    • BACKGROUND/AIM: The survival rate of women diagnosed with triple-negative breast-cancer (TNBC) remains low. Hence, this study aimed at the chemical and biological optimization of furanosteroid derivatives for the treatment of this type of malignancy using TNBC cells.MATERIALS AND METHODS: Semi-synthetic analogs of wortmannolone (1-6) that negatively affected the aberrant pathways in tumor cells were evaluated in hormone-independent breast cancer cells using western blot and cell-cycle analysis.RESULTS: Wortmannolone derivatization generated NF-ĸB inhibitors as new lead structures for further development. Compound (3) was found to be the most significantly active lead.CONCLUSION: Structure-activity analysis in the present study showed that acetylation of the hydroxyl groups and substitution on C3 and C17 of wortmannolone enhanced biological activity. Alpha-substitution of the acetyl group in C3 on ring A (compound 3) resulted in ROS inducing effect; however, presence of an acetyl group in β-position of C3 displayed the highest NF-ĸB p65 inhibitory activity (0.60 μM).

    • A tin oxide (SnO2) nanostructure was prepared using Matricaria recutita leaf extract to investigate its anticancer activity against SK-MEL-28 cells. The tetragonal crystal structure of tin oxide nanoparticles with an average crystal size of 27 nm was confirmed by X-ray diffraction (XRD) analysis. The tetragonal crystal structure of the tin oxide nanoparticles, with an average crystallite size of 27 nm, was confirmed by XRD an absorbance peak at 365 nm was identified by UV-visible spectroscopy analysis as belonging to the bio-mediated synthesis of SnO2 nanoparticles. The SnO2 NPs are capped and stabilized with diverse functional groups derived from bioactive molecules, including aldehydes, benzene rings, amines, alcohols, and carbonyl stretch protein molecules. Fourier transform infrared spectroscopy (FTIR) analysis validated the presence of these capping and stabilizing chemical bonds. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) studies revealed the cauliflower-shaped morphology of the SnO2 nanoparticles with an average particle size of 28 nm. The antimicrobial activity of both prepared and encapsulated samples confirmed their biological activities. Furthermore, both prepared and encapsulated tin oxide samples exhibited excellent anticancer activity against SK-MEL-28 human cancer cells. The present study introduces a reliable and uncomplicated approach to produce SnO2 nanoparticles and demonstrates their effectiveness in various applications, including cancer therapy, drug administration, and disinfectant.

    • In this study, we describe the anion recognition ability of isatin-3-thiosemicarbazone 2, which contains two different anion recognition units i.e. isatin NH and the thiourea moiety. Both have the ability to act as proton donors. Most importantly, a significant colour change of 2 was observed (from light yellow to reddish orange) in organic medium only after the addition of the F– anion. No such colour change could be observed for any other anions including Cl–, Br–, I–, H2PO4−,NO2−,PF4−etc. The UV-Vis spectroscopic studies also indicate the potential of this compound for selective detection of fluoride anions. 1H-NMR titrations clearly indicate the formation of the 2.F– anionic complex. The Density-functional theory (DFT) calculations are also performed to get further insights on the formation of 2.F– complex.

  • Antioxidant and Organic Dye Removal Potential of Cu-Ni Bimetallic Nanoparticles Synthesized Using Gazania rigens Extract

    • Copper-nickel bimetallic nanoparticles (Cu-Ni BNPs) were fabricated using an eco-friendly green method of synthesis. An extract of synthesized Gazania rigens was used for the synthesis of BNPs followed by characterization employing different techniques including UV/Vis spectrophotometer, FTIR, XRD, and SEM. Spectrophotometric studies (UV-Vis and FTIR) confirmed the formation of bimetallic nanoparticles. The SEM studies indicated that the particle size ranged from 50 to 100 nm. Analysis of the BNPs by the XRD technique confirmed the presence of both Cu and Ni crystal structure. The synthesized nanoparticles were then tested for their catalytic potential for photoreduction of methylene blue dye in an aqueous medium and DPPH radical scavenging in a methanol medium. The BNPs were found to be efficient in the reduction of methylene blue dye as well as the scavenging of DPPH free radicals such that the MB dye was completely degraded in just 17 min at the maximum absorption of 660 nm. Therefore, it is concluded that Cu-Ni BNPs can be successfully synthesized using Gazania rigens extract with suitable size and potent catalytic and radical scavenging activities.
