(författare) Tyr-716 in the platelet-derived growth factor beta-receptor kinase insertis involved in GRB2 binding and Ras activation 1994 Ingår i: Molecular and Cellular Biology. - : American Society for Microbiology. - 0270-7306 .- 1098-5549. ; 14:10, s. 6715-6726 Tidskriftsartikel (refereegranskat)abstract Ligand stimulation of the platelet-derived growth factor (PDGF) beta-receptor leads to activation of its intrinsic tyrosine kinase and autophosphorylation of the intracellular part of the receptor. The autophosphorylated tyrosine residues mediate interactions with downstream signal transduction molecules and thereby initiate different signalling pathways. A pathway leading to activation of the GTP-binding protein Ras involves the adaptor molecule GRB2. Here we show that Tyr-716, a novel autophosphorylation site in the PDGF beta-receptor kinase insert, mediates direct binding of GRB2 in vitro and in vivo. In a panel of mutant PDGF beta-receptors, in which Tyr-716 and the previously known autophosphorylation sites were individually mutated, only PDGFR beta Y716F failed to bind GRB2. Furthermore, a synthetic phosphorylated peptide containing Tyr-716 bound GRB2, and this peptide specifically interrupted the interaction between GRB2 and the wild-type receptor. In addition, the Y716(P) peptide significantly decreased the amount of GTP bound to Ras in response to PDGF in permeabilized fibroblasts as well as in porcine aortic endothelial cells expressing transfected PDGF beta-receptors. The mutant PDGFR beta Y716F still mediated activation of mitogen-activated protein kinases and an increased DNA synthesis in response to PDGF, indicating that multiple signal transduction pathways transduce mitogenic signals from the activated PDGF beta-receptor.

A fold line concept is used to evaluate the dependence of these states on an added variable, describing a disturbing load case or a disturbed geometry. The concept describes the local behaviour for small disturbances, but can also be used for parameter dependence analyses, e.g. in connection with optimization algorithms. Two different augmentations of the equilibrium relations are discussed; they describe the criticality of a solution state in different ways. Numerical adoption for a general equilibrium path following algorithm is discussed. A postponed factorization method for solution of the augmented sets of equations is proposed. Two simple examples are used to show the properties and the possibilities of the fold line concept. It is concluded that the suggested numerical procedure can give a better description of critical structural behaviour, especially with respect to imperfections in the structure and idealizations in the model. © 1994.

In this paper, a signal model is developed that, apart from the TOF, accounts for an unknown, linear frequency dependent distortion as well as for additive noise. We derive a TOF estimator for this model based on the criteria of maximum likelihood. The resulting receiver can be seen as an extension or generalization of the well known cross-correlation, or "matched filter", estimator described, e.g., by Nilsson. The novel receiver is found to be more robust against unknown pulse shape distortion than the cross-correlation estimator, giving less biased TOF estimates. Also, bias versus noise sensitivity can be controlled by proper model order selection

Previously unobserved inner valence states are revealed in the direct photoemission and are compared with resonant Auger results. The interpretation and assignment of the different spectral features to different main final state configurations are based on energy, symmetry and intensity arguments, as well as comparisons with previous results.
