  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Changes in adaptive capacity of Kenyan fishing communities

    • Coastal communities are particularly at risk from the impacts of a changing climate(1). Building the capacity of coastal communities to cope with and recover from a changing environment is a critical means to reducing their vulnerability(2,3). Yet, few studies have quantitatively examined adaptive capacity in such communities. Here, we build on an emerging body of research examining adaptive capacity in natural resource-dependent communities in two important ways. We examine how nine indicators of adaptive capacity vary: among segments of Kenyan fishing communities; and over time. Socially disaggregated analyses found that the young, those who had migrated, and those who do not participate in decision-making seemed least prepared for adapting to change in these resource-dependent communities. These results highlight the most vulnerable segments of society when it comes to preparing for and adapting to change in resource-dependent communities. Comparisons through time showed that aspects of adaptive capacity seemed to have increased between 2008 and 2012 owing to higher observed community infrastructure and perceived availability of credit.

  • Four ways blue foods can help achieve food system ambitions across nations

    • Blue foods, sourced in aquatic environments, are important for the economies, livelihoods, nutritional security and cultures of people in many nations. They are often nutrient rich1, generate lower emissions and impacts on land and water than many terrestrial meats2, and contribute to the health3, wellbeing and livelihoods of many rural communities4. The Blue Food Assessment recently evaluated nutritional, environmental, economic and justice dimensions of blue foods globally. Here we integrate these findings and translate them into four policy objectives to help realize the contributions that blue foods can make to national food systems around the world: ensuring supplies of critical nutrients, providing healthy alternatives to terrestrial meat, reducing dietary environmental footprints and safeguarding blue food contributions to nutrition, just economies and livelihoods under a changing climate. To account for how context-specific environmental, socio-economic and cultural aspects affect this contribution, we assess the relevance of each policy objective for individual countries, and examine associated co-benefits and trade-offs at national and international scales. We find that in many African and South American nations, facilitating consumption of culturally relevant blue food, especially among nutritionally vulnerable population segments, could address vitamin B12 and omega-3 deficiencies. Meanwhile, in many global North nations, cardiovascular disease rates and large greenhouse gas footprints from ruminant meat intake could be lowered through moderate consumption of seafood with low environmental impact. The analytical framework we provide also identifies countries with high future risk, for whom climate adaptation of blue food systems will be particularly important. Overall the framework helps decision makers to assess the blue food policy objectives most relevant to their geographies, and to compare and contrast the benefits and trade-offs associated with pursuing these objectives.

  • Towards a typology of interactions between small-scale fisheries and global seafood trade

    • Fish and fish-related products are among the most highly traded commodities globally and the proportion of globally harvested fish that is internationally traded has steadily risen over time. Views on the benefits of international seafood trade diverge, partly as a result from adopting either an aggregate national focus or a focus on local market actors. However, both views generally assume that the trade in question is characterized by export of fisheries resources to international markets. This is potentially misleading as empirical evidence suggests that import of seafood can also have impacts on local SSF dynamics. A systematic analysis of the different ways in which local production systems connect to international seafood markets can therefore help shed more light on why small-scale fisheries exhibit such differences in outcomes as they engage in an increasingly global seafood trade. This paper conducts a synthesis across 24 cases from around the world and develops a typology of small-scale fisheries and how they connect to and interact with international seafood trade. The analysis is based on key features drawn from trade theory regarding how trade interacts with local production. The implications of the findings for social and ecological sustainability of small-scale fisheries are discussed with the aim of identifying further research topics which deserve attention to better inform trade policy for more sustainable fisheries and more just wealth distribution from their trade.

  • Humans and Seagrasses in East Africa : A social-ecological systems approach

    • The present study is one of the first attempts to analyze the societal importance of seagrasses (marine flowering plants) from a Natural Resource Management perspective, using a social-ecological systems (SES) approach. The interdisciplinary study takes place in East Africa (Western Indian Ocean, WIO) and includes in-depth studies in Chwaka Bay, Zanzibar, Tanzania. Natural and social sciences methods were used. The results are presented in six articles, showing that seagrass ecosystems are rich in seagrass species (13) and form an important part of the SES within the tropical seascape of the WIO. Seagrasses provide livelihoods opportunities and basic animal protein, in from of seagrass associated fish e.g. Siganidae and Scaridae. Research, management and education initiatives are, however, nearly non-existent. In Chwaka Bay, the goods and ecosystem services associated with the meadows and also appreciated by locals were fishing and collection grounds as well as substrate for seaweed cultivation. Seagrasses are used as medicines and fertilizers and associated with different beliefs and values. Dema (basket trap) fishery showed clear links to seagrass beds and provided the highest gross income per capita of all economic activities. All showing that the meadows provide social-ecological resilience. Drag-net fishery seems to damage the meadows. Two ecological studies show that artisanal seaweed farming of red algae, mainly done by women and pictured as sustainable in the WIO, has a thinning effect on seagrass beds, reduces associated macrofauna, affects sediments, changes fish catch composition and reduces diversity. Furthermore, it has a negative effect on i.a. women’s health. The two last papers are institutional analyses of the human-seagrass relationship. A broad approach was used to analyze regulative, normative and cultural-cognitive institutions. Cooperation and conflict take place between different institutions, interacting with their slow or fast moving characteristics, and are thus fundamental in directing the system into sustainable/unsustainable paths. Ecological knowledge was heterogeneous and situated. Due to the abundance of resources and high internal control, the SES seems to be entangled in a rigidity trap with the risk of falling into a poverty trap. Regulations were found insufficient to understand SES dynamics. “Well” designed organizational structures for management were found insufficient for “good” institutional performance. The dynamics between individuals embedded in different social and cultural structures showed to be crucial. Bwana Dikos, monitoring officials, placed in villages or landing sites in Zanzibar experienced four dilemmas – kinship, loyalty, poverty and control – which decrease efficiency and affect resilience. Mismatches between institutions themselves, and between institutions and cognitive capacities were identified. Some important practical implications are the need to include seagrass meadows in management and educational plans, addressing a seascape perspective, livelihood diversification, subsistence value, impacts, social-ecological resilience, and a broad institutional approach.

  • Circulating Folate, Vitamin B6, and Methionine in Relation to Lung Cancer Risk in the Lung Cancer Cohort Consortium (LC3)

    • Background: Circulating concentrations of B vitamins and factors related to one-carbon metabolism have been found to be strongly inversely associated with lung cancer risk in the European Prospective Investigation into Cancer and Nutrition (EPIC) study. The extent to which these associations are present in other study populations is unknown.Methods: Within 20 prospective cohorts from the National Cancer Institute Cohort Consortium, a nested case-control study was designed including 5364 incident lung cancer case patients and 5364 control subjects who were individually matched to case patients by age, sex, cohort, and smoking status. Centralized biochemical analyses were performed to measure circulating concentrations of vitamin B6, folate, and methionine, as well as cotinine as an indicator of recent tobacco exposure. The association between these biomarkers and lung cancer risk was evaluated using conditional logistic regression models.Results: Participants with higher circulating concentrations of vitamin B6 and folate had a modestly decreased risk of lung cancer risk overall, the odds ratios when comparing the top and bottom fourths (OR 4vs1 ) being 0.88 (95% confidence interval [CI] = 0.78 to 1.00) and 0.86 (95% CI = 0.74 to 0.99), respectively. We found stronger associations among men (vitamin B6: OR 4vs1 = 0.74, 95% CI = 0.62 to 0.89; folate: OR 4vs1 = 0.75, 95% CI = 0.61 to 0.93) and ever smokers (vitamin B6: OR 4vs1 = 0.78, 95% CI = 0.67 to 0.91; folate: OR 4vs1 = 0.87, 95% CI = 0.73 to 1.03). We further noted that the association of folate was restricted to Europe/Australia and Asia, whereas no clear association was observed for the United States. Circulating concentrations of methionine were not associated with lung cancer risk overall or in important subgroups.Conclusions: Although confounding by tobacco exposure or reverse causation cannot be ruled out, these study results are compatible with a small decrease in lung cancer risk in ever smokers who avoid low concentrations of circulating folate and vitamin B6.

    • Vitamin B supplementation can have side effects for human health, including cancer risk. We aimed to elucidate the role of vitamin B12 in lung cancer etiology via direct measurements of pre‐diagnostic circulating vitamin B12 concentrations in a nested case–control study, complemented with a Mendelian randomization (MR) approach in an independent case–control sample. We used pre‐diagnostic biomarker data from 5183 case–control pairs nested within 20 prospective cohorts, and genetic data from 29,266 cases and 56,450 controls. Exposures included directly measured circulating vitamin B12 in pre‐diagnostic blood samples from the nested case–control study, and 8 single nucleotide polymorphisms associated with vitamin B12 concentrations in the MR study. Our main outcome of interest was increased risk for lung cancer, overall and by histological subtype, per increase in circulating vitamin B12 concentrations. We found circulating vitamin B12 to be positively associated with overall lung cancer risk in a dose response fashion (odds ratio for a doubling in B12 [ORlog2B12] = 1.15, 95% confidence interval (95%CI) = 1.06–1.25). The MR analysis based on 8 genetic variants also indicated that genetically determined higher vitamin B12 concentrations were positively associated with overall lung cancer risk (OR per 150 pmol/L standard deviation increase in B12 [ORSD] = 1.08, 95%CI = 1.00–1.16). Considering the consistency of these two independent and complementary analyses, these findings support the hypothesis that high vitamin B12 status increases the risk of lung cancer.

    • Increasing the production of food from the ocean is seen as a pathway toward more sustainable and healthier human diets. Yet this potential is being overshadowed by competing uses of ocean resources in an accelerating blue economy.'' The current emphasis on production growth, rather than equitable distribution of benefits, has created three unexamined or flawed assumptions that growth in the blue economy will lead to growth in blue food'' production, increased production will inevitably lead to improved food and nutrition security, and mariculture production will replace marine capture fisheries. In this perspective, we argue that if research and policies are pursued without addressing these blind spots,'' blue food contributions to reducing hunger and malnutrition, and to meeting the Sustainable Development Goals, will be limited. Taking a broader food-system approach beyond production to also considering food access, affordability, and consumption will refocus the blue food agenda on making production and consumption more equitable and sustainable while increasing access for those who need it most.
