  • Genome-wide association study and functional characterization identifies candidate genes for insulin-stimulated glucose uptake

    • Distinct tissue-specific mechanisms mediate insulin action in fasting and postprandial states. Previous genetic studies have largely focused on insulin resistance in the fasting state, where hepatic insulin action dominates. Here we studied genetic variants influencing insulin levels measured 2 h after a glucose challenge in >55,000 participants from three ancestry groups. We identified ten new loci (P < 5 × 10-8) not previously associated with postchallenge insulin resistance, eight of which were shown to share their genetic architecture with type 2 diabetes in colocalization analyses. We investigated candidate genes at a subset of associated loci in cultured cells and identified nine candidate genes newly implicated in the expression or trafficking of GLUT4, the key glucose transporter in postprandial glucose uptake in muscle and fat. By focusing on postprandial insulin resistance, we highlighted the mechanisms of action at type 2 diabetes loci that are not adequately captured by studies of fasting glycemic traits.

  • Simulation of shock-induced melting of Ni using molecular dynamics coupled to a two-temperature model

    • Using nonequilibrium molecular dynamics (MD) simulations we study shock-induced melting in Ni with an embedded atom method (EAM). Dynamic melting is probed by the pair correlation function, and we find a melting lattice temperature of T-melt=6400 +/- 300 K for a melting pressure of P-melt=275 +/- 10 GPa. When a combined MD+TTM (two-temperature model) approach is used to include electronic heat conduction and electron-phonon coupling, P-melt and T-melt change. For a given pressure, the temperature behind the shock decreases due to electronic heat diffusion into the cold, unshocked material. This cooling of the material behind the shock slightly increases the melting pressure compared to simulations without electronic heat conduction and electron-phonon coupling. The decrease in the temperature behind the shock front is enhanced if the electron-phonon coupling is artificially made larger. We also explore the feasibility of using x-ray diffraction to detect melting.

    • We present the first ever Compton scattering study on WxCu1-x(x=0.60, 0.72) alloys. The Compton profile measurements have been made using 20Ci (CS)-C-137 gamma-ray source. The experimental data are compared with the superposition of APW-based Compton profiles of constituent metals. A schematic study on charge transfer has been reported using the experimental valence band Compton profiles of both the alloys, W and Cu. Our first ever data support the charge transfer from W to Cu on alloying, which is also confirmed by our band structure calculations employing exact muffin-tin orbitals method (EMTO).   • High-pressure melting of MgSiO3

    • The melting curve of MgSiO3 perovskite has been determined by means of ab initio molecular dynamics complemented by effective pair potentials, and a new phenomenological model of melting. Using first principles ground state calculations, we find that the MgSiO3 perovskite phase transforms into post perovskite at pressures above 100 GPa, in agreement with recent theoretical and experimental studies. We find that the melting curve of MgSiO3, being very steep at pressures below 60 GPa, rapidly flattens on increasing pressure. The experimental controversy on the melting of the MgSiO3 perovskite at high pressures is resolved, confirming the data by Zerr and Boehler.

    • In this study, amorphous TiNi phase was successfully prepared using mechanically milling for a very short time of 8 h. HRTEM confirms the formation of amorphous phase with the presence of nanocrystalline Fe particles. After hydrogenation (30 bars of H-2 for a duration of 2 h), the electrochemical reaction shows that TiNi hydride/Li cell discharges at a current of one Li for 10 h between 3 V and 0.005 V. The discharge of TiNiH electrode around x = 1 Li corresponds to a capacity of 251 mAh g(-1) and a hydride composition of TiNiH1.0 at an average voltage of 0.4 V. Ex-situ X-ray diffraction pattern collected at the end of the discharge shows a mixture of amorphous TiNi compound and LiH. A general mechanism for the electrochemical reaction is then proposed: alpha-TiNiH + Li+ + e(-) -> alpha-TiNi + LiH. The results from DFT calculations yield an average cell voltage of 0.396 V, which is in good agreement with the experimental pseudo-plateau occurring at 0.4 V.

    • BACKGROUND AND AIMS: Clinical trials evaluating biologics and small molecules in patients with ulcerative colitis are predominantly excluding ulcerative proctitis. The objective of the PROCTRIAL (Definition and endpoints for ulcerative PROCtitis in clinical TRIALs) initiative was to develop consensus statements for definitions, inclusion criteria, and endpoints for the evaluation of ulcerative proctitis in adults.METHODS: Thirty-five international experts held a consensus meeting to define ulcerative proctitis, and the endpoints to use in clinical trials. Based on a systematic review of the literature, statements were generated, discussed, and approved by the working group participants using a modified Delphi method. Consensus was defined as at least 75% agreement among voters.RESULTS: The group agreed that the diagnosis of ulcerative proctitis should be made by ileocolonoscopy and confirmed by histopathology, with the exclusion of infections, drug-induced causes, radiation, trauma, and Crohn's disease. Ulcerative proctitis was defined as macroscopic extent of lesions limited to 15 cm distance from the anal verge in adults. Primary and secondary endpoints were identified to capture response of ulcerative proctitis to therapy. A combined clinical and endoscopic primary endpoint for the evaluation of ulcerative proctitis disease activity is proposed. Secondary endpoints which should be evaluated include endoscopic remission, histological remission, mucosal healing, histologic endoscopic mucosal improvement, disability, fecal incontinence, urgency, constipation, and health-related quality of life.CONCLUSION: In response to the need for guidance on the design of clinical trials in patients with ulcerative proctitis, the PROCTRIAL consensus provides recommendations on the definition and endpoints for ulcerative proctitis clinical trials.
