    • The influence of the trace metals, copper, aluminum and iron, and of the strong complexing agents, EDTA and NTA, on phytoplankton growth in water from a brackish water bay was investigated through bioassay experiments. A diatom (Skeletonema costatum (Grev.) Cleve) and a dinoflagellate (Prorocentrum minimum (Pav.) J. Schiller) were used as test organisms. The growth of both phytoplankton species was strongly inhibited by copper. This inhibition was generally eliminated by EDTA and NTA. Both phytoplankton species were considerably less inhibited by aluminum than by copper at the same total metal concentration. While S. costatum responded to copper and chelator additions in the same way in sea water samples from different seasons, the growth of P. minimum exhibited pronounced seasonal variation. Other parameters than the values of pCu must be considered in order to account for the experimental results. This work supports the theory that alterations in contents of trace metals and natural chelators in sea water are important factors behind shifts in phytoplankton species composition.

    • The potential of eel (Anguilla anguilla) as a monitoring species for the Thames Estuary, UK, was examined. Hepatic cytochrome P4501A [7-ethoxyresorufin O-deethylase (EROD) activity] and blood vitellogenin (Western analysis) were investigated as biomarkers of exposure to, respectively, organic contaminants and to contaminants showing estrogenic activity. Hepatic microsomal EROD activities in A. anguilla from seven sites in the Thames Estuary in May 1998 varied three-fold (111 +/- 24 to 355 +/- 42 pmol min-1 mg protein-1) (mean +/- S.E.M.) and showed correlation with salinity; however, the latter relationship was not maintained at other times of the year. The range of EROD activities was two- to eight-fold higher than the 37 +/- 8 pmol min-1 mg-1 for A. anguilla from the relatively clean Tamar Estuary. beta-Naphthoflavone treatment (5 mg kg-1 wet wt.; 2 days) of Thames A. anguilla produced a two-fold increase in hepatic microsomal EROD activity. Comparing the Thames EROD data with those for A. anguilla from well-characterised contaminated sites in the Netherlands (Van der Oost, R., Goksøyr, A., Celander, M., Heida, H., & Vermeulen, N. P. E. 1996. Aquatic Toxicology, 36, 189-222), the Thames is suggested to be moderately impacted by polycyclic aromatic hydrocarbons and related contaminants. 17-beta-Estradiol treatment produced the appearance of a plasma protein of 211 Kd app. mol. wt. (recognised by antibodies to vitellogenin of Morone saxatilis), but putative vitellogenin could not be detected in A. anguilla from selected sites in the Thames Estuary.

    • Carbonyl reductase (CBR) is an enzyme involved in protection from oxidative stress. In rainbow trout (Oncorhynchus mykiss), the hepatic mRNA abundance of the two isoforms (A and B) is increased after exposure to treated sewage effluents, as well as after exposure with beta-naphthoflavone (beta-NF) and the pro-oxidant paraquat In this study, we show that the same chemicals similarly increase the single known hepatic CBR mRNA level and CBR catalytic activity in the coastal living eelpout (Zoarces viviparus). Hepatic CBR mRNA abundance and catalytic activity were also compared between eelpout collected at contaminated and reference sites on the Swedish west coast, but no differences were observed. In conclusion, CBR is a potential biomarker candidate for monitoring the exposure and effects of AhR agonists and/or pro-oxidants in the marine environment, but more research is needed to investigate temporal regulation as well as dose dependency for different chemicals. The mRNA and enzymatic assays presented in this study provide two additional tools for researchers interested in expanding their biomarker battery. (c) 2012 Elsevier Ltd. All rights reserved.

  • Biomarker responses in eelpouts from four coastal areas in Sweden, Denmark and Germany

    • To increase our understanding of possible chemical impacts on coastal fish populations in the Baltic Sea, Kattegat and Skagerrak, the viviparous eelpout (Zoarces viviparus) was used as sentinel species in two major sampling campaigns (spring and autumn) in 16 different coastal sites. Condition factor (CF), liver somatic index (LSI), gonad somatic index (GSI) were measured and the activity of the hepatic enzymes ethoxyresorufin-O-deethylase (EROD), glutathione reductase GR), glutathione S-transferase (GST), catalase (CAT) and muscular activity of acetylcholinesterase (AChE) were assessed. PAH metabolites in bile were also analyzed. The most notable finding in the data set was the low EROD activity in eelpouts collected at the relatively polluted region in Germany compared to the other regions, which could be due to an inhibition of the CYP1A-system or to adaptation to chronic exposure of pollutants in this area. Additionally, low AChE activity was noted in the German region in the autumn campaign and low AChE activity detected in the Danish region in the spring campaign. These differences suggest possible season-specific differences in the use and release of AChE-inhibiting chemicals in the Danish and German regions. Clustering of biomarkers on site level indicated a relationship between CF and GSI and suggested that sites with a high CF contained eelpout that put a larger effort into their larvae development. Clustering of the oxidative stress markers GR, GST and CAT on the individual level reflected a possible coordinated regulation of these enzymes. Overall, the results support the importance of taking into account general regional differences and seasonal variation in biomarker activity when monitoring and assessing the effects of pollution. Despite the expected seasonal variation for most of the measured endpoint, several markers (GSI, EROD and CF) vary similarly between all selected sites in both spring and autumn. This suggests that the differences between sites for these endpoints are independent of season.

    • In laboratory tests, manganese accumulation by the appendages of the sediment burrowing Norway lobster, Nephrops norvegicus (L.) (including the lateral antennules) was approximately three times greater [600 mug Mn g(-1) (dry weight) after 5 days in 20 mg Mn l(-1)] than that by the carapace. The accumulation was linearly dose-dependent (10-40 mg Mn l(-1)) and duration-dependent (2-30 days), and showed no decrease after 3 weeks in undosed seawater. A high manganese uptake to the lateral antennules during hypoxia in the field was verified from the SE Kattegat, Sweden. These results indicate that accumulation of Mn on the mobile appendages of the Norway lobster fulfils most of the criteria for a biomarker of exposure to hypoxia. Using these measurements in conjunction with Mn concentrations in the internal tissues, it may be possible to resolve both the timing and the extent of the Mn exposure and the underlying hypoxic event.   • The role epiphytes play in particle capture of seagrass canopies

    • Seagrass epiphytic communities act as ecological indicators of the quality status of vegetated coastal environments. This study aims to determine the effect leaf epiphytes has on the sediment capture and distribution from outside sources. Thirteen laboratory experiments were conducted under a wave frequency of 0.5 Hz. Three epiphyte models were attached to a Zostera marina canopy of 100 plants/m2 density. The sediment deposited to the seabed, captured by the epiphytic leaf surface, and remaining in suspension within the canopy were quantified. This study demonstrated that the amount of epiphytes impacts on the sediment stocks. Zostera marina canopies with high epiphytic areas and long effective leaf heights may increase the sediment captured on the epiphyte surfaces. Also, reducing suspended sediment and increasing the deposition to the seabed, therefore enhancing the clarity of the water column. For largest epiphytic areas, a 34.5% increase of captured sediment mass is observed. The sediment trapped on the leaves can be 10 times greater for canopies with the highest epiphytic areas than those without epiphytes. Therefore, both the effective leaf length and the level of epiphytic colonization are found to determine the seagrass canopy ability at distributing sediment.

  • Dissimilatory bioreduction of iron(III) oxides by Shewanella loihica under marine sediment conditions

    • Shewanella is a genus of marine bacteria capable of dissimilatory iron reduction (DIR). In the context of deep-sea mining activities or submarine mine tailings disposal, dissimilatory iron reducing bacteria may play an important role in biogeochemical reactions concerning iron oxides placed on the sea bed. In this study, batch experiments were performed to evaluate the capacity of Shewanella loihica PV-4 to bioreduce different iron oxides (ferrihydrite, magnetite, goethite and hematite) under conditions similar to those in anaerobic sea sediments. Results showed that bioreduction of structural Fe(III) via oxidation of labile organic matter occurred in all these iron oxides. Based on the aqueous Fe (II) released, derived Fe(II)/acetate ratios and bioreduction coefficients seem to be only up to about 4% of the theoretical ones, considering the ideal stoichiometry of the reaction. A loss of aqueous Fe (II) was caused by adsorption and mineral transformation processes. Scanning electron microscope images showed that Shewanella lohica was attached to the Fe(III)-oxide surfaces during bioreduction. Our findings suggest that DIR of Fe(III) oxides from mine waste placed in marine environments could result in adverse ecological impacts such as liberation of trace metals in the environment.

  • Colonisation of wave power foundations by mobile mega- and macrofauna - a 12 year study

    • Environmental impacts from wave energy generators on the local mobile mega- and macrofauna community have been investigated in the Lysekil project by Uppsala University. Offshore renewable energy installations provide hard, artificial substrates, and as such, they could act as artificial reefs. Foundations with manufactured holes served as complex habitats and foundations without served as non-complex. In this long-term study, SCUBA surveys of mobile fauna in the years 2007, 2008 and 2016-2019 were analyzed. The results show a distinct reef effect on the foundations with significant greater species richness, total number of individuals, greater values of the Shannon-Wiener biodiversity index, and greater abundance of specific reef fauna. Complex foundations accommodated a greater abundance of brown crabs than non-complex foundations, other taxa did not show differences between the two foundation types. A successional increase of species richness, numbers of individuals and Shannon-Wiener biodiversity could be revealed from the first to the second survey period. Inter-annual variation was visible throughout all taxa and years.
