Application of real time RT-PCR for the detection of type 1 and type 3 of bovine viral diarrhea virus (BVDV) in biological material

- The aim of the study was to use real time RT-PCR for the detection of genetic material of bovine viral diarrhea virus (BVDV) type 1 and type 3 in serum and milk samples. Material tested included the fetal calf serum used for cell culture, serum samples from healthy calves and from calves experimentally inoculated with BVDV type 1 and type 3, and milk samples (pasteurized and treated with ultra high temperature). Sensitivity of the test was 200 copies of RNA per reaction (10(5) viral RNA copies per ml) for both types of BVDV, using dedicated primers and standards. Detection limit was 102 tissue culture infectious dose 50 (TCID(50)) and 1 TCID(50) for type 1 and type 3, respectively. Diagnostic specificity of the method was 100%. Out of 10 samples of milk, 3 were positive for BVDV type I, while none was positive for type 3. On the other hand, BVDV type 3 was found in 6 out of 10 samples of fetal calf serum. Real Time RT-PCR for BVDV type 1 and type 3 proved to be a highly sensitive and highly specific technique, enabling the detection of viral genetic material in various samples, even when its detection by virus isolation or antigen ELISA tests is impossible because of virus inactivation by such processes as high temperature, gamma irradiation, or the presence of virus neutralizing antibodies.

- Gdzie leżą granice między tym co ludzkie a światem zwierzęcym? Jaką rolę odgrywa w tym kontekście dźwięk i skomponowany pejzaż dźwiękowy? Czym jest pejzaż dźwiękowy jako forma stworzona z myślą o zwierzęciu jako odbiorcy? W tym eseju punktem wyjścia do próby zaadresowania powyższych pytań jest urządzenie o tajemniczo brzmiącej nazwie UOZ1, czyli Urządzenie Ochrony Zwierząt. Jest to system głośników rozmieszczanych wzdłuż torów kolejowych w celu generowania bariery dźwiękowej odstraszającej zwierzęta leśne. Chroniąc je przed kolizją z nadjeżdżającym pociągiem, ten graniczny pejzaż dźwiękowy jednocześnie zabezpiecza infrastrukturę i interes człowieka. Dźwięk staje się medium jednocześnie uwypuklającym granice między światem ludzkim i zwierzęcym, jak i je rozmywającym. Projekt jest również powodem do dyskusji na temat działalności zmarłej 14 lat temu profesor Simony Kossak. To jej badania i lata spędzone w Puszczy Białowieskiej, gdzie poświęciła się pracy nad zrozumieniem psychologii dzikich zwierząt, przyczyniły się do opracowania tzw. „atrapy bodźców kluczowych”, czyli sekwencji alarmująco-informacyjnych sygnałów dźwiękowych inspirowanych naturą i wykorzystanych w UOZ1. Ten wątek działalności prof. Kossak pozwala nam uznać jej pracę jako nie tylko pionierską w dziedzinie akustyki ekologicznej w tej części świata, ale również sugeruje konieczność przemyślenia koncepcji pejzażu dźwiękowego w kontekście rosnącej potrzeby kontestacji idei granicy między kulturą człowieka a naturą.

- This paper aims to reflect upon the future direction of development in post-PGR (State Agricultural Farms) estates in Poland. Using the estate Chotel (gmina Izbica Kujawska, Kujawsko-Pomorskie voivodship) as a case study, we analyze the current human resources as represented by the local youth. Our point of departure is the assumption that youths, as a social category, will in the nearest future influence the structure of human resources, which in turn will determine both the pace and the direction of change in rural areas. Given that post-PGR estates are considered some of the most problematic settlement forms with respect to rural planning, and given that their adult residents are known to exhibit loose social bonds, intensified enmity and lack of initiative for co-operation, a number of important questions arises. Firstly, what are the specific human resources of youths in post-PGR estates? Secondly, how do these resources differ from those of their parents? Thirdly, do these resources give hope for future melioration of socio-economic problems inherent of post-PGR estates? The conducted analysis is prognosticating – a quality, which otherwise is extremely difficult to obtain in the context of the studied estates.
