1).     • The antibacterial agent Triclosan (TCS) is a ubiquitous environmental contaminant due to its widespread use. Sensitivity to TCS varies substantially among eu- and pro-karyotic species and its risk for the marine environment remains to be better elucidated. In particular, the effects that TCS causes on marine microbial communities are largely unknown. In this study we therefore used 16S amplicon rDNA sequencing to investigate TCS effects on the bacterial composition in marine periphyton communities that developed under long-term exposure to different TCS concentrations. Exposure to TCS resulted in clear changes in bacterial composition already at concentrations of 1 to 3.16 nM. We conclude that TCS affects the structure of the bacterial part of periphyton communities at concentrations that actually occur in the marine environment. Sensitive taxa, whose abundance decreased significantly with increasing TCS concentrations, include the Rhodobiaceae and Rhodobacteraceae families of Alphaproteobacteria, and unidentified members of the Candidate division Parcubacteria. Tolerant taxa, whose abundance increased significantly with higher TCS concentrations, include the families Erythrobacteraceae (Alphaproteobacteria), Flavobacteriaceae (Bacteroidetes), Bdellovibrionaceae (Deltaproteobacteria), several families of Gammaproteobacteria, and members of the Candidate phylum Gracilibacteria. Our results demonstrate the variability of TCS sensitivity among bacteria, and that TCS can change marine bacterial composition at concentrations that have been detected in the marine environment.

  • Effects of biodiversity and environment-related attitude on perception of urban green space

    • Green space in cities contributes to the quality of life for city dwellers, e.g., by increasing the opportunity for recreation. However, perception of urban green space is influenced by multiple factors. We investigated effects of biodiversity and environment-related attitudes on visual and auditory perceptions of urban green space. Field measurements of biodiversity were conducted in six sites across an urban gradient in Gothenburg, Sweden, and three categories of biodiversity—high, medium, low—were established. Households were sent a survey on aesthetic perception of urban green space, sound perception and the importance of trees and plants for the perception of bird species. Each respondent focused on the site that was located nearby. The environment-related attitudes comprised “Nature-oriented” and “Urban-oriented” persons and were based on participants’ own attitude estimations. It was shown that participants’ “subjective” aesthetic and sound-related perception of urban greenery were in line with the “objectively” measured subdivisions of high, medium and low biodiversity. So also were their estimations of the importance of trees and plants for perception of bird species in urban greenery, although differing only between high and medium/low biodiversity conditions. Persons rating themselves as highly nature-oriented were shown to give higher scores to urban green space aesthetics and to value greenery-related sounds higher, and to attach greater importance to trees and plants in their perception of bird species in urban greenery, than less nature-oriented persons. Highly urban-oriented persons compared to less urban-oriented persons did the same, but only regarding urban greenery-related aesthetics and sounds of nature. We conclude that environment-related attitudes influence perceptions of green space. Moreover, our findings support the idea that biodiversity per se also influences perceptions; people value green space significantly more with high than with low measured biodiversity. Urban planning needs to provide city inhabitants with green spaces that are species-rich, lush, varied and rich with natural sounds.

  • Induced tolerance in situ to chronically PAH exposed ammonium oxidizers

    • Sediment was sampled in the vicinity of a long-term source of Polycyclic Aromatic Hydrocarbons (PAHs) to evaluate whether tolerance can be induced in situ. Total PAH concentrations as well as the bioavailable PAHs were measured, and for nine PAHs the pore water concentration could be calculated. An induced tolerance in the ammonium oxidizing community was detected at the site with highest PAH concentration and tolerance was strongest, although not significantly, correlated to bioavailable alkylated PAHs. In addition, the tolerant microbial community showed a significant lower baseline capability for nitrification with an on average 35% reduction compared to the other sites. Meiofaunal community structure differed between all sites, and the difference was significantly correlated to bioavailable alkylated PAHs and PAH31 concentrations. The results suggest that in order to judge magnitude of long-term effects, the bioavailable fraction is to be preferred, and when possible as estimation of the freely dissolved concentration.

    • Medetomidine is a new antifouling substance, highly effective against barnacles. As part of a thorough ecotoxicological evaluation of medetomidine, its short-term effects on algal and bacterial communities were investigated and environmental concentrations were predicted with the MAMPEC model. Photosynthesis and bacterial protein synthesis for three marine communities, viz. periphyton, epipsammon and plankton were used as effect indicators, and compared with the predicted environmental concentrations (PECs). The plankton community showed a significant decrease in photosynthetic activity of 16% at 2 mg l(-1) of medetomidine, which was the only significant effect observed. PECs were estimated for a harbor, shipping lane and marina environment using three different model scenarios (MAMPEC default, Baltic and OECD scenarios). The highest PEC of 57 ng l(-1), generated for a marina with the Baltic scenario, was at least 10,000-fold lower than the concentration that significantly decreased photosynthetic activity. It is concluded that medetomidine does not cause any acute toxic effects on bacterial protein synthesis and only small acute effects on photosynthesis at high concentrations in marine microbial communities. It is also concluded that the hazard from medetomidine on these processes is low since the effect levels are much lower than the highest PEC.

  • Effect of Geographical Location on the Variation in Products Formed from the Hydrothermal Liquefaction of Ulva intestinalis

    • Hydrothermal liquefaction (HTL) of macroalgae offers a promising route to advanced biofuel production, although the distinct biochemical compositions of different macroalgae species can lead to widely different product yields and compositions. On the basis of this, there is an implicit assumption that there exists a universal optimal feedstock species for a bioenergy-based biorefinery, which could be exploited across a wide region. However, no studies to date have examined the effect of this large geographical variation on a single macroalgae species for biofuel production. In this study, 24 samples of Ulva intestinalis were collected along 1200 km of Swedish coastline and assessed as a feedstock for HTL. Significant variation in composition was observed between samples from Baltic and Atlantic regions, but substantial variation also existed between sites within close proximity. This was reflected in the HTL biocrude oil yields, which varied between 9 and 20% (14-28% dry and ash-free basis) across the sample set. In a number of cases, greater variation was seen for adjacent sites than for sites at opposite ends of the sampling spectrum. Biocrude oil yields in this study also differed substantially from those previously obtained for U. intestinalis from the United Kingdom and Vietnam. Localized environmental conditions affected the HTL product composition significantly, in particular, the elemental distribution within the sample set. The variability observed in this study suggests that no single species will be dominant within a macroalgal biorefinery concept, but rather a species would need to be selected to match the needs of the exact local environment.

  • Metagenomic Sequencing of Marine Periphyton: Taxonomic and Functional Insights into Biofilm Communities

    • Periphyton communities are complex phototrophic, multispecies biofilms that develop on surfaces in aquatic environments. These communities harbor a large diversity of organisms comprising viruses, bacteria, algae, fungi, protozoans and metazoans. However, thus far the total biodiversity of periphyton has not been described. In this study, we use metagenomics to characterize periphyton communities from the marine environment of the Swedish west coast. Although we found approximately ten times more eukaryotic rRNA marker gene sequences compared to prokaryotic, the whole metagenome-based similarity searches showed that bacteria constitute the most abundant phyla in these biofilms. We show that marine periphyton encompass a range of heterotrophic and phototrophic organisms. Heterotrophic bacteria, including the majority of proteobacterial clades and Bacteroidetes, and eukaryotic macro-invertebrates were found to dominate periphyton. The phototrophic groups comprise Cyanobacteria and the alpha-proteobacterial genus Roseobacter, followed by different micro- and macro-algae. We also assess the metabolic pathways that predispose these communities to an attached lifestyle. Functional indicators of the biofilm form of life in periphyton involve genes coding for enzymes that catalyze the production and degradation of extracellular polymeric substances, mainly in the form of complex sugars such as starch and glycogen-like meshes together with chitin. Genes for 278 different transporter proteins were detected in the metagenome, constituting the most abundant protein complexes. Finally, genes encoding enzymes that participate in anaerobic pathways, such as denitrification and methanogenesis, were detected suggesting the presence of anaerobic or low-oxygen micro-zones within the biofilms.

  • Effect of anode material and dispersal limitation on the performance and biofilm community in microbial electrolysis cells

    • In a microbial electrolysis cell (MEC), the oxidization of organic compounds is facilitated by an electrogenic biofilm on the anode surface. The biofilm community composition determines the function of the system. Both deterministic and stochastic factors affect the community, but the relative importance of different factors is poorly understood. Anode material is a deterministic factor as materials with different properties may select for different microorganisms. Ecological drift is a stochastic factor, which is amplified by dispersal limitation between communities. Here, we compared the effects of three anode materials (graphene, carbon cloth, and nickel) with the effect of dispersal limitation on the function and biofilm community assembly. Twelve MECs were operated for 56 days in four hydraulically connected loops and shotgun metagenomic sequencing was used to analyse the microbial community composition on the anode surfaces at the end of the experiment. The anode material was the most important factor affecting the performance of the MECs, explaining 54–80 % of the variance observed in peak current density, total electric charge generation, and start-up lag time, while dispersal limitation explained 10–16 % of the variance. Carbon cloth anodes had the highest current generation and shortest lag time. However, dispersal limitation was the most important factor affecting microbial community structure, explaining 61–98 % of the variance in community diversity, evenness, and the relative abundance of the most abundant taxa, while anode material explained 0–20 % of the variance. The biofilms contained nine Desulfobacterota metagenome-assembled genomes (MAGs), which made up 64–89 % of the communities and were likely responsible for electricity generation in the MECs. Different MAGs dominated in different MECs. Particularly two different genotypes related to Geobacter benzoatilyticus competed for dominance on the anodes and reached relative abundances up to 83 %. The winning genotype was the same in all MECs that were hydraulically connected irrespective of anode material used.
