  • This study was conducted to investigate the complex interactions between oviducts and cryopreserved spermatozoa. Herein we report the dynamic changes in bull sperm functions during in vitro incubation with bovine estrus and luteal oviductal fluid. Frozen-thawed bull spermatozoa was incubated either in non-capacitating medium, capacitating medium, non-capacitating medium containing 20% v/v estrus oviductal fluid or non-capacitating medium containing 20% v/v luteal oviductal fluid for 6 h at 38 degrees C under 5% CO2. At hourly interval spermatozoa were evaluated for kinematics, tyrosine phosphorylation and acrosome reaction. The sperm velocity parameters were higher (P < 0.05) in capacitating medium compared to the other treatments. At 4 and 5 h of incubation, the proportion of live tyrosine phosphorylated spermatozoa was higher (P < 0.05) in estrus oviductal fluid compared to all other treatments. From 4 to 6 h of incubation the proportion of live acrosome reacted spermatozoa was higher (P < 0.05) in estrus oviductal fluid compared to the other treatments. We conclude that estrus oviductal fluid induced tyrosine phosphorylation and acrosome reaction in a higher proportion of frozen-thawed bull spermatozoa compared to luteal oviductal fluid, although sperm kinematics were not significantly influenced by oviductal during incubation.
