  • Managing an altered social context—Patients experiences of staying away from home while undergoing proton beam therapy

    • Aim: To illuminate the experience of an altered social context for patients with primary brain tumours living away from home while undergoing proton beam therapy. Design: A descriptive, qualitative cross-sectional interview study. Methods: Nineteen patients were interviewed between December 2015–August 2016, either during (N = 7) or before and after (N = 12) their proton beam therapy. A hermeneutical analysis was performed. Results: Participants made adjustments to achieve control and well-being during the treatment period. The analysis also revealed two interrelated patterns that helped participants adjust: being part of the family from a distance and seeking affinity. Conclusion: It is important that patients receiving treatment far from home find a way to remain a part of their family and find affinity in the altered social context. Health professionals can prepare patients for the treatment period and can implement interventions to promote well-being for both patients and their relatives. © 2020 The Authors.

  • Embracing First-Person Perspectives in Soma-Based Design

    • A set of prominent designers embarked on a research journey to explore aesthetics in movement-based design. Here we unpack one of the design sensitivities unique to our practice: a strong first person perspective-where the movements, somatics and aesthetic sensibilities of the designer, design researcher and user are at the forefront. We present an annotated portfolio of design exemplars and a brief introduction to some of the design methods and theory we use, together substantiating and explaining the first-person perspective. At the same time, we show how this felt dimension, despite its subjective nature, is what provides rigor and structure to our design research. Our aim is to assist researchers in soma-based design and designers wanting to consider the multiple facets when designing for the aesthetics of movement. The applications span a large field of designs, including slow introspective, contemplative interactions, arts, dance, health applications, games, work applications and many others.

  • Formate overflow drives toxic folate trapping in MTHFD1 inhibited cancer cells

    • Cancer cells fuel their increased need for nucleotide supply by upregulating one-carbon (1C) metabolism, including the enzymes methylenetetrahydrofolate dehydrogenase–cyclohydrolase 1 and 2 (MTHFD1 and MTHFD2). TH9619 is a potent inhibitor of dehydrogenase and cyclohydrolase activities in both MTHFD1 and MTHFD2, and selectively kills cancer cells. Here, we reveal that, in cells, TH9619 targets nuclear MTHFD2 but does not inhibit mitochondrial MTHFD2. Hence, overflow of formate from mitochondria continues in the presence of TH9619. TH9619 inhibits the activity of MTHFD1 occurring downstream of mitochondrial formate release, leading to the accumulation of 10-formyl-tetrahydrofolate, which we term a ‘folate trap’. This results in thymidylate depletion and death of MTHFD2-expressing cancer cells. This previously uncharacterized folate trapping mechanism is exacerbated by physiological hypoxanthine levels that block the de novo purine synthesis pathway, and additionally prevent 10-formyl-tetrahydrofolate consumption for purine synthesis. The folate trapping mechanism described here for TH9619 differs from other MTHFD1/2 inhibitors and antifolates. Thus, our findings uncover an approach to attack cancer and reveal a regulatory mechanism in 1C metabolism.

    • Background: Skin reaction is a common side-effect of radiotherapy and often only assessed as clinician-reported outcome (CRO). The aim was to examine and compare patient-reported outcome (PRO) of skin reactions with CRO for signs of acute skin reactions for patients with primary brain tumour receiving proton beam radiotherapy (PBT). A further aim was to explore patients’ experiences of the skin reactions. Methods: Acute skin reactions were assessed one week after start of treatment, mid-treatment and end of treatment among 253 patients with primary brain tumour who underwent PBT. PRO skin reactions were assessed with the RSAS and CRO according to the RTOG scale. Fleiss’ kappa was performed to measure the inter-rater agreement of the assessments of skin reactions. Results: The results showed a discrepancy between PRO and CRO acute skin reactions. Radiation dose was associated with increased skin reactions, but no correlations were seen for age, gender, education, occupation, other treatment or smoking. There was a poor agreement between patients and clinicians (κ = −0.016) one week after the start of PBT, poor (κ = −0.045) to (κ = 0.396) moderate agreement at mid treatment and poor (κ = −0.010) to (κ = 0.296) moderate agreement at end of treatment. Generally, patients’ symptom distress toward skin reactions was low at all time points. Conclusion: The poor agreement between PRO and CRO shows that the patient needs to be involved in assessments of skin reactions for a more complete understanding of skin reactions due to PBT. This may also improve patient experience regarding involvement in their own care. © 2021 The Author(s)

    • This case study analyzes five Swedish printing houses' pricing with respect to their investments in new printing technology. The new printing technology made it possible for the printing houses to market new products and services to meet the demand for shorter delivery times and full service solutions. Although this demand was apparent, the printing houses' opportunities to capitalize on their investments depended on the characteristics of the market segment that they served. Findings indicate that the new printing technology made it possible to change prices when the new services reduced delivery time and costs, and when there were substantial differences between the new services and available substitutes. Thus, customers accepted new pricing when the utilization of the new technology resulted in financial gains and time reductions.

  • Stove performance and emission characteristics in residential wood log and pellet combustion : Part 1: Pellet stoves

    • Stove performance, characteristics and quantities of gaseous and particulate emissions were determined for two different pellet stoves, varying fuel load, pellet diameter and chimney draught. This approach aimed at covering variations in emissions from stoves in use today. The extensive measurement campaign included CO, NOx, organic gaseous carbon, volatile organic compounds (VOCs), polycyclic aromatic hydrocarbons (PAHs), total particulate matter (PMtot) as well as particle mass and number concentrations, size distributions and inorganic composition. At high load, most emissions were similar. For stove B, operating at high residual oxygen and solely with primary air, the emissions of PMtot and particle numbers were higher while the particles were smaller. Lowering the fuel load, the emissions of CO and hydrocarbons increased dramatically for stove A, which operated continuously also at lower fuel loads. On the other hand for stove B, which had intermittent operation at lower fuel loads, the emissions of hydrocarbons increased only slightly lowering the fuel load, while CO emissions increased sharply, due to high emissions at the end of the combustion cycle. Beside methane, dominating VOCs were ethene, acetylene and benzene and the emissions of VOC varied in the range 1.1-47 mg/MJfuel. PAH emissions (2-340 µg/MJfuel) were generally dominated by phenantrene, fluoranthene and pyrene. PMtot (15-45 mg/MJfuel) were in all cases dominated by fine particles with mass median diameters in the range 100-200 nm, peak mobility diameters of 50-85 nm and number concentrations in the range 4×1013- 3×1014 particles/MJfuel. During high load conditions the particulate matter was totally dominated by inorganic particles at 15-25 mg/MJfuel consisting of potassium, sodium, sulfur and chlorine, in the form of K2SO4, K3Na(SO4)2 and KCl. The study shows that differences in operation and modulation principles for the tested pellet stoves, relevant for appliances in use today, will affect the performance and emissions significantly, although with lower scattering in the present study compared to compiled literature data.

    • The Masonite Flexible Building (MFB) system is a complete timber building system for commercial and residential multi-storey houses. The system is subdivided into two market variants; XL and Light. The XL version is for tall and large buildings with long floor spans while the Light version is adapted for smaller buildings with lower loads. Though differing in technical performance, the functional criteria arc the same for both variants. The MFB system uses prefabricated wall, floor and roof elements which are delivered in flat packages and erected on the construction site. The MFB system might be classified as a panel construction, where the load-carrying structure consists of composite light-weight timber I-beams mechanically integrated with a composite laminated wood panel called PlyBoard™. The I-beams and the panel form a strong and rigid carcass for wall and floor elements, making the system well suited for high rise construction. A key feature of the MFB system is the connection technique which enables swift erection of the system units on site. The plyboard panels are provided with a continuous slot along the periphery. The slot is used as a general connection interface for the joining of the wall elements. The floor elements are suspended and hooked onto the bearing walls using sheet steel hangers, allowing swift assembling of the floor deck and enabling direct vertical wall-to-wall load transfer parallel to grain. The paper presents the construction principles, system components and units, erection technique and functional and architectural aspects of the Masonite Building System.

    • This paper presents an experimental and analytical study of a steel plate connection for joining walls in the Masonite Flexible Building (MFB) system. These connections are used partly for splicing the wall elements and partly for tying down uplifting forces and resisting horizontal shear forces in stabilizing shear walls. The steel plates are inserted in a perimeter slot in the plyboard panel (a composite laminated wood panel) and fixed mechanically with screw fasteners. The load-bearing capacity of the slotted-in steel plate connections are determined experimentally and derived analytically for different failure modes. The test results show ductile post-peak load-slip characteristics, indicating that a plastic design method for shear walls can be applied to calculate the horizontal load-bearing capacity. The slotted-in steel plate connection concept can also be used for joining shear walls to transverse walls for tying down purposes in order to simplify the stabilization system of the building. The use of transverse walls for resisting uplifting forces introduces a three-dimensional behaviour of the wall junction and a more effective load transfer.

    • The authors present an experimental study of a suspended floor element connection (sheet steel hangers) employed in the Masonite Flexible Building (MFB) system. The hangers are mounted with screws and are pre-attached to the floor elements at manufacturing. This arrangement makes the design of the hanger critical with respect to safety and load transfer redundancy, since the screws transfer all the loads, both withdrawal and shear forces can act simultaneously. Tests have been carried out to examine the structural behaviour of the hanger. The two most critical load cases, vertical floor load and horizontal wind suction load, and three different screw joint configurations were investigated. The results indicate that the vertical distance between the screw joint and the upper edge of the rim beam should be increased and that withdrawal forces on the screws should be kept as low as possible. Some suggestions for improving the present design are given and a modified design is proposed to enhance the load-bearing capacity and to improve the overall safety and redundancy.

  • Impact of xylan on field productivity and wood saccharification properties in aspen

    • Xylan that comprises roughly 25% of hardwood biomass is undesirable in biorefinery applications involving saccharification and fermentation. Efforts to reduce xylan levels have therefore been made in many species, usually resulting in improved saccharification. However, such modified plants have not yet been tested under field conditions. Here we evaluate the field performance of transgenic hybrid aspen lines with reduced xylan levels and assess their usefulness as short-rotation feedstocks for biorefineries. Three types of transgenic lines were tested in four-year field tests with RNAi constructs targeting either Populus GT43 clades B and C (GT43BC) corresponding to Arabidopsis clades IRX9 and IRX14, respectively, involved in xylan backbone biosynthesis, GATL1.1 corresponding to AtGALT1 involved in xylan reducing end sequence biosynthesis, or ASPR1 encoding an atypical aspartate protease. Their productivity, wood quality traits, and saccharification efficiency were analyzed. The only lines differing significantly from the wild type with respect to growth and biotic stress resistance were the ASPR1 lines, whose stems were roughly 10% shorter and narrower and leaves showed increased arthropod damage. GT43BC lines exhibited no growth advantage in the field despite their superior growth in greenhouse experiments. Wood from the ASPR1 and GT43BC lines had slightly reduced density due to thinner cell walls and, in the case of ASPR1, larger cell diameters. The xylan was less extractable by alkali but more hydrolysable by acid, had increased glucuronosylation, and its content was reduced in all three types of transgenic lines. The hemicellulose size distribution in the GALT1.1 and ASPR1 lines was skewed towards higher molecular mass compared to the wild type. These results provide experimental evidence that GATL1.1 functions in xylan biosynthesis and suggest that ASPR1 may regulate this process. In saccharification without pretreatment, lines of all three constructs provided 8-11% higher average glucose yields than wild-type plants. In saccharification with acid pretreatment, the GT43BC construct provided a 10% yield increase on average. The best transgenic lines of each construct are thus predicted to modestly outperform the wild type in terms of glucose yields per hectare. The field evaluation of transgenic xylan-reduced aspen represents an important step towards more productive feedstocks for biorefineries.
