    • Microextraction by packed sorbent (MEPS) is a new technique for sample preparation that can be connected on-line with liquid chromatography (LC) or gas chromatography (GC) systems without any modifications. This article describes the use of MEPS in clinical and preclinical studies to quantify different drugs in whole blood samples. MEPS was used to determine cyclophosphamide in mouse blood from preclinical g studies using 20 mu L of blood samples. The interday accuracies and 0 precisions ranged from 107-109% and from 2.0-7.0%, respectively. The determination of four immunosuppressive drugs in human blood by MEPS and liquid chromatography-mass spectrometry (LC-MS) is described. The method showed a good selectivity and sensitivity. The calibration curves for everolimus, sirolimus, and tacrolimus ranged from 0.5 to 50 ng/mL and for cyclosporine from 3.0 to 1500 ng/mL. Intraday precisions for the studied immunosuppressive drugs were 2.0-11.7% and interday precision ranged from 5.1 to 13.7% (CV).

    • The selectivity of different combinations.of organic modifiers, pH, and types of reversed-phase liquid chromatography (LC) materials has been characterized using Tanaka column characterization, linear solvent energy relationships (LSER), and selectivity correlations. The three characterization techniques highlighted the potential complementary selectivity of these phases and conditions as well as the type and dominancy of some of the retention mechanisms involved. Subsequently, selectivity differences were proven to be valid in the practical separation of acids, bases, and neutral analytes. This paper aims to help chromatographers to produce highly efficient method development strategies for reversed phase LC separations in a relatively short time frame.

    • A new concept for the extraction of solid samples has been developed. The popular pressurized liquid extraction (PLE) technique has been modified so that the extraction cell can be divided, which greatly increases the flexibility and versatility of this technique. The system uses simple adaptors to couple commercially available extraction cells, which may be loaded with either sample or adsorbent(s). These segmented extraction cells make it possible to selectively retain analytes or matrix to achieve simultaneous extraction and clean-up or fractionation. The ultimate aim is to perform simultaneous extraction and clean-up and obtain a purified extract that is ready for instrumental analysis, all in line with current trends in analytical chemistry to automate and streamline the analytical procedures to reduce the analysis costs, solvent consumption, manual labour and increase the quality of analysis. Selected modes of operations are presented and a few recent environmental applications of modular PLE are reviewed.

The profiling approach is complemented by UHPLC-LTQ-Orbitrap mass spectrometry in MSn mode for de novo lipid identification.
