  • Role of the Vibrio cholerae Matrix Protein Bap1 in Cross-Resistance to Antimicrobial Peptides

    • Outer membrane vesicles (OMVs) that are released from Gram-negative pathogenic bacteria can serve as vehicles for the translocation of effectors involved in infectious processes. In this study we have investigated the role of OMVs of the Vibrio cholerae O1 El Tor A1552 strain in resistance to antimicrobial peptides (AMPs). To assess this potential role, we grew V. cholerae with sub-lethal concentrations of Polymyxin B (PmB) or the AMP LL-37 and analyzed the OMVs produced and their effects on AMP resistance. Our results show that growing V. cholerae in the presence of AMPs modifies the protein content of the OMVs. In the presence of PmB, bacteria release OMVs that are larger in size and contain a biofilm-associated extracellular matrix protein (Bap1). We demonstrated that Bap1 binds to the OmpT porin on the OMVs through the LDV domain of OmpT. In addition, OMVs from cultures incubated in presence of PmB also provide better protection for V. cholerae against LL-37 compared to OMVs from V. cholerae cultures grown without AMPs or in presence of LL-37. Using a bap1 mutant we showed that cross-resistance between PmB and LL-37 involved the Bap1 protein, whereby Bap1 on OMVs traps LL-37 with no subsequent degradation of the AMP.

    • PurposeThe purpose of this paper is to present an experimental study which aims at assessing the potentially misleading effect of graphic elements on food packaging. The authors call these elements potentially misleading elements (PMEs) as they can give customers false expectations. They are either highlighted numerical information (30 per cent fibre, 8 per cent fat, 100 per cent natural […]) or pictorial information with no relation to the product (e.g. images of happy people).Design/methodology/approachIn a combined decision task monitored by eye-tracking and a subsequence survey, the authors tested the impact of PMEs on common products. Combining different pairs of products, where one product had a PME, whereas the other did not, the authors could evaluate if preference correlated with the presence of a PME.FindingsThe authors found both types of PMEs to have analogous effects on participants’ preferences and correlate with participants’ visual attention. The authors also found evidence for a positive influence on a later explicit justification for the specific choice.Research limitations/implicationsThis study was conducted in a lab environment and solely related to health-related decisions. The authors still need to know if these findings are transferable to real in-store decisions and other needs such as high quality or low price. This calls for further research.Practical implicationsThe topic is important for food companies, and it might become a priority in managing brand equity, combining consumer preferences, loyalty and communicative fairness.Originality/valueUsing eye-tracking and retrospective interviews brings new insights to consumer’s decision-making and how misleading potentially occurs.

  • Garbage collecting reactive real-time systems

    • As real-time systems become more complex, the need for more sophisticated runtime kernel features arises. One such feature that substantially lessens the burden of the programmer is automatic memory management, or garbage collection. However, incorporating garbage collection in a real-time kernel is not an easy task. One needs to guarantee, not only that sufficient memory will be reclaimed in order to avoid out of memory errors, but also that the timing properties of the systems real-time tasks are unaffected. The first step towards such a garbage collector is to define the algorithm in a manageable way. It has to be made incremental in such way that induced pause times are small and bounded (preferably constant). The algorithm should not only be correct, but also provably useful. That is, in order to guarantee that sufficient memory is reclaimed each time the garbage collector is invoked, one need to define some measure of usefulness. Furthermore, the garbage collector must also be guaranteed to be schedulable in the system. That is, even though the collector is correct and proved useful, it still has to be able to do its work within the system. In this thesis, we present a model of an incremental copying garbage collector based on process terms in a labeled transition system. Each kind of garbage collector step is captured as an internal transition and each kind of external heap access (read, write, and allocate) is captured as a labeled transition. We prove correctness and usefulness of the algorithm. We also deploy the garbage collector in a real-time system, to wit, the runtime kernel of Timber. Timber is a strongly typed, object-oriented, purely reactive, real-time programming language based on reactive objects. We show how properties of the language can be used in order to accomplish very efficient and predictable garbage collection.

  • Deponering eller återvinning i anläggningsarbeten - en LCA-studie av Boliden järnsand

    • Vid utvinning av koppar på Boliden, Rönnskärsverken i Skellefteå uppkommer en glasartat slagg som går under benämningen järnsand. I järnsanden finns bl.a. olika metaller i relativt låga halter. Tekniska undersökningar av materialet har visat att detta mycket väl kan användas som ersättningsmaterial för krossade stenmaterial i olika anläggningsapplikationer bl.a. i vägkonstruktioner. Naturvårdsverket har nu emellertid tagit fram förslag på nya kriterier för återvinning av avfall i anläggningsarbeten. Kriterierna är relativt strikta vilket kan medföra att den praktiska användningen av järnsanden kan omöjliggöras och att järnsanden därmed kommer att läggas på deponi i stället vilket inte sker idag. IVL Svenska Miljöinstitutet har på uppdrag Boliden AB genomfört föreliggande studie rörande en utvärdering av det föreliggande förslaget från Naturvårdsverket. Den föreliggande rapporten belyser avfallshanteringen av järnsand ur ett systemperspektiv med livscykelanalysmetodik (LCA) för två olika alternativa omhändertagandemetoder för järnsand. Det ena alternativet visar användningen av järnsand i en vägapplikation och det andra alternativet visar konsekvenserna av att lägga järnsanden på en deponi.

    • Context adaptation is an important aspect of prognostics and diagnostics as it facilitates the ability to provide relevant information to the consumers of that information. For example, eTechnologies facilitate the communication of data between systems and they assist stakeholders in different parts of an enterprise to make decisions based on the same data. However, decisions may differ, depending on individual stakeholders and specific prognostic and diagnostic processes and techniques, e.g. data fusion or data mining. The correct use of eTechnologies can improve the maintenance of an item or system, thus extending its remaining useful life. The challenge is learning how to make good use of eTechnology and ensure that the right information is provided to the right information consumer, e.g. stakeholders within various processes of an enterprise.Technical solutions in industry are often complex; they are created in heterogeneous environments and the information generated must adapt to different user contexts. Hence, context adaptation can be considered a key requirement for systems operating in heterogeneous environments. This paper considers context to be a template that describes a set of generalised characteristics of a real-world situation; the information in the template differs depending on the situation but the type of information is the same. It defines context adaption as the process of gathering information about the context template, such as structure and information type, evaluating this information and changing the observable behaviour to fit the current context

    • The aim of this special issue is to understand better the many changes in adolescent psychopathology have taken place over the last decades. The factors associated with adjustment problems and psychopathology in adolescence today are not necessarily the same as the factors that predicted problems and psychopathology in the past. But the basic strategies for connecting negative experiences with adolescent psychopathology remain as important today as they were for understanding adolescent psychopathology decades ago. This is well exemplified in the studies included in this Special Issue. What all this studies have in common is that parenting and the family environment are assumed to play a key role in adolescents' adjustment and psychopathology. Finally, given that all papers in this special issue are based on conference presentations at the 15th Biennial Conference of the European Association for Research on Adolescence (EARA), some more information on that conference in included in this introduction.

    • The pure cultures of microalgae Chlorella vulgaris ATCC 13482 and Scenedesmus obliquus FACHB 417 were grown in municipal wastewater in 7-L airlift bubble column photobioreactor supplied with 5% CO2/air (v/v). Batch experiments were conducted at 25 A degrees C with 14-h light/10-h dark cycle for a period of 10 days. The CO2 capture efficiencies for both the microalgae were monitored in terms of their respective biomass productivities, carbon contents, and CO2 consumption rates. In the present study, the initial concentration of ammonia (43.7 mg L-1) was decreased to 2.9 and 3.7 mg L-1 by C. vulgaris and S. obliquus, respectively. And, the initial concentration of phosphate (18.5 mg L-1) was decreased to 1.1 and 1.6 mg L-1 by C. vulgaris and S. obliquus, respectively. CO2 biofixation rates by C. vulgaris and S. obliquus, cultivated in municipal wastewater, were calculated to be 140.91 and 129.82 mg L-1 day(-1), respectively. The findings from the present study highlight the use of microalgae for wastewater treatment along with CO2 uptake and biomass utilization for pilot scale production of biodiesel, biogas, feed supplements for animals, etc., thus minimizing the production costs.
