    • Genetic discoveries of Alzheimer’s disease are the drivers of our understanding, and together with polygenetic risk stratification can contribute towards planning of feasible and efficient preventive and curative clinical trials. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer’s disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer’s disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer’s disease. © 2021, The Author(s).

    • This chapter presents the methodology employed for the Integrated Socio-Economic Assessment (MISEA) of different designs of Multi-Use Offshore Platforms (MUOPs). The methodology allows for the identification, the valuationand the assessment of the potential impacts and their magnitude. The analysis considers a number of feasible designs of MUOP investments, and the likely responsesof those impacted by the investment project. The approach provides decision-makers with a valuable tool to assess whether a MUOP project increases the overall social welfare and hence should be undertaken. This is performed under alternative specifications regarding platform design, the discount rate and the stream of net benefits, if a Cost-Benefit Analysis (CBA) is to be followed or a sensitivity analysis of selected criteria in a Multi-Criteria Decision Analysis (MCDA) framework. Themethodology can support the implementation of policies aiming at achieving a goodenvironmental status of the EU’s marine waters and the protection of the resource base upon which marine-related economic and social activities depend.

    • The Antikythera Mechanism is the most sophisticated extant ancient astronomical instrument and analogue computer known and was assembled sometime between 150 and 100 BCE, almost a century after the death of Archimedes. The mechanism has a great educational potential as it appeals to inquiring minds as an astonishing artefact of science and technology. The latest research findings reveal significant cultural and social functions in its operations. This astonishing astronomical instrument has a clear interdisciplinary valueand it has that it may be used as an educational medium, to engage the general public, and especially to attract students both to/from exact sciences and to/from the humanities. The astronomical and technical knowledge embedded in the mechanism can also be used to introduce some aspects of modern science through the unknown technological achievements of Hellenic antiquity.

  • Differential expansion of circulating human MDSC subsets in patients with cancer, infection and inflammation

    • Myeloid-derived suppressor cells (MDSC) are a functional myeloid cell subset that includes myeloid cells with immune suppressive properties. The presence of MDSC has been reported in the peripheral blood of patients with several malignant and non-malignant diseases. So far, direct comparison of MDSC across different diseases and Centers is hindered by technical pitfalls and a lack of standardized methodology. To overcome this issue, we formed a network through the COST Action Mye-EUNITER (www.mye-euniter.eu) with the goal to standardize and facilitate the comparative analysis of human circulating MDSC in cancer, inflammation and infection. In this manuscript, we present the results of the multicenter study Mye-EUNITER MDSC Monitoring Initiative, that involved 13 laboratories and compared circulating MDSC subsets across multiple diseases, using a common protocol for the isolation, identification and characterization of these cells.MethodsWe developed, tested, executed and optimized a standard operating procedure for the isolation and immunophenotyping of MDSC using blood from healthy donors. We applied this procedure to the blood of almost 400 patients and controls with different solid tumors and non-malignant diseases. The latter included viral infections such as HIV and hepatitis B virus, but also psoriasis and cardiovascular disorders.ResultsWe observed that the frequency of MDSC in healthy donors varied substantially between centers and was influenced by technical aspects such as the anticoagulant and separation method used. Expansion of polymorphonuclear (PMN)-MDSC exceeded the expansion of monocytic MDSC (M-MDSC) in five out of six solid tumors. PMN-MDSC expansion was more pronounced in cancer compared with infection and inflammation. Programmed death-ligand 1 was primarily expressed in M-MDSC and e-MDSC and was not upregulated as a consequence of disease. LOX-1 expression was confined to PMN-MDSC.ConclusionsThis study provides improved technical protocols and workflows for the multi-center analysis of circulating human MDSC subsets. Application of these workflows revealed a predominant expansion of PMN-MDSC in solid tumors that exceeds expansion in chronic infection and inflammation.

    • Using simple symmetry arguments we classify the ungauged D = 4, N = 2 supergravity theories, coupled to both vector and hyper multiplets through homogeneous scalar manifolds, that can be built as the product of N = 2 and N= 0 matter-coupled Yang-Mills gauge theories. This includes all such supergravities with two isolated exceptions: pure supergravity and the T-3 model.

    • We consider "twin supergravities"-pairs of supergravities with N+ and N- supersymmetries, N+ > N-, with identical bosonic sectors-in the context of tensoring super Yang-Mills multiplets. It is demonstrated that the pairs of twin supergravity theories are related through their left and right super YangMills factors. This procedure generates newtheories from old ones. In particular, the matter coupled N(-)twins in D = 3, 5, 6 and the N- = 1 twins inD = 4 have not, as far as we are aware, been obtained previously using the double-copy construction, adding to the growing list of double-copy constructible theories. The use of fundamental matter multiplets in the double-copy construction leads us to introduce a bifundamental scalar that couples to the well-known biadjoint scalar field. It is also shown that certain matter coupled supergravities admit more than one factorization into left and right super Yang-Mills-matter theories.

  • Multimodal conversational interaction with a humanoid robot

    • The paper presents a multimodal conversational interaction system for the Nao humanoid robot. The system was developed at the 8th International Summer Workshop on Multimodal Interfaces, Metz, 2012. We implemented WikiTalk, an existing spoken dialogue system for open-domain conversations, on Nao. This greatly extended the robot's interaction capabilities by enabling Nao to talk about an unlimited range of topics. In addition to speech interaction, we developed a wide range of multimodal interactive behaviours by the robot, including face-tracking, nodding, communicative gesturing, proximity detection and tactile interrupts. We made video recordings of user interactions and used questionnaires to evaluate the system. We further extended the robot's capabilities by linking Nao with Kinect.
