    • During the transition from a healthy state to cardiometabolic disease, patients become heavily medicated, which leads to an increasingly aberrant gut microbiome and serum metabolome, and complicates biomarker discovery1–5. Here, through integrated multi-omics analyses of 2,173 European residents from the MetaCardis cohort, we show that the explanatory power of drugs for the variability in both host and gut microbiome features exceeds that of disease. We quantify inferred effects of single medications, their combinations as well as additive effects, and show that the latter shift the metabolome and microbiome towards a healthier state, exemplified in synergistic reduction in serum atherogenic lipoproteins by statins combined with aspirin, or enrichment of intestinal Roseburia by diuretic agents combined with beta-blockers. Several antibiotics exhibit a quantitative relationship between the number of courses prescribed and progression towards a microbiome state that is associated with the severity of cardiometabolic disease. We also report a relationship between cardiometabolic drug dosage, improvement in clinical markers and microbiome composition, supporting direct drug effects. Taken together, our computational framework and resulting resources enable the disentanglement of the effects of drugs and disease on host and microbiome features in multimedicated individuals. Furthermore, the robust signatures identified using our framework provide new hypotheses for drug–host–microbiome interactions in cardiometabolic disease.

    • Microbiota-host-diet interactions contribute to the development of metabolic diseases. Imidazole propionate is a novel microbially produced metabolite from histidine, which impairs glucose metabolism. Here, we show that subjects with prediabetes and diabetes in the MetaCardis cohort from three European countries have elevated serum imidazole propionate levels. Furthermore, imidazole propionate levels were increased in subjects with low bacterial gene richness and Bacteroides 2 enterotype, which have previously been associated with obesity. The Bacteroides 2 enterotype was also associated with increased abundance of the genes involved in imidazole propionate biosynthesis from dietary histidine. Since patients and controls did not differ in their histidine dietary intake, the elevated levels of imidazole propionate in type 2 diabetes likely reflects altered microbial metabolism of histidine, rather than histidine intake per se. Thus the microbiota may contribute to type 2 diabetes by generating imidazole propionate that can modulate host inflammation and metabolism.

  • Cracking and Fatigue of Heavy Loaded Prestressed Concrete Bridge in Sweden

    • A prestressed concrete bridge was built in 1963 with BBRV cables. It has three spans and a total length of 134.8 m. Due to mining activities the bridge was loaded with trucks with a total weight of 90 ton during 2012-2014 and from 2019. Crack development has been monitored manually and from 2020 with strain gauges and LVDTs.Cracks normally vary between 0.1 to 0.3 mm in width and grow in length with time. In November 2020 some of the strain gauges on the concrete showed alarming growth and the bridge was closed for traffic. Additional strain gauges were installed on vertical reinforcement bars and an assessment was carried out of the fatigue capacity of the bridge. It was found that the new strain gauges did not indicate any growth in strain and that the fatigue capacity was sufficient. The bridge could be opened again for traffic after being closed for five weeks. Monitoring drift in the strain gauges and fatigue are discussed.

    • Due to restrictions following the Covid-19 pandemic,major changes regarding teaching have been called for. Sincestudents’ presence in lab classrooms has been very restricted alsophysical labs have had to be transformed into e.g. simulationsperformed at home or via remote connections. After lifting of therestrictions, hybrid learning where both physical and simulatedlab environment are combined have been introduced. This reportdescribes such a case regarding a lab in data communication forelectrical engineering students on bachelor level. The result showsadded value to the learning outcome of the lab.

    • This paper investigates the outcome of the efforts to economically catch up during the so-called development era in French speaking West Africa. An attempt is made to measure and discuss key elements of social capability over the period 1930-1980 in Côte d'Ivoire and Senegal following Moses Abramovitz' interpretation of social capability. The paper distinguishes between four elements of social capability: degree of structural transformation, social and economic inclusion, the state's autonomy and its accountability. We find that there was significant but uneven progress in social capability in both countries during the development era. Despite their differences in economic performance both countries confronted fundamental shared challenges. Most notably, our analysis highlights how persistent lack of broad-based access to economic opportunities played a significant role in disrupting sustained economic and social progress in the two countries. This gives an opportunity to reflect on similarities and differences between the development era and the recent African growth phase.

  • Evidence of a causal and modifiable relationship between kidney function and circulating trimethylamine N-oxide

    • The host-microbiota co-metabolite trimethylamine N-oxide (TMAO) is linked to increased cardiovascular risk but how its circulating levels are regulated remains unclear. We applied "explainable" machine learning, univariate, multivariate and mediation analyses of fasting plasma TMAO concentration and a multitude of phenotypes in 1,741 adult Europeans of the MetaCardis study. Here we show that next to age, kidney function is the primary variable predicting circulating TMAO, with microbiota composition and diet playing minor, albeit significant, roles. Mediation analysis suggests a causal relationship between TMAO and kidney function that we corroborate in preclinical models where TMAO exposure increases kidney scarring. Consistent with our findings, patients receiving glucose-lowering drugs with reno-protective properties have significantly lower circulating TMAO when compared to propensity-score matched control individuals. Our analyses uncover a bidirectional relationship between kidney function and TMAO that can potentially be modified by reno-protective anti-diabetic drugs and suggest a clinically actionable intervention for decreasing TMAO-associated excess cardiovascular risk.

  • Oligothiophene Assemblies Defined by DNA Interaction : From Single Chains to Disordered Clusters

    • The organization of conjugated polyelectrolytes (CPEs) interacting with biomolecules sets conditions for the biodetection of biological processes and identity, through the use of optical emission from the CPE. Herein, a well-defined CPE and its binding to DNA is studied. By using dynamic light scattering and circular dichroism spectroscopy, it is shown that the CPE forms a multimolecule ensemble in aqueous solution that is more than doubled it? size when interacting with a small DNA chain, while single chains are evident in ethanol. The related changes in the fluorescence spectra upon polymer aggregation are assigned to oscillator strength redistribution between vibronic transitions in weakly coupled H-aggregates. An enhanced single-molecule spectroscopy technique that allows full control of excitation and emission light polarization is applied to combed and decorated;,DNA chains. It is found that the organization of combed CPE-lambda DNA complexes (when dry on the surface) allows considerable variation of CPE distances and direction relative to the DNA chain. By analysis of the polarization data. energy transfer between the polymer chains in individual complexes is confirmed and their sizes estimated.

  • Thermal degradation of TiN and TiAlN coatings during rapid laser treatment

    • In this research, a fundamental study was conducted on damage behavior of cathodic arc evaporated TiN and Ti0.44Al0.56N coatings, in terms of oxidation and cracking/spallation, when they were exposed to single-pulse laser treatment in a temperature range of 1200-2100 degrees C. Moreover, a multiple-pulse laser treatment was designed to apply thermo-mechanical loads on the coatings in order to evaluate their thermal degradation during rapid heating/cooling cycles between 200 and 1200 degrees C. Single-pulse treatment of TiN up to 1500 degrees C led to the intercolumnar cracking and formation of ultrafine TiO grains. An increase in temperature up to 2100 degrees C resulted in a notable bulging of the surface, and formation of TiO2 of various morphologies such as grainy structure, dense molten and re-solidified structure, droplets from melt expulsion and, more interestingly, nanofibers. Multiplepulse treatment of TiN was accompanied by a severe cracking and spallation, which divided the surface into two layers: a heavily cracked top layer composed of dense TiO2 grains, and a bottom layer having porous TiO2 grains indicating incomplete oxidation. Conversely, Ti0.44Al0.56N did not show any visible cracking and oxidation after single-pulse treatment. Multiple-pulse treatment did not also yield cracking and spallation for Ti0.44Al0.56N, and its ablated region consisted of TiO2 grains combined with thin Al2O3 platelets. An excellent combination of properties including higher oxidation resistance and greater fracture toughness at high temperatures led to a higher thermal damage resistance for Ti0.44Al0.56N coating compared to TiN when undergoing single- and multiple-pulse laser treatments.
