  • Type I Interferon signature in Nova Scotia duck tolling retriever dogs with steroid responsive meningitis-arteritis

    • Objective: Dogs of the breed Nova Scotia duck tolling retriever (NSDTR) are prone to develop a disease complex in some aspects resembling human systemic lupus erythematosus (SLE). Peripheral blood mononuclear cells (PBMCs) from human SLE patients have an increased mRNA expression type I interferon (IFN) regulated genes. However, it is unknown whether diseased dogs also display the typical type I IFN signature. Methods: To test canine sera for their capacity to induce type I IFN response Mardin-Darby canine kidney (MDCK) cells were cultured with sera from healthy dogs (n=25),  immune-mediated rheumatic disease (IMRD) dogs with anti-nuclear antibodies (ANA+) (n=30) or dogs with steroid responsive meningitis-arteritis (SRMA) (n=25). mRNA expression of the genes MX1, IFIT1 and CXCL10 was measured by quantitative Real Time PCR. Results: A highly significant (p=0.0009) increase in mRNA expression of the type I IFN responsive gene MX1 was detected in cells stimulated by sera from dogs with SRMA, but not from IMRD ANA+ dogs. Expression of IFIT1 was twice as high in cells stimulated by sera from dogs with SRMA compared to both healthy dogs and ANA+ dogs. The mean expression of CXCL10 was nearly ten times higher in cells stimulated by sera from SRMA dogs than by ANA+ dogs and four times higher compared to cells stimulated by control dogs. Conclusion: Presence of type I IFN in sera from diseased NSDTR dogs was found in this study. This implies that this canine model can be used for identification of pathways of importance for autoimmune disorders in humans and for testing of novel therapeutic approaches. Our results can also be a step on the way towards personalized drugs in these dogs.

  • Development and Characterization of Large Area Pressure Sensors and Sitting Posture Monitoring Systems

    • With the emergence of the Internet and rapid development of science and technology over the past few decades, many individuals worldwide now rely on the Internet to conduct daily activities ranging from education, business and creativity to communication and shopping. As we tend to spend more and more time on the Internet and engage less in physical activities, this persistent behaviour could result in some health-related issues within a relatively short period of time. This behaviour, known as sedentary lifestyle, may be related to a higher risk of cardiovascular disease, osteoporosis, obesity, anxiety, pressure ulcers and many other illnesses. As a consequence, there has been great interest in developing non-invasive and unobtrusive measurement techniques for a variety of health care-monitoring applications, such as for blood oxygen saturation, stress levels, electrocardiograms and glucose monitoring. In such systems, wearable and flexible electronics technologies may enable monitoring of vital signs, offering significant potential for early screening as well as long-term behaviour modelling.In this thesis, large area pressure sensors based on non-conventional materials are proposed and realised by screen printing technique for monitoring sitting postures. The developed pressure sensing system measures distributed pressure when an individual sits on a chair equipped with a pressure sensor array. This technology could provide grounding for the advancement of health-related monitoring systems for both able-bodied and disabled individuals and inform them of their sitting time and sitting posture, and this could be used to establish a sitting pattern. To accomplish this, pressure sensors have been designed using non-conventional flexible electronics. A blend of non-conductive and low-resistance ink is used as pressure-sensitive material to enable the realization of screen-printed sensors. To characterise the performance of the suggested pressure sensor, several tests, such as repeatability, drift and flexibility, are conducted. The sensor has also been exposed to different humidity and temperature conditions in a climate chamber to examine its functionalities.A graphical user interface was developed for real-time demonstration of data from distributed pressure points in the form of a pressure map to display the pressure values. Four sitting postures are identified: forward, backward, left, and right leaning. Furthermore, a stretchable pressure sensor is proposed that could follow slight stretching with regard to changes in the shape of the human skin. Machine learning algorithms have been employed to further enhance the sitting posture identification, and accuracy of 99.03% is attained. A standalone embedded system capable of illustrating real-time pressure data has been developed with the potential to be used in portable health monitoring systems. In summary, this work provides a promising framework for measuring pressure distribution and identifying irregular sitting postures that may help to reduce the potential risks of developing health-related issues associated with prolonged sitting time.

    • Dogs can spontaneously develop complex systemic autoimmune disorders, with similarities to human autoimmune disease. Autoantibodies directed at self-antigens are a key feature of these autoimmune diseases. Here we report the identification of interleukin enhancer-binding factors 2 and 3 (ILF2 and ILF3) as autoantigens in canine immune-mediated rheumatic disease. The ILF2 autoantibodies were discovered in a small, selected canine cohort through the use of human protein arrays; a method not previously described in dogs. Subsequently, ILF3 autoantibodies were also identified in the same cohort. The results were validated with an independent method in a larger cohort of dogs. ILF2 and ILF3 autoantibodies were found exclusively, and at a high frequency, in dogs that showed a speckled pattern of antinuclear antibodies on immunofluorescence. ILF2 and ILF3 autoantibodies were also found at low frequency in human patients with SLE and Sjogren's syndrome. These autoantibodies have the potential to be used as diagnostic biomarkers for canine, and possibly also human, autoimmune disease.

  • The meaning of health, well-being, and quality of life perceived by Roma people in West Sweden

    • Many Roma people in Sweden are on the margins of society and face problems of social exclusion, institutional discrimination, low education, unemployment, and poor health. The aim is to describe how a group of Roma people, in West Sweden, understand health, well-being, and quality of life within the Roma context, and how they cope with their life-situation. Data consisted of qualitative interviews. The data were analyzed qualitatively using a phenomenological hermeneutic approach. The respondents mainly understood the concept of Health as "being healthy" and "feeling good". Elements that were crucial part of the respondents' health perception were being employed, having an education, social support from family and friends, freedom and security, and the extent of involvement in society. The results indicate that the respondents perceive their health and life situation as good, despite of their marginalized situation and discrimination.

    • Systemic lupus erythematosus (SLE) is an autoimmune disorder with heterogeneous clinical presentation and complex etiology involving the interplay between genetic, epigenetic, environmental and hormonal factors. Many common SNPs identified by genome wide-association studies (GWAS) explain only a small part of the disease heritability suggesting the contribution from rare genetic variants, undetectable in GWAS, and complex epistatic interactions. Using targeted re-sequencing of coding and conserved regulatory regions within and around 215 candidate genes selected on the basis of their known role in autoimmunity and genes associated with canine immune-mediated diseases, we identified a rare regulatory variant rs200395694:G > T located in intron 4 of the MEF2D gene encoding the myocyte-specific enhancer factor 2D transcription factor and associated with SLE in Swedish cohorts (504 SLE patients and 839 healthy controls, p = 0.014, CI = 1.1-10). Fisher's exact test revealed an association between the genetic variant and a triad of disease manifestations including Raynaud, anti-U1-ribonucleoprotein (anti-RNP), and anti-Smith (anti-Sm) antibodies (p = 0.00037) among the patients. The DNA-binding activity of the allele was further studied by EMSA, reporter assays, and minigenes. The region has properties of an active cell-specific enhancer, differentially affected by the alleles of rs200395694:G > T. In addition, the risk allele exerts an inhibitory effect on the splicing of the alternative tissue-specific isoform, and thus may modify the target gene set regulated by this isoform. These findings emphasize the potential of dissecting traits of complex diseases and correlating them with rare risk alleles with strong biological effects.

    • Syncytiotrophoblast and invasive extravillous trophoblast arise from a common stem cell, namely villous cytotrophoblast, but have very different characteristics. The study of the differentiation process relies on the availability of suitable markers for these different cell types of developing placenta. In this work, we have produced monoclonal antibodies that are specific to human villous cytotrophoblast. Monoclonal antibody (MAb) MG2 was specific to villous cytotrophoblast across gestation,                     and recognizes hepatocyte growth factor activator inhibitor type 1. MAb MD10 stained villous cytotrophoblast across gestation and also some endothelial cells, particularly in the second or third trimester. MAb MD10 recognizes human integrin α6β4. As a test for specificity, the novel MAbs were also used for staining of frozen tissue from human colon carcinoma. The results show that the two antibodies can be used as tools to study human villous cytotrophoblasts and also human tumors. The MG2 antibody seems most specific and promising for the study of various aspects of human villous cytotrophoblast.
