  • Considering the importance of user profiles in interface design

    • User profile is a popular term widely employed during product design processes by industrial companies. Such a profile is normally intended to represent real users of a product. The ultimate purpose of a user profile is actually to help designers to recognize or learn about the real user by presenting them with a description of a real user’s attributes, for instance; the user’s gender, age, educational level, attitude, technical needs and skill level. The aim of this chapter is to provide information on the current knowledge and research about user profile issues, as well as to emphasize the importance of considering these issues in interface design. In this chapter, we mainly focus on how users’ difference in expertise affects their performance or activity in various interaction contexts. Considering the complex interaction situations in practice, novice and expert users’ interactions with medical user interfaces of different technical complexity will be analyzed as examples: one focuses on novice and expert users’ difference when interacting with simple medical interfaces, and the other focuses on differences when interacting with complex medical interfaces. Four issues will be analyzed and discussed: (1) how novice and expert users differ in terms of performance during the interaction; (2) how novice and expert users differ in the perspective of cognitive mental models during the interaction; (3) how novice and expert users should be defined in practice; and (4) what are the main differences between novice and expert users’ implications for interface design. Besides describing the effect of users’ expertise difference during the interface design process, we will also pinpoint some potential problems for the research on interface design, as well as some future challenges that academic researchers and industrial engineers should face in practice.

  • Optical characterization of colloidal CdSe quantum dots in endothelial progenitor cells.

    • We have quantitatively analyzed the confocal spectra of colloidal quantum dots (QDs) in rat endothelial progenitor cells (EPCs) by using Leica TCS SP5 Confocal Microscopy System. Comparison of the confocal spectra of QDs located inside and outside EPCs revealed that the interaction between the QDs and EPCs effectively reduces the radius of the exciton confinement inside the QDs so that the excitonic energy increases and the QD fluorescence peak blueshifts. Furthermore, the EPC environment surrounding the QDs shields the QDs so that the excitation of the QDs inside the cells is relatively weak, whereas the QDs outside the cells can be highly excited. At high excitations, the occupation of the ground excitonic state in the QD outside the cells becomes saturated and high-energy states excited, resulting in a large relaxation energy and a broad fluorescence peak. This permits, in concept, to use QD biomarkers to monitor EPCs by characterizing QD fluorescence spectra.

    • In order to study the role of surface ligands in determining optical properties of colloidal quantum dots (QDs), we have selectively fabricated and studied CdSe/CdS core-shell QDs with strongly confined electron and hole states attached with commonly used surface ligands. Optical properties, viz. absorption and fluorescence of these QDs, are characterized from which salient changes have been observed for different ligand substitutions which, through theoretical analysis, can be associated with electronic structure properties of the QD-ligand composite systems, in particular localization of wave functions of electrons and holes in the QDs and the band matching of the HOMO-LUMO gap of the ligands. The findings can be utilized to facilitate the understanding and optimization of properties of QD biomarkers with functionalizing surface ligands for targeting cellular objects.

  • High-temperature superconducting magnetometers for on-scalp MEG

    • In the growing field of on-scalp magnetoencephalography (MEG), brain activity is studied by non-invasively mapping the magnetic fields generated by neuronal currents with sensors that are flexibly placed in close proximity to the subject's head. This thesis focuses on high-temperature superconducting magnetometers made from YBa2Cu3Ox-7 (YBCO), which enables a reduction in the sensor-to-room temperature standoff distance from roughly 2 cm (for conventional MEG systems) down to 1 mm. Because of the higher neuromagnetic signal magnitudes available to on-scalp sensors, simulations predict that even a relatively low-sensitivity (higher noise) full-head on-scalp MEG system can extract more information about brain activity than conventional systems. In the first part of this thesis, the development of high critical temperature (high-Tc) superconducting quantum interference device (SQUID) magnetometers for a 7-channel on-scalp MEG system is described. The sensors are single layer magnetometers with a directly coupled pickup loop made on 10 mm × 10 mm substrates using bicrystal grain boundary Josephson junctions. We found that the kinetic inductance strongly varies with film quality and temperature. Determination of all SQUID parameters by combining measurements and inductance simulations led to excellent agreement between experimental results and theoretical predictions. This allowed us to perform an in-depth magnetometer optimization. The best magnetometers achieve a magnetic field noise level of 44 fT/√Hz at 78 K. Fabricated test SQUIDs provide evidence that noise levels below 30 fT/√Hz are possible for high quality junctions with fairly low critical currents and in combination with the optimized pickup loop design. Different feedback methods for operation in a densely-packed on-scalp MEG system were also investigated. Direct injection of current into the SQUID loop was identified as the best on-chip feedback method with feedback flux crosstalk below 0.5%. By reducing the operation temperature, the noise level can be further reduced, however, the effective area also decreases because of the decreasing kinetic inductance contribution. We present a method that allows for one-time sensor calibration independent of temperature. In the second part, the design, operation, and performance of the constructed 7-channel on-scalp MEG system based on the fabricated magnetometers is presented. With a dense (2 mm edge-to-edge) hexagonal head-aligned array, the system achieves a small sensor-to-head standoff distance of 1-3 mm and dense spatial sampling. The magnetic field noise levels are 50-130 fT/√Hz and the sensor-to-sensor feedback flux crosstalk is below 0.6%. MEG measurements with the system demonstrate the feasibility of the approach and indicate that our on-scalp MEG system allows retrieval of information unavailable to conventional MEG. In the third part, two alternative magnetometer types are studied for the next generation system. The first alternative is magnetometers based on Dayem bridge junctions instead of bicrystal grain boundary junctions. With a magnetometer based on the novel grooved Dayem bridge junctions, a magnetic field noise level of 63 fT/√Hz could be achieved, which shows that Dayem bridge junctions are starting to become a viable option for single layer magnetometers. The second alternative are high-Tc SQUID magnetometers with an inductively coupled flux transformer. The best device with bicrystal grain boundary junctions reaches a magnetic field noise level below 11 fT/√Hz and outperforms the best single layer device for frequencies above 20 Hz. In the last part, the potential of kinetic inductance magnetometers (KIMs) is investigated. We demonstrate the first high-Tc KIMs, which can be operated in fields of 9-28 µT and achieve a noise level of 4 pT/√Hz at 10 kHz.

    • Glycoprotein C (gC) mediates the attachment of herpes simplex virus type 1 (HSV-1) to susceptible host cells by interacting with glycosaminoglycans (GAGs) on the cell surface. gC contains a mucin-like region located near the GAG-binding site, which may affect the binding activity. Here, we address this issue by studying an HSV-1 mutant lacking the mucin- like domain in gC and the corresponding purified mutant protein (gCΔmuc), in cell culture and GAG-binding assays, respectively. The mutant virus exhibited two functional alterations as compared to native HSV-1, i.e. decreased sensitivity to GAG-based inhibitors of virus attachment to cells, and reduced release of viral particles from the surface of infected cells. Kinetic and equilibrium binding characteristics of purified gC were assessed using surface plasmon resonance-based sensing together with a surface platform consisting of end-on immobilized GAGs. Both native gC and gCΔmuc bound via the expected binding region to chondroitin sulfate and sulfated hyaluronan but not to the non-sulfated hyaluronan, confirming binding specificity. In contrast to native gC, gCΔmuc exhibited a decreased affinity for GAGs and a slower dissociation, indicating that once formed, the gCΔmuc-GAG complex is more stable. It was also found that a larger number of gCΔmuc bound to a single GAG chain, compared to native gC. Taken together, our data suggest that the mucin-like region of HSV-1 gC is involved in the modulation of the GAG-binding activity, a feature of importance both for unrestricted virus entry into the cells and release of newly produced viral particles from infected cells.

  • Automated Malaria Parasite Detection

    • A system for malaria parasite detection in thin blood-smears is presented. Sample slides prepared with standard methods are accepted. A low-cost TV-camera mounted on an ordinary microscope with a computer controlled stage is used as a picture sensor. Frames, digitized in windows of 64 x 64 pixels are fed into a special purpose picture processor at normal frame rate (25 frames/sec). In the picture processor measurements are made on the images at high speed. The classification problem is split into different levels each having different characteristics such as different sampling density. Four classes, three of which are different types of malaria parasites, are recognized. As a whole the classification is best labelled as a sequential pattern recognition procedure.In its preliminary version the system has been run at a speed comparable to that of a human operator, that is l 500 cells per minute. A test on 80 000 cells gave 25 false negatives out of 283 parasites (9%) and 41 false positives (0.05%).

    • Fabrication of multifunctional ultrasound contrast agents (UCAs) has been recently addressed by several research groups. A versatile strategy for the synthesis of UCA precursors in the form of biodegradable vesicles with a biocompatible crosslinked polymer shell is described. Upon ultrasound irradiation, acoustic droplet vaporization transforms such particles into microbubbles behaving as UCAs. This proof of concept entails the features of a potential theranostic microdevice.

    • Optical mapping (OM) has been used in microbiology for the past 20 years, initially as a technique to facilitate DNA sequence-based studies; however, with decreases in DNA sequencing costs and increases in sequence output from automated sequencing platforms, OM has grown into an important auxiliary tool for genome assembly and comparison. Currently, there are a number of new and exciting applications for OM in the field of microbiology, including investigation of disease outbreaks, identification of specific genes of clinical and/or epidemiological relevance, and the possibility of single-cell analysis when combined with cell-sorting approaches. In addition, designing lab-on-a-chip systems based on OM is now feasible and will allow the integrated and automated microbiological analysis of biological fluids. Here, we review the basic technology of OM, detail the current state of the art of the field, and look ahead to possible future developments in OM technology for microbiological applications.

    • Analytical tools for quantitative measurements of glutamate, the principal excitatory neurotransmitter in the brain, are lacking. Here, we introduce a new enzyme-based amperometric sensor technique for the counting of glutamate molecules stored inside single synaptic vesicles. In this method, an ultra-fast enzyme-based glutamate sensor is placed into a solution of isolated synaptic vesicles, which stochastically rupture at the sensor surface in a potential-dependent manner at a constant negative potential. The continuous amperometric signals are sampled at high speed (10 kHz) to record sub-millisecond spikes, which represent glutamate release from single vesicles that burst open. Glutamate quantification is achieved by a calibration curve that is based on measurements of glutamate release from vesicles pre-filled with various glutamate concentrations. Our measurements show that an isolated single synaptic vesicle encapsulates about 8000 glutamate molecules and is comparable to the measured exocytotic quantal glutamate release in amperometric glutamate sensing in the nucleus accumbens of mouse brain tissue. Hence, this new methodology introduces the means to quantify ultra-small amounts of glutamate and to study synaptic vesicle physiology, pathogenesis, and drug treatments for neuronal disorders where glutamate is involved.
