  • Structure-Function Relationship of Artificial Spider Silk Fibers Produced by Straining Flow Spinning

    • The production of large quantities of artificial spider silk fibers that match the mechanical properties of the native material has turned out to be challenging. Recent advancements in the field make biomimetic spinning approaches an attractive way forward since they allow the spider silk proteins to assemble into the secondary, tertiary, and quaternary structures that are characteristic of the native silk fiber. Straining flow spinning (SFS) is a newly developed and versatile method that allows production under a wide range of processing conditions. Here, we use a recombinant spider silk protein that shows unprecedented water solubility and that is capable of native-like assembly, and we spin it into fibers by the SFS technique. We show that fibers may be spun using different hydrodynamical and chemical conditions and conclude that these spinning conditions affect fiber mechanics. In particular, it was found that the addition of acetonitrile and polyethylene glycol to the collection bath results in fibers with increased beta-sheet content and improved mechanical properties.

  • Cellulose from the green macroalgae Ulva lactuca: isolation, characterization, optotracing, and production of cellulose nanofibrils

    • We report (1) successful extraction and characterization of cellulose from northern hemisphere green macroalgae Ulva lactuca (Ulva fenestrata) collected along the Swedish west coast and cultivated indoors under controlled conditions, followed by (2) its utilization in the production of lignin-free cellulose nanofibrils (CNF). Cellulose was extracted by sequential treatment with ethanol, hydrogen peroxide, sodium hydroxide, and hydrochloric acid, yielding a cellulose-rich insoluble fraction. The extracted cellulose was disintegrated into CNF using a mechanical homogenization process without any further enzymatic pre-treatments. In addition, regenerated cellulose was prepared. XRD characterization of the CNF showed characteristic peaks for the cellulose I allomorph and confirmed that the nanofibrils were semicrystalline with a crystallinity index of 48%. Regenerated cellulose was mostly amorphous with an XRD pattern indicating the presence of the cellulose II allomorph. The cellulose fractions were essentially free from inorganic substances and thermally stable up to around 260 degrees C. Structural mapping with CP-MAS C-13-NMR sustains the cellulose content of CNF and regenerated cellulose, respectively, yet ion chromatography identified the presence of 10-15% xylose in the fractions. Optotracing was used as a novel and non-disruptive tool to selectively assess the polysaccharide composition of the cellulose fractions and produced CNF aiming to shed light on this hitherto non-resolved origin of xylose in Ulva cell wall matter. Fluorescence excitation and emission spectra of a panel of 4 oligothiophenes identified and verified the presence of cellulose and sustain the conclusion that the isolated fractions consist of cellulose intertwined with a small amount of a xylose-containing glucan copolymer.   • Tailoring the effect of antibacterial polyelectrolyte multilayers by choice of cellulosic fiber substrate

    • There is a need for new, antibacterial cellulose-based materials. Antibacterial cellulosic fibers with irreversibly attached polyvinylamine (PVAm) and polyacrylic acid (PAA) in multilayers were developed based on a water-based physical adsorption process. The antibacterial substance is thus prevented from leaching, in contrast to materials containing, for example, silver. It was shown on fibers from different sources that the antibacterial effect against both Escherichia coli and Bacillus subtilis can be tailored. The efficiency correlated with the initial fiber charge, which in turn correlated with the amount of adsorbed PVAm. In the case of highly charged fibers, the antibacterial efficacy was more than 99.9%. A sustainable antibacterial material can be obtained in a simple way based on a water-based process.

    • Studies of biological and artificial membrane systems, such as niosomes, currently rely on the use of fluorescent tags, which can influence the system under investigation. For this reason, the development of label-free, non-invasive detection techniques is of great interest. We demonstrate an open volume label-free millimeter-wave sensing platform based on a coplanar waveguide, developed for identification and characterization of niosome constituents. A design based on a λ/2-line resonator was used and on-wafer measurements of transmission and reflection parameters were performed up to 110 GHz. Our sensor was able to clearly distinguish between common niosome constituents, non-ionic surfactants Tween 20 and Span 80, measuring a resonance shift of 3 GHz between them. The complex permittivities of the molecular compounds have been extracted. Our results indicate insignificant frequency dependence in the investigated frequency range (3 GHz – 110 GHz). Values of permittivity around 3.0 + 0.7i and 2.2 + 0.4i were obtained for Tween 20 and Span 80, respectively.

  • Disruption of the Clock Component Bmal1 in Mice Promotes Cancer Metastasis through the PAI-1-TGF-beta-myoCAF-Dependent Mechanism

    • The circadian clock in animals and humans plays crucial roles in multiple physiological processes. Disruption of circadian homeostasis causes detrimental effects. Here, it is demonstrated that the disruption of the circadian rhythm by genetic deletion of mouse brain and muscle ARNT-like 1 (Bmal1) gene, coding for the key clock transcription factor, augments an exacerbated fibrotic phenotype in various tumors. Accretion of cancer-associated fibroblasts (CAFs), especially the alpha smooth muscle actin positive myoCAFs, accelerates tumor growth rates and metastatic potentials. Mechanistically, deletion of Bmal1 abrogates expression of its transcriptionally targeted plasminogen activator inhibitor-1 (PAI-1). Consequently, decreased levels of PAI-1 in the tumor microenvironment instigate plasmin activation through upregulation of tissue plasminogen activator and urokinase plasminogen activator. The activated plasmin converts latent TGF-beta into its activated form, which potently induces tumor fibrosis and the transition of CAFs into myoCAFs, the latter promoting cancer metastasis. Pharmacological inhibition of the TGF-beta signaling largely ablates the metastatic potentials of colorectal cancer, pancreatic ductal adenocarcinoma, and hepatocellular carcinoma. Together, these data provide novel mechanistic insights into disruption of the circadian clock in tumor growth and metastasis. It is reasonably speculated that normalization of the circadian rhythm in patients provides a novel paradigm for cancer therapy.

    • Recognition of specific antigens expressed in cancer cells is the initial process of cytolytic T cell-mediated cancer killing. However, this process can be affected by other non-cancerous cellular components in the tumor microenvironment. Here, it is shown that interleukin-33 (IL-33)-activated macrophages protect melanoma cells from tumor-infiltrating lymphocyte-mediated killing. Mechanistically, IL-33 markedly upregulates metalloprotease 9 (MMP-9) expression in macrophages, which acts as a sheddase to trim NKG2D, an activating receptor expressed on the surface of natural killer (NK) cells, CD8+ T cells, subsets of CD4+ T cells, iNKT cells, and gamma delta T cells. Further, MMP-9 also cleaves the MHC class I molecule, cell surface antigen-presenting complex molecules, expressed in melanoma cells. Consequently, IL-33-induced macrophage MMP-9 robustly mitigates the tumor killing-effect by T cells. Genetic and pharmacological loss-of-function of MMP-9 sheddase restore T cell-mediated cancer killing. Together, these data provide compelling in vitro and in vivo evidence showing novel mechanisms underlying the IL-33-macrophage-MMP-9 axis-mediated immune tolerance against cancer cells. Targeting each of these signaling components, including IL-33 and MMP-9 provides a new therapeutic paradigm for improving anticancer efficacy by immune therapy.

    • A bioanalytical method for the determination of lumefantrine in 100 μl blood applied onto sampling paper, by solid-phase extraction and liquid chromatography, has been developed and validated. Whatman 31 ET Chr sampling paper was pre-treated with 0.75 M tartaric acid before sampling capillary blood to enable a high recovery of lumefantrine. Lumefantrine was extracted from the sampling paper, then further purified using solid-phase extraction and finally quantified with HPLC. The between-day variation was below 10% over the range 0.4-25 μM. The lower limit of quantification was 0.25 μM in 100 μl capillary blood. No decrease in lumefantrine concentration in dried blood spot is seen after 4 months storage at 22 °C. The method was also evaluated in field samples from patients in Tanzania after treatment with lumefantrine/artemether. Lumefantrine could be estimated accurately enough to assess bioavailability and treatment compliance on day 7 (i.e. 4 days after the last dose) after a standard regimen with the lumefantrine/artemether combination.

    • Bacterial biofilms on medical devices may result in infections with significant societal burden. One drug-free strategy against biofilms is photocatalysis, in which a semiconducting coating is applied on the medical device and irradiated with light to generate reactive oxygen species providing an on-demand disinfection approach. However, most photocatalytic materials are active in the harmful UV range rendering them unsuitable for biomedical applications. Furthermore, the main manufacturing bottleneck today for antibiofilm coatings is their poor durability. To address these challenges, here we produced silver/titanium-suboxide nanoparticles that are photocatalytically active in the visible-light range. Moreover, we directly deposited the nanoparticles as porous coatings on substrates in situ during their aerosol synthesis. To enhance their durability, we infused the fabricated porous coatings with a polymer solution barely covering the photocatalytic particle film, resulting in the formation of polymer nanocomposite coatings. The optimized polymer nanocomposite films exhibit several cycles of triggered, on-demand biofilm eradication activity under short visible light illumination of 15–90 min with no significant intrinsic cytotoxicity to mammalian cells. The developed films can be considered as a suitable coating material for medical devices, such as catheters, ventilators, wound meshes, and others, that may require repeated disinfection during use.

    • Oxidative DNA damage is recognized by 8-oxoguanine (8-oxoG) DNA glycosylase 1 (OGG1), which excises 8-oxoG, leaving a substrate for apurinic endonuclease 1 (APE1) and initiating repair. Here, we describe a small molecule (TH10785) that interacts with the phenylalanine-319 and glycine-42 amino acids of OGG1, increases the enzyme activity 10-fold, and generates a previously undescribed b,d-lyase enzymatic function. TH10785 controls the catalytic activity mediated by a nitrogen base within its molecular structure. In cells, TH10785 increases OGG1 recruitment to and repair of oxidative DNA damage. This alters the repair process, which no longer requires APE1 but instead is dependent on polynucleotide kinase phosphatase (PNKP1) activity. The increased repair of oxidative DNA lesions with a small molecule may have therapeutic applications in various diseases and aging. © 2022 The Authors, some rights reserved; exclusive licensee American Association for the Advancement of Science.     • Blinking and time correlation between fluorescences of neighboring negatively charged CdSe/CdS and CdSe/ZnS colloidal quantum dots have been studied experimentally. A tendency of synchronous blinking, that is, a bunching effect, is clearly observed from two neighboring QDs with a spatial separation up to 1.1 mu m. We believe that our observations will help to better understand the mechanisms for the blinking.
