  • Diverse fractionation patterns of Rare Earth Elements in deep fracture groundwater in the Baltic Shield - Progress from utilisation of Diffusive Gradients in Thin-films (DGT) at the Aspo Hard Rock Laboratory

    • Rare earth elements (REEs) were studied in groundwater in fractures at depths between 144 m and 450 m in Proterozoic granitoids on the Baltic Shield at a coastal site in south eastern Sweden (Aspo Hard Rock Laboratory). A specially designed device was used to sample the groundwater under in situ high pressure and low redox conditions. The REEs were measured via both diffusive gradients in thin-films (DGT) samplers after approximately three-week deployment times, and after conventional filtration (0.45 mu m membrane filters). The concentrations of REEDGT were lower than the REE0.45 mu m concentrations in all 14 studied groundwaters. This is explained by development of a diffuse boundary layer (DBL) in the solution at the DGTs caused by the low flow, and in some of the groundwaters additionally by the presence of organically complexed REE that diffuse slowly in the diffusive gel of the DGT. Among the 14 studied groundwaters there was a huge range in REEDGT-fractionation patterns, ranging from enrichment to a nearly three-order-of-magnitude depletion of the heavy REEs (HREEs), despite a relatively homogenous bedrock (granitoids). The HREE enrichment is explained by preservation of the fractionation signature as HREE enriched sea water intruded and flowed through the fractures, supported by the high proportion of modelled REE fulvic-acid complexes in these waters. The strong HREE depletion, which occurred in saline groundwater, was ascribed to an advanced REE equilibrium between the groundwater and primary and/or secondary minerals and the pore water in the bedrock. Yet other groundwaters had flat or moderately HREE depleted patterns, which in some cases may have been caused by mixing of small portions of surficial waters with saline groundwater.     • Several countries are preparing to dispose of radioactive nuclear waste deep underground in crystalline rock. This type of bedrock is commonly extensively fractured and consequently carries groundwater that serves as a medium for transporting metals and radionuclides. A group of metals of particular interest in this context is the rare earth elements (REEs), because they are analogues of actinides contained within radioactive waste and are tracers of hydrological pathways and geochemical processes. Concentrations of REEs are commonly low in these groundwaters, leading to values below detection limits of standard monitoring methods, particularly for the heavy REEs. We present a new technical set-up for monitoring REEs (and other trace metals) in groundwater in fractured crystalline rock. The technique consists of passing the fracture groundwater, commonly under high pressure and containing reduced chemical species, through a device that maintains the physicochemical character of the groundwater. Within the device, diffusive gradient in thin-film (DGT) discs are installed in triplicate. With this set-up, we studied REEs in groundwater in fractures at depths of approximately -144, -280, and -450 m in granitoids in the A"spo Hard Rock Laboratory in southern Sweden. The entire REE suite was detected (concentrations down to 0.1 ng L-1) and was differently fractionated among the groundwaters. The shallowest groundwater, composed of dilute modern Baltic Sea water, was enriched in the heavy REEs, whereas the deeper groundwaters, dominated by old saline water, were depleted in the heavy REEs. Deployment periods varying from 1 to 4 weeks delivered similar REE concentrations, indicating stability and reproducibility of the experimental set-up. The study finds that 1 week of deployment may be enough. However, if the overall setting and construction allow for longer deployment times, 2-3 weeks will be optimal in terms of reaching reliable REE concentrations well above the detection limit while maintaining the performance of the DGT samplers.

  • Open Archives and their Significance in the Communication of Science (Workshop in Uppsala 16-17 November)

    • Today there are several projects concerned with how to use usage data from open archives in order to understand the new ways of how science is communicated. The key issue in this scientific field is to develop secure and reliable statistical analysis for the usage of the objects deposited in open archives. With the increasing efforts to evaluate science in different ways it is important to understand the metrics from open archives. At the Swedish University of Agricultural Sciences (SLU) a two-day workshop was held on the topic in November. The aim was to examine how usage metrics can provide new knowledge about how science is communicated through open archives and if it also can be a tool in the future evaluation of science. The workshop was financed by the OpenAccess.se program and we were more than 60 participants taking part of interesting lectures and discussions. This was a multisited workshop. Most of the participants were in Uppsala but some of us participated via video from SLU-sites in Alnarp and Umeå. Some of the presenters also connected via the same video link during the workshop

    • Rapporten presenterar resultatet av projektet ” Analys av påverkan från EU-förslag för omställningen inom svensk industri”. Projektets syfte har varit att analysera hur Kommissionens förslag till lagstiftningspaket (Fit for 55) för att göra EU redo för att minska utsläppen i linje med det skärpta utsläppsmålet om -55 procent netto till 2030 jämfört med 1990 påverkar möjligheterna till omställning av svensk industri. Analysen visar att förslagen från EU täcker flera av de hinder som finns för att ställa om industrin, men att det inte fullt ut räcker för att uppfylla den svenska målsättningen om nettonollutsläpp till 2045. Analysen visar att EU-förslagen framför allt brister avseende stöd och stöttning för en uppbyggnad av supportinfrastruktur för en transformativ omställning. Alltså stöd för att bygga upp de nya samhällen som krävs för den omställning industrin ska gå mot, både vad gäller kompetensuppbyggnad och samhällsfunktioner, men även frågor kopplade till infrastruktur som eldistribution och vägar. Rapporten har utgjort ett underlag i Naturvårdsverkets och Energimyndighetens arbete med rapporten ”Industrins klimatomställning”

    • Här presenterar resultatet av projektet "Styrmedel för ökad efterfrågan på klimateffektiva produkter". Projektets syfte har varit att analysera hur styrmedel – antingen direkt eller indirekt – kan skapa efterfrågan på produkter med mycket låga eller inga utsläpp.Rapporten är avgränsad till att undersöka, föreslå och analysera styrmedel för att få till transformativa förändringar på systemnivå snarare än åtgärder för inkrementella förbättringar. Detta i syfte att möjliggöra klimatneutral basindustri. Ett antal hinder kopplat till cement-, järn-och stål-, kemi-, och raffinaderiindustrins klimatomställning har identifierats i rapporten och ett antal styrmedel som kan adressera hinder kopplade till en osäker efterfrågan på klimateffektiva produkter analyseras.

    • Background: Chronic sickness behavior is implicated in ME/CFS (Myalgic Encephalomyelitis/Chronic Fatigue Syndrome) and chronic pain but the level of subjective sickness behavior in these conditions has not been investigated or compared to other clinical and non-clinical samples, or to the level in experimental inflammation. Furthermore, the relationship between sickness behavior and self-rated health and functioning is not known in patients with ME/CFS and chronic pain. The aim of the present study was to investigate how sickness behavior in patients with chronic conditions differs from that in individuals with experimental acute sickness, primary care patients, the general population and healthy subjects. In addition, we wanted to explore how sickness behavior is related to self-rated health and health-related functioning.Methods: Sickness behavior was quantified using the sickness questionnaire (SicknessQ). Self-ratings were collected at one time-point in 6 different samples. Levels of sickness behavior in patients with ME/CFS (n ​= ​38) and patients with chronic pain (n ​= ​190) were compared to healthy subjects with lipopolysaccharide(LPS)-induced inflammation (n ​= ​29), primary care patients (n ​= ​163), individuals from the general population (n ​= ​155) and healthy subjects (n ​= ​48), using linear regression. Correlations and moderated regression analyses were used to investigate associations between sickness behavior and self-rated health and health-related functioning in ME/CFS, chronic pain and the general population.Results: LPS-injected individuals (M ​= ​16.3), patients with ME/CFS (M ​= ​16.1), chronic pain (M ​= ​16.1) and primary care patients (M ​= ​10.7) reported significantly higher SicknessQ scores than individuals from the general population (M ​= ​5.4) and healthy subjects (M ​= ​3.6) all p’s ​< ​0.001). In turn, LPS-injected individuals, patients with ME/CFS and chronic pain reported significantly higher SicknessQ scores than primary care patients (p’s ​< ​0.01). Higher levels of sickness behavior were associated with poorer self-rated health and health-related functioning (p’s ​< ​0.01), but less so in patients with ME/CFS and chronic pain than in individuals from the general population.Conclusions: Patients with ME/CFS and chronic pain report similar high levels of sickness behavior; higher than primary care patients, and comparable to levels in experimental inflammation. Further study of sickness behavior in ME/CFS and chronic pain populations is warranted as immune-to-brain interactions and sickness behavior may be of importance for functioning as well as in core pathophysiological processes in subsets of patients.

    • Cerebrospinal fluid (CSF) biomarkers reflect brain pathophysiology and are used extensively in translational research as well as in clinical practice for diagnosis of neurological diseases, e.g., Alzheimer's disease (AD). However, CSF biomarker concentrations may be influenced by non-disease related inter-individual variability. Here we use a data-driven approach to demonstrate the existence of inter-individual variability in mean standardized CSF protein levels. We show that these non-disease related differences cause many commonly reported CSF biomarkers to be highly correlated, thereby producing misleading results if not accounted for. To adjust for this inter-individual variability, we identified and evaluated high-performing reference proteins which improved the diagnostic accuracy of key CSF AD biomarkers. Our reference protein method attenuates the risk for false positive findings, and improves the sensitivity and specificity of CSF biomarkers, with broad implications for both research and clinical practice.

  • Baseline Pro-Inflammatory Cytokine Levels Moderate Psychological Inflexibility in Behavioral Treatment for Chronic Pain

    • Background: The medical and scientific communities struggle to understand chronic pain and find effective treatments. Multimodal approaches are encouraging but show significant individual differences. Methods: Seventy-eight persons (56 women) with chronic pain received Acceptance and Commitment Therapy and provided blood samples before and after treatment. The participants completed surveys with the blood sampling. Blood plasma was analyzed for IL-6 and TNF-α levels with the Olink Inflammation Panel (Olink Bioscience Uppsala, Sweden). The treatment effects and moderating effects of low-grade inflammation on changes in outcomes were analyzed using linear mixed models. Results: Pain interference (p < 0.001) and psychological inflexibility (p < 0.001) improved significantly during treatment, but pain intensity did not (p = 0.078). Cytokine levels did not change over the course of the treatment (IL-6/TNF-α p = 0.086/0.672). Mean baseline levels of IL-6 and TNF-α moderated improvement in psychological inflexibility during the course of treatment (p = 0.044), but cytokine levels did not moderate changes in pain interference (p = 0.205) or pain intensity (p = 0.536). Conclusions: Higher baseline inflammation levels were related to less improvement in psychological inflexibility. Low-grade inflammation may be one factor underlying the variability in behavioral treatment in chronic pain.

  • Inflammatory Blood Signature Related to Common Psychological Comorbidity in Chronic Pain

    • Chronic pain is characterized by high psychological comorbidity, and diagnoses are symptom-based due to a lack of clear pathophysiological factors and valid biomarkers. We investigate if inflammatory blood biomarker signatures are associated with pain intensity and psychological comorbidity in a mixed chronic pain population. Eighty-one patients (72% women) with chronic pain (>6 months) were included. Patient reported outcomes were collected, and blood was analyzed with the Proseek Multiplex Olink Inflammation Panel (Bioscience Uppsala, Uppsala, Sweden), resulting in 77 inflammatory markers included for multivariate data analysis. Three subgroups of chronic pain patients were identified using an unsupervised principal component analysis. No difference between the subgroups was seen in pain intensity, but differences were seen in mental health and inflammatory profiles. Ten inflammatory proteins were significantly associated with anxiety and depression (using the Generalized Anxiety Disorder 7-item scale (GAD-7) and the Patient Health Questionnaire (PHQ-9): STAMBP, SIRT2, AXIN1, CASP-8, ADA, IL-7, CD40, CXCL1, CXCL5, and CD244. No markers were related to pain intensity. Fifteen proteins could differentiate between patients with moderate/high (GAD-7/PHQ-9 > 10) or mild/no (GAD-7/PHQ-9 < 10) psychological comorbidity. This study further contributes to the increasing knowledge of the importance of inflammation in chronic pain conditions and indicates that specific inflammatory proteins may be related to psychological comorbidity.
