    • Full thickness wounds, such as deep burns, need restoration of both the dermal and epidermal layers of the skin. In normal wound healing, re-epithelialization occurs by migration and proliferation of keratinocytes from the wound edges and by differentiation of stem cells from remaining hair follicles. Restoration of dermis occurs by influx of growth factors secreted by macrophages, platelets, and fibroblasts; by fibroblast proliferation and subsequent synthesis and remodeling of collagenous dermal matrix. In the case of full-thickness acute burn injuries and chronic wounds (e.g. pressure ulcers, venous ulcers and diabetic foot ulcers), these processes are defective. With the principles of tissue engineering in mind (to correct, improve and maintain tissues and their functions), researchers have developed promising materials and methods to make it possible to restore either the dermal (Integra® DRT, Alloderm®) or the epidermal layer (split thickness skin grafts (STSG), cultured epithelial autografts (CEA), autologous keratinocytes in single cell suspension). It is now well established that superior results are obtained if both dermal and epidermal components are combined, for example in a bilayered skin equivalent. Apligraf® is recommended for use on venous ulcers and is the only bilayered living skin equivalent currently approved by the FDA. Studies on different factors affecting the wound healing capacity as well as techniques in use provide valuable information for further development.In this licentiate thesis, we evaluated different transplantation techniques for delivering cultured human keratinocytes in single cell suspension, a measure becoming more frequently used in addition to STSG and CEA for restoring the epidermal layer of the skin. We found that the pressure device, commonly used to spray cell suspension onto the wound with pressures as high as 200 kPa, killed around 0% of the cells. In comparison, an ordinary syringe with the attachment of a spray nozzle showed almost 90% viable cells post transplantation and provided an equally good distribution of the cell suspension.We also studied different silver containing dressings regarding silver accumulation in human skin. In addition, we graded the re-epithelialization to evaluate whether the dressings caused any delay in the wound healing process. We found that the silver dressings tested, with few exceptions, caused dermal accumulation of silver, primarily aggregated around blood vessels. We could also show that most of the dressings had negative effect on the re-epithelialization.For the restoration of the dermal layer of the skin, Integra® DRT functions as a scaffold for guided tissue regeneration of the dermis. We had the possibility to study a case of necrotizing fasciitis were the treatment consisted of the use of Integra® DTR together with sub-atmospheric pressure (after initial surgical debridement) and later transplantation of split thickness skin grafts. This measure proved to be safe as well as giving satisfactory pliable and aesthetically acceptable result.

  • User-driven innovation in tourism : A review of methodologies

    • This literature study reviews user-driven innovation and establishes a typology of its forms in a tourism context. Sixteen methods are distinguishable. They comprise situations where users are actively involved and methods where information is collected without direct user involvement. The nature and intensity of the dialogue between companies and their customers are addressed. Drawing on this existing research, the article concludes that there is still little comprehensive follow-up on user-driven innovation in tourism and its impact on quality improvements and assurance. Key areas for future studies are identified.

    • The self-organization of monolayer gold nanoparticles (AuNPs) on 3-aminopropyltriethoxysilane (APTES)-functionalized glass substrate is reported. The orientation of APTES molecules on glass substrates plays an important role in the interaction between AuNPs and APTES molecules on the glass substrates. Different orientations of APTES affect the self-organization of AuNps on APTES-functionalized glass substrates. The as grown monolayers and films annealed in ultrahigh vacuum and air (600 degrees C) were studied by water contact angle measurements, atomic force microscopy, X-ray photoelectron spectroscopy, UV-visible spectroscopy and ultraviolet photoelectron spectroscopy. Results of this study are fundamentally important and also can be applied for designing and modelling of surface plasmon resonance based sensor applications.

    • Sm–Nd data for rocks of granitic composition in an east–west traverse across the Namaqua Sector of the Mesoproterozoic c. 1.2 Ga Namaqua–Natal Province of southern Africa provide new evidence about the timing of crustal extraction from the mantle. Recent ion probe zircon dating has shown that, contrary to previous indications that pre-Namaqua basement had been preserved in parts of the Namaqua Sector, the majority of the magmatic rocks were emplaced during the 1.4–1.0 Ga Namaqua tectogenesis and very little U–Pb evidence of older precursors remains. Sm–Nd model ages show that Mesoproterozoic crustal extraction did occur, but was also strong in the Palaeoproterozoic, with local evidence for the existence of Archaean crust. The distribution of model ages generally corresponds with the established terranes, subdivided using lithostratigraphic and structural criteria. The northern part of the Bushmanland Terrane shows early Palaeoproterozoic to Archaean model ages and is clearly different from the southern part, which has Mesoproterozoic model ages. This supports the previously published results of a north–south traverse and justifies the separation of the Garies Terrane from the Bushmanland Terrane, though further subdivisions are not supported. The continuation of the Richtersveld Province east of Pofadder is supported by one sample, and the terrane boundary between the Bushmanland and Kakamas terranes is marked by an abrupt change from Palaeoproterozoic to Mesoproterozoic model ages. Model ages for the Kakamas and Areachap terranes do not distinguish them well. They suggest a Mesoproterozoic to late Palaeoproterozoic origin for both terranes, neither of which has a purely juvenile character. The influence of Palaeoproterozoic crust-forming events is clear in the Kaaien Terrane to the east, possibly reflecting reworking of the Kaapvaal Craton. The Namaqua Sector of the Province thus has a history of crustal extraction and evolution which reaches back locally to the Archaean, with major Palaeoproterozoic and Mesoproterozoic crust-forming events. This differs from the Natal Sector, which has a largely juvenile character related to a Mesoproterozoic Wilson cycle. Attempts to reconstruct the Mesoproterozoic Supercontinent Rodinia will have to take into account the extensive Nd-isotopic evidence for older crustal events in the Namaqua sector.
