  • Genetic analysis of a temperament test as a tool to select against everyday life fearfulness in Rough Collie

    • Fear-related problems are common among Rough Collies in Sweden. Annually, on average, >200 Rough Collies are subjected to the dog mentality assessment (DMA), a temperament test during which 33 behavioral reactions are rated. Previous research has shown that a dog's DMA result can be condensed into 5 underlying personality traits. The aim of the study was to evaluate if it is possible to use the DMA for selection for temperament in Swedish Rough Collies, in particular to decrease everyday life fearfulness. We also wanted to compare 2 methods to compute the personality traits: summated scales (SS) and factor scores (FS). The DMA data for 2,953 Rough Collies were used to estimate genetic parameters for the 5 personality traits (both SS and FS), using a linear animal model including fixed effects of sex, year and month of test, and random effects of litter, judge, test occasion, genetic effect of the individual, and residual. Age at test was included as linear and quadratic regressions. The DMA personality trait heritability estimates ranged from 0.13 to 0.25. The SS showed greater or equal heritability estimates compared with the FS. To validate the DMA, data on everyday life behavior of 1,738 Rough Collies were collected using an extended version of the Canine Behavioral Assessment and Research Questionnaire. Each dog's questionnaire result was condensed into 18 underlying behavioral subscales. Genetic parameters for the subscales were estimated using a linear animal model, including a fixed effect of sex and random genetic effect of the individual and residual. Age when the questionnaire was completed was included as linear and quadratic regressions. Heritability estimates for the questionnaire subscales were 0.06 to 0.36. There were high and significant genetic correlations between DMA personality traits and questionnaire subscales. For instance, the DMA personality trait Curiosity/Fearlessness correlated strongly genetically to the questionnaire subscale Non-social fear (-0.70), DMA Sociability to Stranger-directed interest (0.87) and Stranger-directed fear (-0.80), DMA Playfulness to Human-directed play interest (0.63), and DMA Chase-proneness to Chasing (0.73). We could not detect any obvious difference in validity between DMA SS and FS. We conclude that the DMA is an effective tool for selection of breeding animals with the goal to decrease everyday life fearfulness among Swedish Rough Collies. The DMA can also be used for breeding for other traits. The SS method seems to perform at least as good the FS method.

    • A ratio of 1.36:1 in relative bioactivity of RRR-alpha-tocopheryl acetate as a natural (Nat-alpha-T) source to all-rac-alpha-tocopheryl-acetate, as a synthetic (Syn-alpha-T) source, is generally accepted. This factor also largely reflects the difference in bioavailability. However, studies indicate that neither bioavailability of alpha-tocopherol stereoisomers nor relative bioavailability between them are constant, but are dose-dependent and differ between organs. However, no information is available about how different ratios between synthetic and natural alpha-tocopherol affect bioavailability of alpha-tocopherol stereoisomers. Thirty lambs were randomly assigned to diets supplied with additives containing 5 different Syn-alpha-T to Nat-alpha-T ratios, including 100: 0, 75: 25, 50: 50, 25: 75, and 0:100. The experiment lasted for 70 d after which the lambs were slaughtered. The amount of RRR-alpha-tocopherol generally increased in plasma and organs with increasing the proportion of Nat-alpha-T in the diet (P < 0.05). However, the relative bioavailability of RRR- and RRS-alpha-tocopherol in plasma, organs, and abdominal fat generally decreased with increasing the proportion of Nat-alpha-T in the diet (P < 0.05), whereas the other stereoisomers only showed minor changes with the exception of liver. However, a linear response was maintained between the ratio of stereoisomers in the feed and the ratio in plasma and organs. In conclusion, regardless of Syn-alpha-T to Nat-alpha-T ratio in the diets, amounts of alpha-tocopherol stereoisomers in plasma, brain, heart, lungs, and abdominal fat were in the following order: RRR > RRS, RSR, RSS > Sigma 2S.

    • Improvement of farrowing rate (FR) and litter size (LS) of sows that are AI with frozen-thawed (FT) semen can hardly be reached without identification of the factors behind the high variability seen among trials. Three experiments using weaned (4-d wean-to-estrus interval) multiparous (parity 2 to 7) sows were conducted to evaluate the effect of period of the year on FR and LS of FT-inseminated sows in southern Spain. Sows were grouped into 2 periods of the year: winter-spring (November to April; WS) and summer-autumn (May to October; SA). Ovarian status was monitored by transrectal ultrasonography to record how long before or after ovulation AI was performed (pre-, peri-, or postovulatory AI) and to determine the onset of estrus-to-ovulation interval (EOI). Inseminations were performed using deep intrauterine AI with 1.5 x 109 FT sperm per dose. The first experiment was designed to determine the influence of the period of the year on FR and LS of FT semen. Sows (116 in WS and 100 in SA) were AI at 33 and 39 h after the onset of estrus. The period of the year influenced the FR and LS (P less than 0.01). Farrowing rate and LS were least in SA (P less than 0.05). This pattern of annual variation was similar to that shown by sows on the same farm currently undergoing AI with liquid semen (cervical AI at 12 and 36 h after the onset of estrus with 3 x 109 sperm per dose). However, the FR reduction in SA respect to WS was more substantial in sows artificially inseminated with FT (77.6 vs. 50%, P less than 0.001) than those artificially inseminated with liquid semen (83.9 vs. 71.8%, P less than 0.05). More pre- and less periovulatory AI were performed in SA sows than in WS sows (P = 0.05). Experiment 2 was designed to evaluate whether the period of the year influenced EOI. Ovarian status was transrectal ultrasonography scanned every 6 h after the onset of estrus until the end of ovulation (WS: 30; SA: 31 sows). There were more sows with long EOI (greater than48 h) in SA than in WS (P = 0.05). Experiment 3 aimed to improve the reduced FR and LS recorded in SA sows when using FT semen (Exp. 1) by inducing ovulation with eCG + hCG. A single AI with FT semen was performed 5 h before the expected ovulation (55 sows). As a control, spontaneously ovulating sows (n = 53) were FT-inseminated as in Exp. 1. Hormonal induction of ovulation did not improve FR and LS (P greater than 0.05). In the Spanish Mediterranean area, a longer EOI during SA negatively influenced the FR and LS of weaned sows after AI. This effect was particularly evident when FT semen was used. These findings were not ameliorated by hormonal induction of ovulation.

  • Effects of source and concentration of neutral detergent fiber from roughage in beef cattle diets: Comparison of methods to measure the effectiveness of fiber

    • Methods have been developed to measure the effectiveness of many roughages, but few evaluations have been conducted with tropical feeds. The objectives of this research were to determine the effectiveness of roughage sources based on bioassay and laboratory methods and identify the biological attributes of the diets that correlate with these methods. Six ruminally cannulated Nellore steers (408 +/- 12 kg of BW) were randomly assigned to a 6 x 6 Latin square design within six diets: negative control diet (NC) with aNDF as 10% from corn silage (CS); positive control diet (PC) with aNDF as 20% from CS; and four diets containing 10% aNDF from CS and 10% aNDF from each of the following sources: sugarcane (SC), sugarcane bagasse (SCB), soybean hulls (SH), or low oil cottonseed hulls (LOCH). Physical effectiveness factor (pef, related to the physical characteristics of aNDF) and effectiveness factor (ef, related to the ruminal pH) were determined based on a linear model approach that uses a bioassay method in which CS aNDF was assumed to be the standard fiber source. Laboratory methods to estimate pef of roughage sources were based on the proportion of DM of roughage retained on a 1.18-mm sieve pef(>1.18 mm) or retained on the 8.0-mm Penn State Particle Separator screen pef(>8.0 mm). The pef calculated by the bioassay method (total chewing time and ruminal mat resistance) for CS, SCB, and SC were higher values (P < 0.05) compared with SH and LOCH. The pef(rumen mat) of SC and SCB were higher (P < 0.05) than that of CS, SH, and LOCH. The pef(rumen mat) of LOCH was 61% higher than SH. The ef(rumen pH) of SC and LOCH was higher (P < 0.05) than CS and SH. The pef(chewing, min/d), pef(chewing, min/kg of DM), pef(rumen mat), and ef(rumen pH) positively correlated with rumination time, total chewing time, and ruminal mat resistance (values from transit time in seconds). No correlation was observed (P > 0.05) between pef(>8.0 mm) and rumination time, chewing time, and ruminal pH. The pef calculated using the bioassay method as well as pef (>8.0 mm) were negatively correlated with rumen pH (P > 0.05). The values of the effectiveness of fiber sources obtained in this research can be used as a guideline for nutritionists aiming to replace roughage sources from tropical regions in beef cattle finishing diets. Under our conditions, the pef using the bioassay method or laboratory methods were not adequate in predicting ruminal pH.

    • The objectives of this research were to evaluate the effects of source and concentration of a-amylase-treated neutral detergent fiber (aNDF) from roughage on feed intake, ingestive behavior, and ruminal kinetics in beef cattle receiving high-concentrate diets. Six ruminally cannulated Nellore steers (408 +/- 12 kg of body weight) were randomly assigned to a 6 x 6 Latin square design with six diets: 10% aNDF from corn silage (10CS); 20% aNDF from corn silage (20CS); or four diets containing 10% aNDF from corn silage and 10% aNDF from one of the following sources: sugarcane (SC), sugarcane bagasse (SCB), soybean hulls (SH), or low oil cottonseed hulls (LOCH). The parameters of passage and degradation kinetics were estimated based on a two-compartmental model with gamma- and exponential-distributed residence times. The nonlinear models were fitted by nonlinear least squares, and a linear mixed-effects model was fitted to all variables measured from the Latin square design that were related to intake, digestibility, digestion kinetic parameters, and residence times. Mean particle size (MPS) between roughage sources (CS, SCB, and SC) and coproducts (SH and LOCH) was affected (P < 0.05). Dry matter intake (DMI) was not affected (P > 0.05) by 20CS, SC, SH, or LOCH. Steers fed 20CS or LOCH diets had 16% and 20% greater DMI, respectively, (P < 0.05) than steers fed 10CS diet. Steers fed SCB consumed the least dry matter (DM). The SH and LOCH diets had lower MPS values (about 8.77 mm) in comparison to 20CS, SCB, and SC diets (about 13.08 mm) and, consequently, affected (P < 0.05) rumen content, ruminal in situ disappearance, nutrient digestibility, and solid fractional passage rate. Chewing time was affected (P < 0.05) by roughage sources and concentration. Lower values of distance travel inside the rumen (min/cm) were observed (P < 0.05) for the SCB and SC diets in comparison with any other diet. Except for SCB, there was no difference (P > 0.05) in rumen fill, among other treatments. Mean daily ruminal pH was not affected (P > 0.05) by 20CS, SCB, SC, and LOCH diets, and it ranged from 6.1 to 6.23. Total short-chain fatty acids concentration was affected (P < 0.05) by roughage source and concentration. Based on our results, we recommend that under Brazilian finishing diets, replacing roughage sources, except for SCB, based on aNDF concentration of the roughage in high-concentrate diets containing finely ground flint corn does not affect DMI.

    • The apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of nutrients were evaluated in 2 commercially available products: wheat (Triticum aestivum) wet distillers grain with solubles (WDGS) and wheat dried distillers grain with solubles (DDGS). Two diets included (DM basis) 50% basal diet with either 50% WDGS (W) or 50% DDGS (D). The basal diet included corn (Zea mays) starch, sugar, vitamins, and minerals. Seven castrated male pigs with post valve T-cecum cannulas were fed the diets according to a changeover design during two 14-d periods. In a pre- and postperiod, casein was given as the only protein source with the basal diet to estimate endogenous losses of N and AA for calculation of standardized ileal digestibility (SID). The AID of OM did not differ between diets, but ATTD of OM was higher (P < 0.05) for diet W. The AID (76.2 vs. 68.9%), SID, and ATTD of CP was higher (P < 0.05) in diet W than diet D. The SID for Lys (75.7 vs. 51.8%) and Met (75.8 vs. 70.1%) was higher (P < 0.01) in WDGS than DDGS. In conclusion, drying of wheat distillers grain products can markedly lower ileal digestibility of Lys and Met whereas negative effects on energy value are small.

  • Inverted teats (Mammillae invertitae) in gilts - Effect on piglet survival and growth rate

    • In the modern pig industry, the increasing number of piglets born per litter augments the importance of the number of functional teats in the sow. The aim of this study was to evaluate the function and importance of inverted teats during nursing and to analyze structural and functional differences between the mammary glands of inverted teats versus normal teats. Nine farrowing gilts (8 purebred Swedish Yorkshire gilts and 1 cross between Swedish Yorkshire and Norwegian Landrace) and 94 piglets (59 piglets suckling normal teats, 32 piglets suckling protruded teats [i.e., previously inverted], 2 piglets suckling inverted teats, and 3 piglets suckling considerably smaller teats) were included in the study. Teat fidelity (keeping the same teat between the nursings) was registered, excluding the first 48 h postpartum. Piglet weight was recorded daily during the first week of life and thereafter once a week until weaning at 4 wk of age. Weight and growth rate were analyzed using repeated observation mixedmodel analysis of variance. The 2 piglets that suckled the inverted teats were not able to emerge the teats and they were euthanized 4 and 8 d after birth, respectively, due to loss of BW. The average weight at weaning (28 d of age) was 8.1 kg (range 3.2-13.8 kg). In the normal teats (n = 53), the weight of the corresponding mammary gland tissue at necropsy was positively correlated to the piglet average daily weight gain during wk 2 (r = 0.33, P < 0.05), 3 (r = 0.55, P < 0.001), and 4 (r = 0.47, P < 0.001). In the protruded teats (n = 32), the weight of the corresponding mammary gland tissue was positively correlated to the piglet average daily weight gain during wk 2 (r = 0.63, P < 0.001) and 3 (r = 0.43, P < 0.05). Among the piglets nursing normal teats, 82% kept fidelity to its teat and the corresponding percent for the protruded teats was 26%. In 7 of the 9 sows, the weaning weight of the piglets suckling protruded teats was numerically lower compared to the piglets suckling normal teats, although the difference was not statistically significant. Piglets nursing small teats had lower weaning weight (4.8 kg) and the corresponding mammary tissue also had lower weight (335 g). This study indicates the importance of an adequate number of well-developed and well-functioning teats in healthy sows for the growth and development of the piglets but also emphasizes the complexity of the interaction between the nursing sow and her piglets.

  • Road transport and diet affect metabolic response to exercise in horses

    • This study investigated the effects of transport and diet on metabolic response during a subsequent race-like test in Standardbred horses in training fed a forage-only diet and a 50: 50 forage: oats diet. Six trained and raced Standardbred trotter mares were used. Two diets, 1 forage-only diet (FONLY) and 1 diet with 50% of DM intake from forage and 50% from oats (FOATS), were fed for two 29-d periods in a crossover design. At Day 21, the horses were subjected to transport for 100 km before and after they performed an exercise test (transport test [TT]). At Day 26, the horses performed a control test (CT), in which they were kept in their stall before and after the exercise test. Blood samples were collected throughout the study, and heart rate and water intake were recorded. Heart rate and plasma cortisol, glucose, and NEFA concentrations were greater for the TT than for the CT (P = 0.008, P = 0.020, P = 0.010, and P = 0.0002, respectively) but were not affected by diet. Plasma acetate concentration was lower during the TT than during the CT (P = 0.034) and greater for the FONLY than for the FOATS (P = 0.003). There were no overall effects of the TT compared with the CT on total plasma protein concentration (TPP), but TPP was lower with the FONLY than with the FOATS (P = 0.016). There was no overall effect of the TT compared with the CT on water intake, but water intake was greater with the FONLY than the FOATS (P = 0.011). There were no overall effects of transport or diet on BW, plasma lactate, or plasma urea concentration. It was concluded that both transport and diet affect metabolic response during exercise in horses. Aerobic energy supply was most likely elevated by transportation and by the FONLY. The FONLY also decreased exercise-induced effects on extracellular fluid regulation. These results highlight the importance of experimental design in nutrition studies. If the aim is to examine how a diet affects exercise response in competition horses, transport should preferably be included in the experimental design, because horses are likely to be transported before a competition.

  • The Saga of Just One QTL; From Laying Hen Bone Strength to One Carbon Metabolism

    • Skeletal damage is a challenge for laying hens, as the adaptations for egg laying make them susceptible to osteoporosis. We know that activity, growth and puberty influence skeletal quality genetics. However, in this story, we follow the process of resolving a QTL for skeletal quality to a single gene. Providing both a nutritional and a genetic solution and illustrating another level of complexity. Using divergently selected populations in an F2 cross, a QTL for skeletal quality was described. Utilising a series of experiments in the commercial founder line the QTL was fine mapped on chromosome 1 and the differences in physiology of hens that segregated for markers at the locus defined e.g. tibia breaking strength 200.4 vs 218.1 Newton (p&lt; 0.002). Transcriptome profiling of tibia from the high and low bone strength genotype revealed a highly differentially expressed gene co localising to the QTL, the enzyme cystathionine beta synthase (CBS). CBS is a component of one carbon metabolism. Plasma homocysteine, the substrate of CBS had a higher concentration in the genotype associated with poor bone quality. Homocysteine interferes with crosslinking of the collagen matrix, critical to bone formation. Homocysteine can be re-methylated into methionine, a limiting amino acid in laying hens, using betaine as a methyl donor. Testing the hypothesis that reducing plasma homocysteine by feeding betaine, a widely available feed additive, would improve bone quality in laying hens proved successful.This is just one component of a complex trait and its elucidation took us along unexpected routes. However, I am optimistic that with modern tools and an open mind we can progress more quickly. Alongside understanding the factors that determine skeletal quality genetics, the discovery of the mechanisms behind complex traits, although rarely easy, offer the potential for novel interventions to improve animal health and production.

    • Biosolids (processed human sewage sludge), which contain low individual concentrations of an array of contaminants including heavy metals and organic pollutants such as polycyclic aromatic hydrocarbons (PAH), polychlorinated biphenyls (PCB), and polychlorinated dibenzodioxins/polychlorinated dibenzofurans known to cause physiological disturbances, are increasingly being used as an agricultural fertilizer. This could pose a health threat to both humans and domestic and wild animal species. This review summarizes results of a unique model, used to determine the effects of exposure to mixtures of environmentally relevant concentrations of pollutants, in sheep grazed on biosolids-treated pastures. Pasture treatment results in nonsignificant increases in environmental chemical (EC) concentrations in soil. Whereas EC concentrations were increased in some tissues of both ewes and their fetuses, concentrations were low and variable and deemed to pose little risk to consumer health. Investigation of the effects of gestational EC exposure on fetal development has highlighted a number of issues. The results indicate that gestational EC exposure can adversely affect gonadal development (males and females) and that these effects can impact testicular morphology, ovarian follicle numbers and health, and the transcriptome and proteome in adult animals. In addition, EC exposure can be associated with altered expression of GnRH, GnRH receptors, galanin receptors, and kisspeptin mRNA within the hypothalamus and pituitary gland, gonadotroph populations within the pituitary gland, and regional aberrations in thyroid morphology. In most cases, these anatomical and functional differences do not result in altered peripheral hormone concentrations or reproductive function (e.g., lambing rate), indicating physiological compensation under the conditions tested. Physiological compensation is also suggested from studies that indicate that EC effects may be greater when exposure occurs either before or during gestation compared with EC exposure throughout life. With regard to human and animal health, this body of work questions the concept of safe individual concentration of EC when EC exposure typically occurs as complex mixtures. It suggests that developmental EC exposure may affect many different physiological systems, with some sex-specific differences in EC sensitivity, and that EC effects may be masked under favorable physiological conditions.
