    • The evolution of CAST’s UDL Guidelines has been a dynamic, collaborative, and developmental process. We shared the first version of the Guidelines—Version 1.0—in 2008. Since that time, we have collected and specifically solicited feedback from the field. This feedback, along with the expanding research in the areas of UDL, education, cognitive science, psychology, and neuroscience, has led us to develop different representations of the Guidelines over time. Each of these representations has a particular goal and traces our learning not only as an organization but as a field more broadly.Version 2.2 (2018)This latest version—Version 2.2—does not represent the “correct” version. Instead, we see this version as yet another representation to more fully explore and articulate the promise of UDL. We have made several structural changes to the graphic organizer to better support educators and other stakeholders who are new to UDL and to more clearly articulate the goal of UDL. About the Graphic Organizer describes these structural changes in more detail, while our FAQ may help answer questions you have about the UDL Guidelines and their relationship to the UDL framework.

  • Preparation of Porous Poly(3-Hydroxybutyrate) Films by Water-Droplet Templating

    • Porous resorbable implants are of great interest since they may deliver bioactives or drugs, facilitate the transport of body fluids or degradation products and provide a favorable environment for cell attachment and growth. In this work we report on a method using concentrated emulsions to template interconnected solid foam materials and to produce highly porous poly(3-hydroxybutyrate) (PHB) materials. Porous PHB films were cast made from water-in-oil template emulsions including Span 80 and lithium sulphate. The films were characterized by SEM-EDX and DMA. The water uptake of the films was recorded in order to determine the fraction water available pores. The results show that the ad-dition of lithium sulphate allows a fine tuning of the film morphology with respect to porosity and interconnected po-rous structure. The film porosity was determined to 51% ± 3%, 52% ± 3% and 45% ± 3% for the films made with 0%, 2.9% and 14.3% lithium sulphate in the template emulsion, respectively. The fraction water available pores was sig-nificantly lower, 11% ±3%, 38% ±12% and 48% ± 7% for films with 0%, 2.9% and 14.3% litium sulphate respectively. Differences in fraction water available pores and total porosity for the films reflects the film morphology and differ-ences in pore interconnection.

    • Syftet med denna rapport är att illustrera hur ett praktiskt inriktat ramverk, tekno- logiska innovationssystem (TIS), kan användas av analytiker och beslutsfattare vid departement och myndigheter för att analysera strategiskt viktiga teknikområden ?????????????????????????????????????????????????????????????????????????????I rapporten analyseras fem TIS centrerade kring havsbaserad vindkraft, marin energi, ????????????????????????????????????????????????????????????????????????????????????? systemsvagheter som bromsar områdets vidare utveckling, vilka som kan åtgärdas av systemets aktörer och vilka som motiverar särskilda politiska åtaganden. Rapporten utgör därmed ett underlag för att formulera åtgärder för att åstadkomma ökad innova- tion, teknikspridning och industrialisering inom ovan nämnda teknikområden.Studien har även möjliggjort en jämförande analys av likheter och skillnader ???????????????????????????????????????????????????????????????????????????????? ?????????????????????????????????????????????????????????????????????????????????????? mellan områdena – de är starka respektive svaga av olika orsaker.   • Fracture risk after three bariatric surgery procedures in Swedish obese subjects : up to 26 years follow-up of a controlled intervention study

    • Background: Previous studies have reported an increased fracture risk after bariatric surgery. Objective: To investigate the association between different bariatric surgery procedures and fracture risk. Methods: Incidence rates and hazard ratios for fracture events were analysed in the Swedish Obese Subjects study; an ongoing, nonrandomized, prospective, controlled intervention study. Hazard ratios were adjusted for risk factors for osteoporosis and year of inclusion. Information on fracture events were captured from the Swedish National Patient Register. The current analysis includes 2007 patients treated with bariatric surgery (13.3% gastric bypass, 18.7% gastric banding, and 68.0% vertical banded gastroplasty) and 2040 control patients with obesity matched on group level based on 18 variables. Median follow-up was between 15.1 and 17.9 years for the different treatment groups. Results: During follow-up, the highest incidence rate for first-time fracture was observed in the gastric bypass group (22.9 per 1000 person-years). The corresponding incidence rates were 10.4, 10.7 and 9.3 per 1000 person-years for the vertical banded gastroplasty, gastric banding and control groups, respectively. The risk of fracture was increased in the gastric bypass group compared with the control group (adjusted hazard ratio [adjHR] 2.58; 95% confidence interval [CI] 2.02–3.31; P < 0.001), the gastric banding group (adjHR 1.99; 95%CI 1.41–2.82; P < 0.001), and the vertical banded gastroplasty group (adjHR 2.15; 95% CI 1.66–2.79; P < 0.001). Conclusions: The risk of fracture is increased after gastric bypass surgery. Our findings highlight the need for long-term follow-up of bone health for patients undergoing this treatment.

  • Macrophage Gene Expression in Adipose Tissue is Associated with Insulin Sensitivity and Serum Lipid Levels Independent of Obesity.

    • Objective: Obesity is linked to both increased metabolic disturbances and increased adipose tissue macrophage infiltration. However, whether macrophage infiltration directly influences human metabolism is unclear. The aim of this study was to investigate if there are obesity-independent links between adipose tissue macrophages and metabolic disturbances. Design and Methods: Expression of macrophage markers in adipose tissue was analyzed by DNA microarrays in the SOS Sib Pair study and in patients with type 2 diabetes and a BMI-matched healthy control group. Results: The expression of macrophage markers in adipose tissue was increased in obesity and associated with several metabolic and anthropometric measurements. After adjustment for BMI, the expression remained associated with insulin sensitivity, serum levels of insulin, C-peptide, high density lipoprotein cholesterol (HDL-cholesterol) and triglycerides. In addition, the expression of most macrophage markers was significantly increased in patients with type 2 diabetes compared to the control group. Conclusion: Our study shows that infiltration of macrophages in human adipose tissue, estimated by the expression of macrophage markers, is increased in subjects with obesity and diabetes and associated with insulin sensitivity and serum lipid levels independent of BMI. This indicates that adipose tissue macrophages may contribute to the development of insulin resistance and dyslipidemia.

  • Grazer-induced transcriptomic and metabolomic response of the chain-forming diatom Skeletonema marinoi

    • Diatoms and copepods are main actors in marine food webs. The prey-predator interactions between them affect bloom dynamics, shape marine ecosystems and impact the energy transfer to higher trophic levels. Recently it has been demonstrated that the presence of grazers may affect the diatom prey beyond the direct effect of grazing. Here, we investigated the response of the chain-forming centric diatom Skeletonema marinoi to grazer cues, including changes in morphology, gene expression and metabolic profile. S. marinoi cells were incubated with Calanus finmarchicus or with Centropages typicus and in both cases responded by reducing the chain length, whereas changes in gene expression indicated an activation of stress response, changes in the lipid and nitrogen metabolism, in cell cycle regulation and in frustule formation. Transcripts linked to G protein-coupled receptors and to nitric oxide synthesis were differentially expressed suggesting involvement of these signalling transduction pathways in the response. Downregulation of a lipoxygenase in the transcriptomic data and of its products in the metabolomic data also indicate an involvement of oxylipins. Our data contribute to a better understanding of the gene function in diatoms, providing information on the nature of genes implicated in the interaction with grazers, a crucial process in marine ecosystems.

  • Evolution of age-related mutation-driven clonal haematopoiesis over 20 years is associated with metabolic dysfunction in obesity

    • Background Haematopoietic clones caused by somatic mutations with >= 2% variant allele frequency (VAF) increase with age and are linked to risk of haematological malignancies and cardiovascular disease. Recent observations suggest that smaller clones (VAF<2%) are also associated with adverse outcomes. Our aims were to determine the prevalence of clonal haematopoiesis driven by clones of variable sizes in individuals with obesity treated by usual care or bariatric surgery (a treatment that improves metabolic status), and to examine the expansion of clones in relation to age and metabolic dysregulation over up to 20 years.Methods Clonal haematopoiesis-driver mutations (CHDMs) were identified in blood samples from participants of the Swedish Obese Subjects intervention study. Using an ultrasensitive assay, we analysed single-timepoint samples from 1050 individuals treated by usual care and 841 individuals who had undergone bariatric surgery, and multiple-timepoint samples taken over 20 years from a subset (n = 40) of the individuals treated by usual care.Findings In this explorative study, prevalence of CHDMs was similar in the single-timepoint usual care and bariatric surgery groups (20.6% and 22.5%, respectively, P = 0.330), with VAF ranging from 0.01% to 31.15%. Clone sizes increased with age in individuals with obesity, but not in those who underwent bariatric surgery. In the multiple-timepoint analysis, VAF increased by on average 7% (range -4% to 24%) per year and rate of clone growth was negatively associated with HDL-cholesterol (R = -0.68, 1.74 E-04).Interpretation Low HDL-C was associated with growth of haematopoietic clones in individuals with obesity treated by usual care.

  • The Contribution of Hot and Cool Self-Regulation in Early Childhood to Later Academic Achievement

    • The aim of the present study was to investigate the contribution of hot (emotional/motivational) and cool (cognitive) self-regulation in early childhood to lateracademic achievement in kindergarten (math) and late elementary school (math andlanguage). In a sample of 235 preschoolers, higher levels of cool self-regulation predicted better math achievement in kindergarten and this head-start effect persisted into late elementary school. In contrast, higher levels of hot self-regulation predicted better language but not math achievement in late elementary school. These findings support the notion that cool self-regulation is important for the development of academic skills already inkindergarten, whereas the effects on academic achievement of hot self-regulation are delayed, which may be a result of increasing demands through elementary school.

  • Gastrointestinal stromal tumors. Pathogenetic mechanisms, phenotypic characterization and prognosis

    • Gastrointestinal stromal tumor (GIST), the most common non-epithelial neoplasm of the gastrointestinal tract, has historically been problematic both conceptually and clinically. Recently, GIST has been shown to share phenotypic features with the interstitial cells of Cajal (ICC), including the almost uniform expression of the tyrosine kinase receptor KIT. GIST have frequently been found to have activating mutations of the KIT gene or more rarely the platelet derived growth factor receptor alpha (PDGFRA) gene; these apparently play a key role in GIST s pathogenesis. The occurrence of GIST lacking KIT and PDGFRA mutations indicates, however, that alternative pathogenetic mechanisms exist. Clinical studies regarding the correlation of mutations with disease outcome in GIST have been contradictory. Mutation analysis of the KIT gene in 14 GIST revealed exon 11 mutations in 9 tumors. Mutations were detected in benign as well as malignant GIST. Cytogenetic and SKY analyses revealed several recurrent structural and numerical abnormalities; losses of 1p, 14 and 22 being the most common. No correlation was found between biologic behavior, KIT mutation status and chromosome aberrations. All tumors preferentially expressed the shorter, tumorigenic, splice variant of exon 9 of the KIT gene.Imatinib mesylate, a new designer drug, selectively inhibits type III receptor tyrosine kinases and has a dramatic, anti-tumor effect on GIST. In a center-based study of 17 patients with GIST, we studied the relationship between KIT exon 11 mutation status and treatment response. Imatinib was especially effective in tumors with exon 11 mutations - KIT exon 11 mutations were detected in 8 of 9 patients with partial response, whereas no mutations were detected in 3 patients with stable or progressive disease.Earlier studies of receptor tyrosine kinase mutations indicate that different types of mutations are associated with distinctive phenotypes and potentially clinical behavior in GIST. In a large, retrospective, population-based series of GIST from the pre-imatinib mesylate era, we examined whether mutation type correlated with phenotype and clinical course. KIT exon 11 mutations were detected in GIST from 117 of 233 patients (69 deletions, 27 missense mutations and 21 duplications). The deletion subgroup had a significantly decreased disease-free survival. KIT exon 11 deletions were found to be an independent adverse prognostic factor with respect to disease-free survival. GIST have been reported to occasionally occur in patients with neurofibromatosis type I (NF1). NF1 is the most common autosomal dominant inherited disorder, with an incidence of 1/3-4000 individuals. The clinical spectrum of NF1 is extremely wide and includes organic as well as mental manifestations. We analyzed GIST arising in 15 NF1 patients and found that this subset of GIST has a unique phenotype, including an almost uniformly benign appearance histologically, a propensity for multicentricity, association with ICC hyperplasia, predominant location in the small intestine, and a spindle cell morphology as well as benign or very low grade malignant behavior. The NF1-associated GIST also have a unique genotype in that they lack KIT and PDGFRA mutations, indicating that different pathogenetic mechanisms are involved in their evolution.In summary, we have shown that the role of KIT mutations in the biological behavior of GIST is more complex than previously recognized and that the type of mutation influences clinical behavior as well as response to imatinib treatment. NF1-associated GIST are a pheno- and genotypically unique subset of GIST that lack activating mutations in the KIT and PDGFRA genes.
